Polyadenylation of the 3-prime ends of eukaryotic mRNAs is a key event that takes place in the nucleus during maturation of mRNA. The reaction includes endoribonucleolytic cleavage of the pre-RNA at the poly(A) site that leads to synthesis of the poly(A) tail at the 3-prime end of the upstream cleavage product. The poly(A) polymerase (PAP) is required The adenosine addition reaction depends on poly(A) polymerase (PAP) activity. The testis express PAP-beta (TPAP) in the cytoplasm of spermatogenic cells. The adenosine addition function of PAP-beta plays a critical role in male germ cell production. PAP-beta-deficient transgenic mice display impaired expression of haploid-specific genes that are necessary for spermatogenesis. The intronless gene encoding human PAP-beta maps to chromosome 7p22.3.

The protein encoded by this gene is a member of the platelet-derived growth factor family. The four members of this family are mitogenic factors for cells of mesenchymal origin and are characterized by a motif of eight cysteines. This gene product can exist either as a homodimer (PDGF-BB) or as a heterodimer with the platelet-derived growth factor alpha polypeptide (PDGF-AB), where the dimers are connected by disulfide bonds. Mutations in this gene are associated with meningioma. Reciprocal translocations between chromosomes 22 and 7, at sites where this gene and that for COL1A1 are located, are associated with a particular type of skin tumor called dermatofibrosarcoma protuberans resulting from unregulated expression of growth factor. Ficolin B is the designation in mouse and rat of a protein also known as L-ficolin, ficolin-2, collagen/fibrinogen domain-containing protein 2, serum lectin p35, EBP-37 or hucolin. Ficolin B is a 313 amino acid member of the ficolin lectin family. It is a secreted innate immunity pattern recognition molecule expressed in liver and plasma that forms a disulfide-linked homopolymer. This extensive N-terminal disulfide bridge formation can lead to a functional dodecamer polypeptide. Ficolin B binds to DNA ligands expressed by late apoptotic and necrotic cells to increase attachment and engulfment. Variation in ficolin B concentrations amongst individuals is associated with polymorphisms in the promoter and structural portion of the FCN2 gene. In patients with Behé´t's disease (BD), there exists a significant difference in allele frequency for FCN2 gene single nucleotide polymorphisms (SNPs) within the -557 and -64 promoter sites within HLA-B51 positive and HLA-B51 negative subgroups.

FOXRED1 is a 486 amino acid single-pass membrane protein. Utilizing FAD as a cofactor, FOXRED1 may act as a chaperone protein essential for the function of mitochondrial complex I. Mutations to FOXRED1 may result in mitochondrial complex I deficiency (MT-C1D), which results in a wide range of clinical maladies from lethal neonatal disease to adult onset neurodegenerative disorders. Common phenotypes of MT-C1D include cardiomyopathy, liver disease, Leigh syndrome, Leber hereditary optic neuropathy, and some forms of Parkinson disease. FOXRED1 exists as three alternatively spliced isoforms and is encoded by a gene mapping to human chromosome 11q24.2. With approximately 135 million base pairs and 1,400 genes, chromosome 11 makes up around 4% of human genomic DNA and is considered a gene and disease association dense chromosome.

Chromosome 1 is the largest human chromosome spanning about 260 million base pairs and making up 8% of the human genome. There are about 3,000 genes on chromosome 1, and considering the great number of genes there are also a large number of diseases associated with chromosome 1. Notably, the rare aging disease Hutchinson-Gilford progeria is associated with the LMNA gene which encodes lamin A. When defective, the LMNA gene product can build up in the nucleus and cause characteristic nuclear blebs. The mechanism of rapidly enhanced aging is unclear and is a topic of continuing exploration. The MUTYH gene is located on chromosome 1 and is partially responsible for familial adenomatous polyposis. Stickler syndrome, Parkinsons, Gaucher disease and Usher syndrome are also associated with chromosome 1. A breakpoint has been identified in 1q which disrupts the DISC1 gene and is linked to schizophrenia. Aberrations in chromosome 1 are found in a variety of cancers including head and neck cancer, malignant melanoma and multiple myeloma. The C1orf183 gene product has been provisionally designated C1orf183 pending further characterization. There are two isoforms of C1orf183 that are produced as a result of alternative splicing events.

The second largest human chromosome, 2 consists of 237 million bases encoding over 1,400 genes and making up approximately 8% of the human genome. A number of genetic diseases are linked to genes on chromosome 2. Harlequin icthyosis, a rare and morbid skin deformity, is associated with mutations in the ABCA12 gene. The lipid metabolic disorder sitosterolemia is associated with ABCG5 and ABCG8. An extremely rare recessive genetic disorder, Alstré°‰ syndrome is due to mutations in the ALMS1 gene. Interestingly, chromosome 2 contains what appears to be a vestigial second centromere and vestigial telomeres which gives credence to the hypothesis that human chromosome 2 is the result of an ancient fusion of two ancestral chromosomes seen in modern form today in apes.

BXDC2 is a 306 amino acid protein encoded by the human gene BXDC2. BXDC2 is a nuclear protein that contains one Brix domain. Brix domain containing proteins represent a family of proteins involved in the biogenesis of large ribosomal subunits. The Brix domain is a region that is homologous to the yeast protein Pitx1 (Ribosome biogenesis protein BRX1). Pitx1 is part of a complex that includes BXDC5, BXDC1 and PPAN. This complex is required for the biogenesis of the 60S ribosomal subunit. BXDC2 exhibits the same functions as Pitx1.
