Description:   In mammalian cells, translation is controlled at the level of polypeptide chain initiation by initiation factors. Eukaryotic translation initiation factor 1 (eIF1) is crucial for the scanning process in vitro. During the scanning process, eIF1 is a component of a complex involved in recognition of the initiator codon. Translation is also initiated by the role of eIF1 in regulating the activity of ribosomal subunits 43S, 48S and 40S. eIF1 enables 43S ribosomal complexes to discern between cognate and near-cognate initiation codons, sensing the nucleotide content of initiation codons. It is also a promotor, along with eukar-yotic translation initiation factor 1A (eIF1A), for assembly of 48S ribosomal complexes at the initiation codon of a conventional capped mRNA. In addition, eIF1 and eIF1A, together with eukaryotic translation initiation factor 5 (eIF5), function in the formation of stable 40S ribosomal preinitiation complexes.

Description:   DNA polymerase lambda (pol lambda), also designated DNA polymerase lambda or Pol Beta2, is a low-fidelity polymerase which plays a role in both spontaneous and DNA damage-induced mutagenesis. Encoded by the POLL gene, pol lambda is a member of the DNA polymerase type-X family. Pol lambda extends primer-terminal mispairs opposite nondamaged DNA templates, suggesting that it may assist in extending mismatched base pairs during normal DNA replication. In addition, pol ?may play a role in the mutagenic bypass of T-T dimers. Proliferating cell nuclear antigen (PCNA), a protein essential to DNA replication, interacts with pol lambda and thus influences the ability of pol ?to synthesize DNA.

Description:   Comprising more than ten subunits, the anaphase-promoting complex (APC) acts in a cell-cycle dependent manner to promote the separation of sister chromatids during the transition between metaphase and anaphase in mitosis. APC, or cyclosome, accomplishes this progression through the ubiquitination of mitotic cyclins and other regulatory proteins that are targeted for destruction during cell division. APC is phosphorylated, and thus activated, by protein kinases Cdk1/cyclin B and polo-like kinase (Plk). APC is under tight control by a number of regulatory factors, including CDC20, CDH1 and MAD2. Specifically, CDC20 and CDH1 directly bind to APC and activates APC’s cyclin-ubiquitination activity. In contrast, MAD2 inhibits APC by forming a ternary complex with CDC20 and APC; thus preventing APC activation. APC11 is a RING-H2 finger protein that allows for the synthesis of multiubiquitin chains in the presence of Ubiquitin carrier protein 4 (Ubc4) and ubiquitin conjugating enzyme (E2). In addition, a heterodimeric complex of either Ubc4 or UbcH10 with APC11 and APC2 catalyzes the ubiquitination of human securin and cyclin B1.

Description:   Mono-ADP-ribosylation is one of the posttranslational protein modifications regulating cellular metabolism (e.g. nitrogen fixation) in prokaryotes. Mono-ADP-ribosylation is a process in which the ADP-ribose moiety of nicotinamide adenine dinucleotide is transferred to an acceptor amino acid. Five mammalian ADP-ribosyltransferases (ART1-ART5) have been cloned, and expression is restricted to tissues such as cardiac and skeletal muscle, leukocytes, brain and testis. ART3 (ADP-ribosyltransferase 3), also known as Ecto-ADP-ribosyltransferase 3, is a testis specific membrane protein that does not appear to have ADP-ribosyltransferase activity. It lacks the R-S-EXE active site motif and is therefore unable to catalyze the reaction. ART3 is predominantly found in spermatocytes and may play a role in spermatogenesis.

Acts as an alarmin or a danger associated molecular pattern (DAMP) molecule and stimulates innate immune cells via binding to pattern recognition receptors such as Toll-like receptor 4 (TLR4) and receptor for advanced glycation endproducts (AGER). Binding to TLR4 and AGER activates the MAP-kinase and NF-kappa-B signaling pathways resulting in the amplification of the proinflammatory cascade. Has antimicrobial activity towards bacteria and fungi and exerts its antimicrobial activity probably via chelation of Zn(2+) which is essential for microbial growth. Can induce cell death via autophagy and apoptosis and this occurs through the cross-talk of mitochondria and lysosomes via reactive oxygen species (ROS) and the process involves BNIP3. Description:   The LIM-only (LMO) proteins, LMO1 and LMO2, are nuclear factors that are characterized by a conserved LIM domain (1). The LIM domain consists of a cysteine-rich zinc-binding motif that is present in a variety of transcription factors, including the LIM homeobox (LHX) proteins expressed in the central nervous system and involved in cell differentiation (2).
