Description:   Involved in the base excision repair (BER) pathway, by catalyzing the poly(ADP-ribosyl)ation of a limited number of acceptor proteins involved in chromatin architecture and in DNA metabolism. This modification follows DNA damages and appears as an obligatory step in a detection/signaling pathway leading to the reparation of DNA strand breaks. May link the DNA damage surveillance network to the mitotic fidelity checkpoint. Negatively influences the G1/S cell cycle progression without interfering with centrosome duplication. Binds DNA. May be involved in the regulation of PRC2 and PRC3 complex-dependent gene silencing.Tissue specificity: Widely expressed; the highest levels are in the kidney, skeletal muscle, liver, heart and spleen; also detected in pancreas, lung, placenta, brain, leukocytes, colon, small intestine, ovary, testis, prostate and thymus.

Description:   The mini-chromosome maintenance (MCM) family of proteins, including MCM2, MCM3, MCM4 (Cdc21), MCM5 (Cdc46), MCM6 (Mis5) and MCM7 (Cdc47), are regulators of DNA replication that act to ensure replication occurs only once in the cell cycle. Expression of MCM proteins increases during cell growth, peaking at G1 to S phase. The MCM proteins each contain an ATP-binding motif, which is predicted to mediate ATP-dependent opening of double-stranded DNA. MCM proteins are regulated by E2F transcription factors, which induce MCM expression, and by protein kinases, which interact with MCM proteins to maintain the postreplicative state of the cell. MCM2/MCM4 complexes function as substrates for Cdc2/cyclin B in vitro. Cleavage of MCM3, which can be prevented by caspase inhibitors, results in the inactivation of the MCM complex (composed of at least MCM proteins 2-6) during apoptosis.

Description:   HERPUD1 is a includes the inhibition of translation to prevent further accumulation of unfolded proteins, the increased expression of proteins involved in polypeptide folding, known as the unfolded protein response (UPR), and the destruction of misfolded proteins by the ER-associated protein degradation (ERAD) system. This gene may play a role in both UPR and ERAD. Its expression is induced by UPR and it has an ER stress response element in its promoter region while the encoded protein has an N-terminal ubiquitin-like domain which may interact with the ERAD system. This protein has been shown to interact with presenilin proteins and to increase the level of amyloid-beta protein following its overexpression. Alternative splicing of this gene produces multiple transcript variants encoding different isoforms. The full-length nature of all transcript variants has not been determined.

Description:   This endogenous retroviral envelope protein has retained its original fusogenic properties and participates in trophoblast fusion and the formation of a syncytium during placenta morphogenesis. The interaction with MFSD2A is apparently important for this process. Endogenous envelope proteins may have kept, lost or modified their original function during evolution but this one can still make pseudotypes with MLV, HIV-1 or SIV-1 virions and confer infectivity. Retroviral envelope proteins mediate receptor recognition and membrane fusion during early infection. The surface protein mediates receptor recognition, while the transmembrane protein anchors the envelope heterodimer to the viral membrane through one transmembrane domain. The other hydrophobic domain, called fusion peptide, mediates fusion of the viral membrane with the target cell membrane.

Description:   The RING-type zinc finger motif is present in a number of viral and eukaryotic proteins and is made of a conserved cysteine-rich domain that is able to bind two zinc atoms. Proteins that contain this conserved domain are generally involved in the ubiquitination pathway of protein degradation. RNF17 (ring finger protein 17) or tudor domain-containing protein 4, TDRD4, SPATA23, Mmip-2 or FLJ11045, is a testis-specific protein and and novel key regulator of spermiogenesis containing 1,623 amino acids. By distributing Mad proteins to the cytoplasm, RNF17 regulates the transcriptional activity of c-Myc. Although showing localization in the nucleus, RNF17 is predominantly observed in cytoplasm and is a component of a novel nuage found in male germ cells. The gene encoding RNF17 maps to human chromosome 13q12.12 and encodes one RING-type zinc finger and four tudor domains. As a result of alternative splice events, five RNF17 isoforms exisit.

Description:   The protein encoded by this gene contains a RING-H2 finger motif. Deletions and mutations in this gene were detected in esophageal squamous cell carcinoma (ESCC), suggesting that this protein may be a potential tumor suppressor. Studies of the mouse counterpart suggested a role of this protein in the transcription regulation that controls germinal differentiation. Multiple alternatively spliced transcript variants encoding the same protein are observed. [provided by RefSeq, Jul 2008].

Description:   FGF20 is secreted heparin binding growth factor that is a member of the FGF family. FGF family members possess broad mitogenic and cell survival activities, and are involved in a variety of biological processes including embryonic development cell growth, morphogenesis, tissue repair, tumor growth and invasion by promoting cellular proliferation and differentiation. The gene for FGF20 was shown to be expressed in normal brain, particularly the cerebellum. FGF20 signals through the FGFR 2c and 3c and is expressed during limb and brain development.

Description:   The cysteine-rich, adipose tissue-specific, secretory factor resistin (resistance to insulin, also known as ADSF) is a secreted hormone that potentially links obesity to diabetes. Resistin is rich in serine and cysteine residues and contains a unique cysteine repeat motif. Resistin and the resistin-like molecules share the characteristic cysteine composition and other signature features. Resistin-like a is a secreted protein that has restricted tissue distribution and is most highly expressed in adipose tissue. Another family member, Resistin-like b, is a secreted protein expressed only in the gastrointestinal tract, particularly in the colon, in both mouse and human. Resistin-like b expression is highest in proliferative epithelial cells and is markedly increased in tumors, suggesting a role in intestinal proliferation.

Description:   Subunit of mTORC2, which regulates cell growth and survival in response to hormonal signals. mTORC2 is activated by growth factors, but, in contrast to mTORC1, seems to be nutrient-insensitive. mTORC2 seems to function upstream of Rho GTPases to regulate the actin cytoskeleton, probably by activating one or more Rho-type guanine nucleotide exchange factors. mTORC2 promotes the serum-induced formation of stress-fibers or F-actin. mTORC2 plays a critical role in AKT1 'Ser-473' phosphorylation, which may facilitate the phosphorylation of the activation loop of AKT1 on 'Thr-308' by PDK1 which is a prerequisite for full activation. mTORC2 regulates the phosphorylation of SGK1 at 'Ser-422'. mTORC2 also modulates the phosphorylation of PRKCA on 'Ser-657'. Plays an essential role in embryonic growth and development.

Description:   PRE7 is a subunit of the Yeast proteasome. The yeast proteasome seems to be composed of 14 different subunits which form a highly ordered ring-shaped structure. The proteasome degrades poly-ubiquitinated proteins in the cytoplasm and in the nucleus. It is essential for the regulated turnover of proteins and for the removal of misfolded proteins.
