The NFKB complex consists of NFKB1 or NFKB2 bound to REL, RELA, or RELB. The NFKB complex is inhibited by I kappa B proteins (NFKBIA, or NFKBIB), which inactivate NF kappa B by trapping it in the cytoplasm. Phosphorylation of serine residues on the I kappa B proteins by kinases (IKBKA, or IKBKB,) marks them for destruction via the ubiquitination pathway, thereby allowing activation of the NF kappa B complex.

The amphoterin-induced gene and ORF (AMIGO) family of proteins consists of AMIGO1, AMIGO2 and AMIGO3. All three members are single pass type I membrane proteins that contain several leucine-rich repeats, one IgG domain, and a transmembrane domain. The AMIGO proteins are specifically expressed on fiber tracts of neuronal tissues and participate in their formation. The AMIGO proteins can form complexes with each other, but can also bind itself. AMIGO1, also designated Alivin-2, promotes growth and fasciculation of neurites and plays a role in myelination and fasciculation of developing neural axons. In cerebellar neurons, AMIGO2 (Alivin-1) is crucial for depolarization-dependent survival. Similar to AMIGO1 and AMIGO2, AMIGO3 (Alivin-3) plays a role in homophilic and/or heterophilic cell-cell interaction and signal transduction.

The 26S proteasome is a multicatalytic proteinase complex with a highly ordered structure composed of 2 complexes, a 20S core and a 19S regulator. In addition to normal tissue expression, p44S10 is also expressed in several melanoma cell lines, such as MCF-7, 451Lu and WM164. Since forced expression of p44S10 in radial growth phase melanoma cells results in an increase in cellular proliferation, p44S10 may represent a potential link between regulation of proteasome activity and tumor cell proliferation in vivo.
