Glycogen phosphorylase (also known as GP) is an allosteric enzyme important in carbohydrate metabolism. Its activity is regulated through either noncovalent binding of metabolites or by covalent modification. Glycogen phosphorylase catalyzes the phosphorylation of glycogen to Glc-1-P. There are three genes which encode the brain, liver and muscle forms of glycogen phosphorylase, PYGB, PYGL and PYGM. Because of its fundamental role in the metabolism of glycogen, glycogen phosphorylase has been a target for the design of inhibitory compounds, which could be valuable in the therapeutic treatment of type 2 diabetes mellitus.

Description:   The human insulin receptor is a heterotetrameric membrane glycoprotein consisting of disulfide linked subunits in a beta-alpha-alpha-beta configuration. Receptor activation occurs through ligand binding, which facilitates receptor dimerization and autophosphorylation of specific tyrosine residues in the cytoplasmic portion. The interaction of insulin with the alpha subunit of the insulin receptor activates the protein tyrosine kinase of the beta subunit, which then undergoes an autophosphorylation that increases its tyrosine kinase activity. Three adapter proteins, IRS1, IRS2 and Shc, become phosphorylated on tyrosine residues following insulin receptor activation. These three phosphorylated proteins then interact with SH2 domain containing signaling proteins.

Description:   KIF13B is also known as Kinesin-like protein GAKIN or GAKIN and is a 1,826 amino acid protein that is widely expressed in tissues, with highest expression in brain and kidney. KIF13B is localized to the cytoplasm, as well as to the cytoskeleton, and is thought to be a microtubule-dependent motor protein which is able to bind to a variety of proteins in order to traffic them to various locations throughout the cell. KIF13B belongs to the kinesin-like protein family and possesses three domains typical of the kinesin-like protein family, namely an N-terminal motor domain with an ATP-binding motif, an FHA domain which is known to bind diverse cargos and a large stalk domain involved in protein-protein binding. Additionally, KIF13B has a microtubule-interacting sequence which is known as the CAP-Gly domain at its C-terminus. Description:   Made up of nearly 146 million bases, chromosome 8 encodes about 800 genes. Translocation of portions of chromosome 8 with amplifications of the c-Myc gene are found in some leukemias and lymphomas, and typically associated with a poor prognosis. Portions of chromosome 8 have been linked to schizophrenia and bipolar disorder. Trisomy 8, also known as Warkany syndrome 2, most often results in early miscarriage but is occasionally seen in a mosaic form in surviving patients who suffer to a varying degree from a number of symptoms including retarded mental and motor development, and certain facial and developmental defects. WRN is a DNA helicase encoded by chromosome 8 and shown defective in those with the early aging disorder Werner syndrome. Chromosome 8 is also associated with Pfeiffer syndrome, congenital hypothyroidism and Waardenburg syndrome. The C8orf31 gene product has been provisionally designated C8orf31 pending further characterisation.

Description:   Chromosome 1 is the largest human chromosome spanning about 260 million base pairs and making up 8% of the human genome. The MUTYH gene is located on chromosome 1 and is partially responsible for familial adenomatous polyposis. Stickler syndrome, Parkinsons, Gaucher disease and Usher syndrome are also associated with chromosome 1. A breakpoint has been identified in 1q which disrupts the DISC1 gene and is linked to schizophrenia. Aberrations in chromosome 1 are found in a variety of cancers including head and neck cancer, malignant melanoma and multiple myeloma.
