IQSEC2 is a 1,478 amino acid protein that belongs to the BRAG family and contains one IQ domain, one PH domain and a SEC7 domain. Localizing to the cytoplasm, IQSEC2 is expressed in brain, kidney and small intestine, with weaker levels of expression in placenta, pancreas, ovary, prostate and liver. IQSEC2 is a component of the postsynaptic density at excitatory synapses, and interacts with ARF family members as a guanine nucleotide exchange factor. Through the activation of ARF substrates, IQSEC2 may play a crucial role in cytoskeletal and synaptic organization. The gene encoding IQSEC2 maps to the human X chromosome. Defects to the IQSEC2 gene have been linked to mental retardation X-linked type 1 (MRX1), a condition characterized by decreased intellectual function. IQSEC2 exists as three isoforms due to alternative splicing events.

BMP2 belongs to the transforming growth factor-beta (TGFB) superfamily of secreted growth factors. It is a disulfide-linked homodimer and induces bone and cartilage formation. In addition to its osteogenic activity, BMP2 plays an important role in cardiac morphogenesis and is expressed in a variety of tissues including lung, spleen, brain, liver, prostate ovary and small intestine. The functional form of BMP2 is a 26 kDa protein composed of two identical 114 amino acid polypeptide chains linked by a single disulfide bond. BMPs control fundamental events in early embryonic development, organogenesis and adult tissue homeostasis.

Mahogany (MG), originally identified as a protein involved in pigmentation, acts in conjunction with melanocortin receptors to suppress diet-induced obesity. Mahogany contains a single transmembrane domain, and it is expressed in a broad range of tissues, including the hypothalamus and pigment cells. Mutations within the mahogany gene were shown to rescue agouti-lethal-yellow mutant mice from obesity. The extracellular domain of mouse mahogany is the ortholog of the human protein attractin. Attractin (also designated DPPT-L) is a human serum glycoprotein and is a member of the CUB family of cell adhesion and guidance proteins. Attractin is expressed on activated T cells and is released from the cells 48 to 72 hours after activation.

The gene that encodes Notum consists of approximately 9334 bases and maps to human chromosome 17q25.3. Encoding more than 1200 genes, chromosome 17 comprises over 2.5% of the human genome. Two key tumour suppressor genes are associated with chromosome 17, namely, p53 and BRCA1. tumour suppressor p53 is necessary for maintenance of cellular genetic integrity by moderating cell fate through DNA repair versus cell death. Malfunction or loss of p53 expression is associated with malignant cell growth and Li-Fraumeni syndrome. Like p53, BRCA1 is directly involved in DNA repair, though specifically it is recognised as a genetic determinant of early onset breast cancer and predisposition to cancers of ovary, colon, prostate gland and fallopian tubes.

Arginase I which is expressed almost exclusively in the liver, catalyzes the conversion of arginine to ornithine and urea . The human arginase I gene, which maps to chromosome 6q23, encodes a 322 amino acid protein. Arginase I exists as a homotrimeric protein and contains a binuclear manganese cluster. Arginase II catalyzes the same reaction as arginase I, but differs in its tissue specificity and subcellular location. Specifically, arginase II localizes to the mitochondria. Arginase II is expressed in non-hepatic tissues, with the highest levels of expression in the kidneys, but, unlike arginase I, is not expressed in liver. ARFs influence vesicle trafficking and signal transduction in eukaryotic cells (1–3). ARF-dependent regulatory mechanisms include the coordination of spectrin interactions with golgi membranes and the association of actin to the golgi via rho family-dependent G-protein localization (Rac, CDC42) and WASP/Arp2/3 complexes (3–7). Additionally, ARFs play a central role in maintenance of organelle integrity, assembly of coat proteins, and activation of phospholipase D (5–7). The ARF proteins are categorized as class I (ARF1, ARF2,and ARF3), class II (ARF4 and ARF5) and class III (ARF6); members of each class share a common gene organization (8,9). The human ARF6 gene maps to chromosome 7q22.1, contains five exons and four introns, and encodes a 175 amino acid protein (8,9).
