The protein encoded by this gene is a member of the protein tyrosine phosphatase (PTP) family. PTPs are known to be signaling molecules that regulate a variety of cellular processes including cell growth, differentiation, mitotic cycle, and oncogenic transformation. This gene is preferentially expressed in a variety of hematopoietic cells, and is an early response gene in lymphokine stimulated cells. The non-catalytic N-terminus of this PTP can interact with MAP kinases and suppress the MAP kinase activities. This PTP was shown to be involved in the regulation of T cell antigen receptor (TCR) signaling, which was thought to function through dephosphorylating the molecules related to MAP kinase pathway. Retinoids are metabolites of vitamin A (retinal) and are believed to represent important signaling molecules during vertebrate development and tissue differentiation. Two families of retinoid receptors have been identified. Retinoic acid receptors (RARs) include RAR alpha, RAR beta and RAR gamma, each of which has a high affinity for all trans retinoic acids and belongs to the same class of nuclear transcription factors as thyroid hormone receptors, vitamin D3 receptor and ecdysone receptor. The ligand binding domains of the RARs are highly conserved and RAR isoforms are expressed in distinct patterns through out development and in the mature organism. Members of the retinoid X receptor (RXR) family, RXR alpha, RXR beta and RXR gamma, are activated by 9 cis retinoic acid, a stereo and photoisomer of all trans RA that is expressed in vivo in both liver and kidney and may represent a widely used hormone.

AHNAK is a 5,890 amino acid protein encoded by the human gene AHNAK. The intronless AHNAK gene is located on human chromosome 11q12 and has three main structural regions: the 251 amino acid N-terminus, a large central region of 4390 amino acids with multiple repeated units of about 128 amino acids in length, and the 1002 amino acid C-terminus. The central region seems to have antiparallel beta-strands connected by intervening loops. Several putative regulatory elements are clustered within the C-terminal region, including nuclear export localization signals, a leucine zipper, and potential phosphorylation sites for Akt1 and PKC. AHNAK is believed to be an important signalling molecule involved in a wide range of physiological activities and may be required for neuronal cell differentiation.. AHNAK also appears to influence b-adrenergic regulation of cardiac L-type Ca2+ channel function.

The HUNK (hormonally upregulated Neu-associated kinase) protein, also designated MAK-V in mouse, has been identified as a novel SNF1-related serine/threonine kinase. The human HUNK gene localizes to chromosome 21q22 and encodes a protein with nucleocytoplasmic distribution and localizes to the centrosome. Overexpression of the HUNK protein associates with approximately 50% of breast carcinomas, and may provide diagnostic-prognostic value as a molecular marker. Serine/threonine-protein kinase SNF1-like kinase 2 (SIK) phosphorylates Ser-794 of IRS1 in insulin-stimulated adipocytes, which may modulate the efficiency of insulin signal transduction. SIK is activated by phosphorylation on Thr-175 by STK11 in complex with STE20-related adapter-α and CAB39.

A number of genetic diseases are linked to genes on chromosome 2. Harlequin icthyosis, a rare and morbid skin deformity, is associated with mutations in the ABCA12 gene. The lipid metabolic disorder sitosterolemia is associated with ABCG5 and ABCG8. An extremely rare recessive genetic disorder, Alstré°‰ syndrome is due to mutations in the ALMS1 gene. Interestingly, chromosome 2 contains what appears to be a vestigial second centromere and vestigial telomeres which gives credence to the hypothesis that human chromosome 2 is the result of an ancient fusion of two ancestral chromosomes seen in modern form today in apes.

Bad is a member of the Bcl2 family and acts to promote apoptosis by forming heterodimers with the survival proteins Bcl2 and BclxL, thus preventing them from binding with BAX. Bad is found on the outer mitochondrial membrane and, once phosphorylated in response to growth stimuli, translocates to the cytoplasm. The phosphorylation status of Bad represents a key checkpoint for death or cell survival. JNK-induced phosphorylation of BAD serine 128 promotes the apoptotic role of Bad by opposing the inhibitory effect of growth factor on Bad-mediated apoptosis. Cdc2-induced phosphorylation of Bad serine 128 has an inhibitory effect on its interaction with 14-3-3 proteins. The latter interaction is critical for Bad phosphorylation at serine 155, a site within the BH3 domain that leads to the release of BclxL and the promotion of cell survival. Alternative splicing of this gene results in two transcript variants which encode the same isoform.

Zinc finger proteins contain DNA-binding domains and have a wide variety of functions, most of which encompass some form of transcriptional activation or repression. The majority of zinc finger proteins contain a Krüppel-type DNA binding domain and a KRAB domain, which is thought to interact with KAP1, thereby recruiting histone modifying proteins. Zinc finger protein 750 is a 723 amino acid member of the Krüppel C2H2-type zinc finger protein family. Localized to the nucleus, ZNF750 contains one conserved C2H2 zinc finger domain and is expressed in the skin, lungs, prostate, placenta and thymus. ZNF750 is also expressed in primary human keratinocytes but not in fibroblasts. Mutations in the gene encoding ZNF750 cause Seborrhea-like dermatitis with psoriasiform, a condition characterized by a chronic and diffuse rash on the face and hyperkeratosis of skin over the elbows, soles, knees and palms.

HECA, also known as headcase homolog, HDC, HDCL or HHDC, is a 543 amino acid mammalian homolog of the Drosophila headcase protein, a highly basic, cytplasmic peptide that plays a role in mitotic re-entry during adult morphogenesis. Expressed in a variety of tissues with highest expression in thymus, spleen and heart, HECA is thought to play a role in the development of epithelial tube networks in lung tissue and may also be involved in the pathogenesis of lung cancer. The gene encoding HECA maps to human chromosome 6, which contains 170 million base pairs and comprises nearly 6% of the human genome. Deletion of a portion of the q arm of chromosome 6 is associated with early onset intestinal cancer, suggesting the presence of a cancer susceptibility locus. Additionally, Porphyria cutanea tarda, Parkinson's disease, Stickler syndrome and a susceptibility to bipolar disorder are all associated with genes that map to chromosome 6.
