Serine/threonine-protein kinase that plays a critical role in initiating innate immune response against foreign pathogens. Involved in Toll-like receptor (TLR) and IL-1R signaling pathways. Is rapidly recruited by MYD88 to the receptor-signaling complex upon TLR activation. In turn, MAP3K7/TAK1 activates IKKs (CHUK/IKKA and IKBKB/IKKB) leading to NF-kappa-B nuclear translocation and activation. Alternatively, phosphorylates TIRAP to promote its ubiquitination and subsequent degradation. Phosphorylates the interferon regulatory factor 7 (IRF7) to induce its activation and translocation to the nucleus, resulting in transcriptional activation of type I IFN genes, which drive the cell in an antiviral state. When sumoylated, translocates to the nucleus and phosphorylates STAT3 (By similarity).

RNF32 (RING finger protein 32), also known as HSD15 or FKSG33, is a 362 amino acid cytoplasmic protein that contains one IQ domain and two RING-type zinc fingers. Highly expressed in testis with lower expression levels in ovary tissue, RNF32 is thought to play a role in spermatogenesis, specifically contributing to the growth and maturation of round spermatids. Mutations of the gene coding for PAK3 are associated with X linked mental retardation and PAK3 is a key regulator of synapse formation and plasticity in the hippocampus. PAK3 is thought to play a key role in regulation of cell shape and motility as well as cell death. P21-activated kinase (PAK) is actually a family of serine/threonine protein kinases, members of which are activated by small molecular weight GTPases. The three most common isoforms are PAK 1, PAK 2, and PAK 3 (also known as alpha PAK, gamma PAK, and beta PAK, respectively). These kinases contain numerous regulatory elements that trigger diverse signaling processes such as those initiated by activated GTPases, interaction with Src homology 3 (SH3) domains, and caspase mediated proteolytic cleavage. NuMA (Nuclear Mitotic Apparatus Protein) is an intranuclear protein and present in nucleus during interphase. At the onset of mitosis, it redistributes from the nucleus to two centrosomal structures that later will become part of the mitotic spindle pole. After anaphase, the protein redistributes from the spindle polar region into reforming nucleus. NuMA is an essential protein during mitosis for the terminal phases of chromosome separation and/or nuclear reassembly. Recently a study shows that NuMA is cleaved to a 180 to 200kDa during apoptosis.

UNCX (UNC homeobox), also known as UNCX4.1, is a 531 amino acid nuclear transcription factor involved in neurogenesis and somitogenesis. UNCX assists in the formation of connections between hypothalamic neurons and the pituitary, which is necessary for central neurons to deliver hormones into peripheral blood. UNCX also plays a role in maintaining differentiation of the axial skeleton and acts upstream of Pax-9. The gene encoding UNCX maps to human chromosome 7, which houses over 1,000 genes and comprises nearly 5% of the human genome.
