Sox genes comprise a family of genes that are related to the mammalian sex determining gene SRY. These genes similarly contain sequences that encode for the HMG-box domain, which is responsible for the sequence-specific DNA-binding activity. Sox genes encode putative transcriptional regulators implicated in the decision of cell fates during development and the control of diverse developmental processes. The highly complex group of Sox genes cluster at least 40 different loci that rapidly diverged in various animal lineages. At present, 30 Sox genes have been identified. Members of this family have been shown to be conserved during evolution and to play key roles during animal development. Some are involved in human diseases, including sex reversal.

RDH13, also known as all-trans and 9-cis retinol dehydrogenase 13 or SDR7C3, is a 331 amino acid mitochondrial protein belonging to the short-chain dehydrogenases/reductases (SDR) family. Widely expressed, mostly in eye, pancreas, placenta and lung, RDH13 localizes on the outer side of the inner mitochondrial membrane. Related to microsomal retinoid oxidoreductase RDH11, RDH13 is considered to be a major enzyme among the RDH family of proteins. Catalytically active, RDH13 recognizes retinoids as substrates and may function in retinoic acid production. RDH13 may function to protect the mitochondria against oxidative stress. Leber congenital amaurosis (LCA) type 3, an inherited autosomal recessive retinal disease, has been associated with defects of RDH13. LCA represents the most common genetic cause of congenital visual impairment in infants and children.

Phelan-McDermid syndrome, Neurofibromatosis type 2 and autism are associated with chromosome 22. A schizophrenia susceptibility locus has been identified on chromosome 22 and studies show that 22q11 deletion symptoms include a high incidence of schizophrenia. Translocations between chromosomes 9 and 22 may lead to the formation of the Philadelphia Chromosome and the subsequent production of the novel fusion protein, BCR-Abl, a potent cell proliferation activator found in several types of leukaemia. The C22orf25 gene product has been provisionally designated C22orf25 pending further characterisation.

ACSVL family members are capable of activating both long (LCFAs) and very long-chain fatty acids (VLCFAs). There are six members of the human ACSVL subfamily, which have been described as solute carrier family 27A (SLC27A) gene products. They represent a group of evolutionarily conserved fatty acid transport proteins (FATPs) recognized for their role in facilitating translocation of long-chain fatty acids across the plasma membrane. The family nomenclature has recently been unified with their respective acyl-CoA synthetase family designations: ACSVL1 (FATP2), ACSVL2 (FATP6), ACSVL3 (FATP3), ACSVL4 (FATP1), ACSVL5 (FATP4) and ACSVL6 (FATP5). ACSVLs have unique expression patterns and are found in major organs of fatty acid metabolism, such as adipose tissue, liver, heart and kidney. ACSVL2 is a 619 amino acid multi-pass membrane protein. Glycogen phosphorylase (also known as GP) is an allosteric enzyme important in carbohydrate metabolism. Its activity is regulated through either noncovalent binding of metabolites or by covalent modification. Glycogen phosphorylase catalyzes the phosphorylation of glycogen to Glc-1-P. There are three genes which encode the brain, liver and muscle forms of glycogen phosphorylase, PYGB, PYGL and PYGM. Because of its fundamental role in the metabolism of glycogen, glycogen phosphorylase has been a target for the design of inhibitory compounds, which could be valuable in the therapeutic treatment of type 2 diabetes mellitus.
