Description:   Glutamate receptors mediate most excitatory neurotransmission in the brain and play an important role in neural plasticity, neural development and neurodegeneration. Ionotropic glutamate receptors are categorized into NMDA receptors and kainate/AMPA receptors, both of which contain glutamate-gated, cation-specific ion channels. Gcom1 (GRINL1A complex locus protein 1), also known as GUP (GRINL1A upstream protein) and Gcom (GRINL1A combined protein), is a 466 amino acid protein that is a component of the GRINL1A complex transcription unit, which is thought to be involved in the modulation of glutamatergic neurotransmission through interaction with the NR1 subunit of the NMDA receptor. Gcom1 is expressed in small intestine, lung, liver, heart, skeletal muscle, testis and prostate and also colocalizes with NR1 in cortical and hippocampal neurons. There are eleven isoforms of Gcom1 that are produced as a result of alternative splicing events.

Description:   In mammalian cells, translation is controlled at the level of polypeptide chain initiation by initiation factors. Eukaryotic translation initiation factor 1 (eIF1) is crucial for the scanning process in vitro. During the scanning process, eIF1 is a component of a complex involved in recognition of the initiator codon. Translation is also initiated by the role of eIF1 in regulating the activity of ribosomal subunits 43S, 48S and 40S. eIF1 enables 43S ribosomal complexes to discern between cognate and near-cognate initiation codons, sensing the nucleotide content of initiation codons. It is also a promotor, along with eukar-yotic translation initiation factor 1A (eIF1A), for assembly of 48S ribosomal complexes at the initiation codon of a conventional capped mRNA. In addition, eIF1 and eIF1A, together with eukaryotic translation initiation factor 5 (eIF5), function in the formation of stable 40S ribosomal preinitiation complexes.

Description:   Cell adhesion molecule-related/down-regulated by oncogenes (CDO) and BOC (brother of CDO) are members of the immunoglobulin/fibronectin type III repeat family and act as cell surface receptors. CDO is a component of a cell-surface receptor complex which also contains BOC, NEO1, CTNNB1 and cadherins and which acts as a mediator of cell-cell interactions between muscle cells. CDO and BOC are single pass membrane proteins that play a role in myogenic cell differentiation. Together, CDO and BOC participate in a positive feedback loop with MyoD, a myogenic transcription factor. The 1,242 amino acid rat CDO protein has a 24 residue signal sequence, five Ig V-like repeats, a 25 residue membrane-spanning region, three FNIII-like repeats and a cytoplasmic region of 256 amino acids containing a proline-rich stretch. The human protein contains 1,225 amino acid residues and shares significant homology with the domain structures of the rat protein.

Description:   CDK5 (Cyclin Dependent Kinase 5) is serine/threonine kinase involved in synaptic regulation and neuronal development; phosphorylates synaptic protein Pctaire1; regulates acetylcholine receptor expression. CDK5 is a member of the cyclindependent kinase family of serine/threonine kinases. It is present in numerous mammalian tissues including kidney, testes, and ovary. Its activity is detected almost exclusively in brain extracts. Neuronal and muscle cells contain the highest amount of this protein. Similar to other Cdks, monomeric Cdk5 displays no enzymatic activity, but Cdk5 is not activated by cyclins. Instead, the p35 protein, which is expressed solely in the brain, activates Cdk5. Cdk5 interacts with D1 and D3 type G1 cyclins and can phosphorylate histone H1, TAU, MAP2 and NF-H and NF-M. Cdk5 activity is involved in terminal differentiation of neurons and muscle cells.
