SKI is a nuclear protein that forms homodimers and heterodimers and binds to DNA to function as transcriptional activators and repressors. The SKI oncoprotein dramatically affects cell growth, differentiation, and/or survival. SKI was shown to act in distinct signaling pathways including those involving nuclear receptors, transforming growth factor beta, and tumor suppressors. SKI inhibits transforming growth factor-beta (TGF-beta) signaling through interaction with Smad proteins. SKI represses Smad-mediated transcriptional activation, probably through its action as a transcriptional co-repressor. SKI also inhibits TGF-beta-induced downregulation of genes such as c-myc.

Defects in G6PD are the cause of chronic non-spherocytic hemolytic anaemia (CNSHA) . Deficiency of G6PD is associated with hemolytic anaemia in two different situations. First, in areas in which malaria has been endemic, G6PD-deficiency alleles have reached high frequencies (1% to 50%) and deficient individuals, though essentially asymptomatic in the steady state, have a high risk of acute hemolytic attacks. Secondly, sporadic cases of G6PD deficiency occur at a very low frequencies, and they usually present a more severe phenotype. Several types of CNSHA are recognized. Class-I variants are associated with severe NSHA; class-II have an activity <10% of normal; class-III have an activity of 10% to 60% of normal; class-IV have near normal activity.

This gene encodes one of several forms of glutamic acid decarboxylase, identified as a major autoantigen in insulin-dependent diabetes. The enzyme encoded is responsible for catalyzing the production of gamma aminobutyric acid from L glutamic acid. A pathogenic role for this enzyme has been identified in the human pancreas since it has been identified as an autoantibody and an autoreactive T cell target in insulin dependent diabetes. This gene encodes a selenoprotein, which contains a selenocysteine (Sec) residue at its active site. The selenocysteine is encoded by the UGA codon that normally signals translation termination. The 3' UTR of selenoprotein genes have a common stem-loop structure, the sec insertion sequence (SECIS), that is necessary for the recognition of UGA as a Sec codon rather than as a stop signal. This protein has been shown to function as a glutathione-dependent antioxidant in vivo.
