Hemogen is a 484 amino acid protein encoded by the human gene HEMGN. Hemogen is a nuclear protein that is expressed in hematopoietic precursor cells and can be detected in CD34+ and K-562 leukemia cell line. It is also expressed in bone marrow, testis, thymus and thyroid tumors, non-Hodgkin lymphoma, various leukemia cell lines, peripheral blood mononuclear cells (PBMCs) and bone marrow mononuclear cells (BMMCs) of patients with leukemia. Hemogen is down-regulated during megakaryocytic differentiation of K-562 cells by 12-O-tetradecanoylphorbol-13-acetate (TPA) (at protein level). It can be up-regulated in normal PBMCs by mitogens.

Description:
Sorting nexin 1 (SNX1) is a member of a large family of hydrophilic proteins that interact with a variety of receptor types and are involved in intracellular trafficking. SNX1 and the related splice variant, SNX1A, bind the epidermal growth factor (EGF) receptor and facilitate its transport to lysosome, thereby contributing to the degradation of the receptor. SNX2 and SNX4 share a high degree of amino acid similarity with SNX1, as they all contain a characteristic phox homology (PX) domain. These proteins are all partially associated with cellular membranes and they likewise associate with EGF, PDGF and insulin receptor tyrosine kinases. These nexins are widely expressed and yet have various tissue distribution patterns. Additionally, the sorting nexins can associate with each other and with a variety of other cellular proteins, suggesting that they exist as part of multi-subunit complexes. The related protein, SNX3, comprises a distinct subgroup of nexins that share less sequence similarity outside of the PX domain and have dramatically different binding affinities for the tyrosine kinase receptors.

It belongs to the GATA family of transcription factors. GATA1 is a transcriptional activator which probably serves as a general switch factor for erythroid development. It binds to DNA sites with the consensus sequence [AT]GATA[AG] within regulatory regions of globin genes and of other genes expressed in erythroid cells. The protein also plays an important role in erythroid development by regulating the switch from fetal hemoglobin production to adult hemoglobin.

Glutathione (GSH) is a tripeptide antioxidant which reduces disulfide bonds between cytoplasmic proteins. The constitutive enzyme glutathione reductase transforms glutathione into its reduced state which ultimately can provide a measure of cellular toxicity. GSTT2 (glutathione S-transferase theta-2), also known as GST class-theta-2, is a 244 amino acid enzyme with sulfatase activity that functions in conjugating reduced glutathione to hydrophobic electrophiles. GSTT2 exists as a homodimer in the cytoplasm and is expressed in low levels in the liver and the lung. GSTT2 belongs to the GST superfamily and contains both a GST C-terminal and a GST N-terminal domain. The gene encoding GSTT2 exists on human chromosome 22.
