They are abundant in the granules of neutrophils and also found in the epithelia of mucosal surfaces such as those of the intestine, respiratory tract, urinary tract, and vagina. Members of the defensin family are highly similar in protein sequence and distinguished by a conserved cysteine motif. Several alpha defensin genes are clustered on chromosome 8. This gene differs from other genes of this family by an extra 83-base segment that is apparently the result of a recent duplication within the coding region. The protein encoded by this gene, defensin, alpha 4, is found in the neutrophils; it exhibits corticostatic activity and inhibits corticotropin stimulated corticosterone production. E2F's are DNA binding proteins, which associate with negative regulators, such as the retinoblastoma p107 protein, resulting in an altered rate of gene transcription. The E2F proteins contain several evolutionally conserved domains found in most members of the family. These domains include a DNA binding domain, a dimerization domain which determines interaction with the differentiation regulated transcription factor proteins (DP), a transactivation domain enriched in acidic amino acids, and a tumor suppressor protein association domain which is embedded within the transactivation domain. This protein and another 2 members, E2F2 and E2F3, have an additional cyclin binding domain. E2F1 is proposed to be involved in several cellular processes that range from tumor suppressor, cell progression and oncogenesis. E2F1 overexpression can also drive cells into apoptosis.

Chromosome 1 is the largest human chromosome spanning about 260 million base pairs and making up 8% of the human genome. There are about 3,000 genes on chromosome 1, and considering the great number of genes there are also a large number of diseases associated with chromosome 1. Notably, the rare aging disease Hutchinson-Gilford progeria is associated with the LMNA gene which encodes lamin A. When defective, the LMNA gene product can build up in the nucleus and cause characteristic nuclear blebs. The mechanism of rapidly enhanced aging is unclear and is a topic of continuing exploration. The MUTYH gene is located on chromosome 1 and is partially responsible for familial adenomatous polyposis. Stickler syndrome, Parkinsons, Gaucher disease and Usher syndrome are also associated with chromosome 1. A breakpoint has been identified in 1q which disrupts the DISC1 gene and is linked to schizophrenia. Aberrations in chromosome 1 are found in a variety of cancers including head and neck cancer, malignant melanoma and multiple myeloma. The C1orf182 gene product has been provisionally designated C1orf182 pending further characterization.

IRGM belongs to a family of interferon-gamma inducible GTPases, belonging to the GTPase superfamily, and are selectively induced by IFN-gamma or bacterial lipopolysaccharide (LPS) stimulation. IRGM is primarily expressed in all cells derived from B-cell lineages, and is highly expressed in macrophages following IFN-gamma stimulation. IRGM has been shown to be required for host defences against a broad range of intracellular pathogens. Specifically, IRGM deficient mice show defects with intracellular microbial killing, phagosome maturation and autophagy. More recently IRGM has been shown to inhibit baseline hematopoietic proliferation and is required for a normal hematopoietic stem cell response to chemical and infectious stimuli.

ZNF318 is a 2279 amino acid endocrine regulatory protein that localizes to the nucleus. Highly expressed in testis, ovaries and kidneys, ZNF318 is a co-repressor of androgen receptor (AR)-mediated transcriptional activation and is thought to regulate transcription during spermatogenesis. ZNF318 interacts with the N-terminal domain of AR and contains two matrin-type zinc fingers. Two isoforms of ZNF318, designated TZF and TZF-L, are produced due to alternative splicing events. Each of these splice variants are thought to have unique roles in transcriptional regulation. While the TZF isoform functions as a repressor of AR-mediated transcriptional activation, the TZF-L isoform is thought to enhance AR-mediated transcriptional activation.
