Description:   This gene encodes an accessory subunit of the mitochondrial membrane respiratory chain NADH dehydrogenase (Complex I), or NADH:ubiquinone oxidoreductase, the first multi-subunit enzyme complex of the mitochondrial respiratory chain. Complex I plays a vital role in cellular ATP production, the primary source of energy for many crucial processes in living cells. It removes electrons from NADH and passes them by a series of different protein-coupled redox centers to the electron acceptor ubiquinone. In well-coupled mitochondria, the electron flux leads to ATP generation via the building of a proton gradient across the inner membrane. Complex I is composed of at least 41 subunits, of which 7 are encoded by the mitochondrial genome and the remainder by nuclear genes. [provided by RefSeq, Jul 2008].

Description:   The survival and development of central neurons require the supply of trophic factors by glial cells. The trophic actions of glial cells on Purkinje neurons are mediated by L-serine and glycine, which are glia-derived trophic factors synthesized by 3PGDH (1). 3PGDH protein is 544 amino acids in length. Two distinct mRNA transcripts that encode for 3PGDH protein in normal human tissues are dominant 2.1 kb mRNA, which is highly expressed in prostate, testis, ovary, brain, liver, kidney, and pancreas, and weakly expressed in thymus, colon, and heart, and 710 bp mRNA, which is highly expressed in heart and skeletal muscle (2). 3PGDH is regulated at the transcriptional level depending on tissue specificity and cellular proliferative status (2). 3PGDH protein is also highly expressed in adult and fetal brain tissues (3). 3PGDH protein plays an important role in the metabolism, development, and function of the central nervous system (3) and its deficiency is a treatable congential error (4-5) that impairs L-serine biosynthesis which is characterized by congenital microcephaly, psychomotor retardation, and seizures (3).

Description:   Cyclin dependent kinases are positively regulated by association with cyclins and negatively regulated by binding to inhibitory subunits. The activity of cyclin dependent kinases is also regulated by the phosphorylation status, which is controlled by the antagonistic action of Wee1 kinase and CDC25 phosphatases. Three CDC25 genes are present in human cells: CDC25A, CDC25B, and CDC25C. These three genes function at different phases of the cell cycle. Whereas CDC25A and CDC25B are expressed throughout the cell cycle, with peak expression in G1 for CDC25A and in both G1 S phase and G2 for CDC25B, CDC25C is predominantly expressed in G2.

Description:   The steroid receptor superfamily acts through direct association with DNA sequences known as hormone response elements (HREs) and binds DNA as either homo- or heterodimers. The promiscuous mediator of heterodimerization, RXR, is the receptor for 9-cis retinoic acid, and dimerizes with VDR, TR, PPAR, and several novel receptors including LXR (also referred to as RLD-1) and FXR. FXR and LXR fall into a category of proteins termed orphan receptors? because of their lack of a defined function, and in the case of LXR, the lack of a defined ligand. FXR has been shown to bind a class of lipid molecules called farnesoids. LXR/RXR heterodimers have highest affinity for DR-4 DNA elements while FXR/RXR heterodimers bind IR-1 elements. Both LXR/RXR and FXR/RXR heterodimers retain their responsiveness to 9-cis retinoic acid.

Description:   LMBR1 is a 490 amino acid multi-pass membrane protein that is widely expressed with strongest expression in heart and pancreas. Belonging to the LIMR family, LMBR1 shares 95% sequence identity with the mouse protein and may play crucial role in the evolution of limb and skeletal system. LMBR1 is critical for expression of sonic hedgehog (Shh) in the developing posterior limb bud mesenchyme. Mutations in the gene encoding LMBR1 is the cause of several rare conditions such as acheiropody (ACHP) and syndactyly type 4 (SDYT4). ACHP is an autosomal recessive inherited disorder characterized by bilateral congenital amputations of the hands and feet. LMBR1L (limb region 1 protein homolog-like), also known as LIMR (Lipocalin-1-interacting membrane receptor), is a 489 amino acid multi-pass membrane protein that is thought to act as a receptor for Lipocalin-1 and may also assist in its endocytosis.

Description:   The human augmin complex (HAUS) is an evolutionarily conserved 8-subunit protein complex that was initially discovered in Drosophila. The HAUS complex is essential for microtubule generation, centrosome integrity, mitotic spindle assembly and completion of cytokinesis. HAUS6 (HAUS augmin-like complex, subunit 6), also known as FAM29A or Dgt6, is a 955 amino acid component of the augmin complex. Required for mitotic progression, HAUS6 localizes to cytoplasm, cytoskeleton, mitotic spindle microtubules and interphase centrosomes, and undergoes post-translational phosphorylation following mitosis on multiple serine and threonine residues. HAUS6 exists as two alternatively spliced isoforms that are encoded by a gene located on human chromosome 9.

Description:   Bid, a BH3 domain containing proapoptotic Bcl2 family member, is localized in the cytosolic fraction of cells as an inactive precursor. Its active form is generated upon proteolytic cleavage by caspase 8 in the Fas signaling pathway. Cleaved Bid translocates to mitochondria and releases its potent proapoptotic activity, which in turn induces cytochrome c release and mitochondrial damage. The cytochrome c releasing activity of Bid was antagonized by Bcl2. Mutation in the SH3 domain can diminish the cytochrome c releasing activity. In animal model studies, Bid deficient mice are found resistant to the lethal effects of death factor signals relayed through Fas.

Description:   Cystatin A and cystatin B (also designated PME, CST6, STFB, CPI-B, stefin B and liver thiol proteinase inhibitor) are thiol protease inhibitors that form complexes with papain and the cathepsins B, H, and L. Cystatin A, a cytoplasmic protein, is one of the precursor proteins of the cornified cell envelope in keratinocytes and plays a role in epidermal development and maintenance. Cystatin B protects against intracellular proteases leaking out of lysosomes and is primarily expressed in heart, liver and kidney.
