Description:
Striatin, SG2NA, and zinedin, the three mammalian members of the striatin family, are multimodular, WD-repeat and calmodulin-binding proteins. Zinedin and SG2NA share with striatin identical protein-protein interaction domains and the same overall domain structure. All three proteins are both cytosolic and membrane-bound and bind calmodulin in the presence of calcium. Striatin is a neuronal, intracellular protein strictly expressed in the somatodendritic compartment, including spines, subsets of neurons, and is considered as a marker of neuronal polarity. Downregulation of striatin, which is expressed in a few subsets of neurons, impairs the growth of dendrites as well as rat locomotor activity. Zinedin is mainly expressed in the central nervous system, whereas SG2NA is mainly expressed in the brain and muscle.

Neugrin, also known as NGRN, mesenchymal stem cell protein DSC92, neurite outgrowth-associated protein or spinal cord-derived protein FI58G, is a 291 amino acid protein that plays a role in neuronal differentiation and belongs to the neugrin family. As both a secreted and nuclear protein, neugrin exists as two alternatively spliced isoforms and is highly expressed in skeletal muscle, brain and heart. Neugrin is upregulated in neuroblastostoma cells by retinoic acid treatment and is encoded by a gene that maps to human chromosome 15q26.1. Chromosome 15 houses over 700 genes and comprises nearly 3% of the human genome. Angelman syndrome, Prader-Willi syndrome, Tay-Sachs disease and Marfan syndrome are all associated with defects in chromosome 15-localised genes.

The SHC gene encodes for a signaling and transforming protein containing Src homology 2 and 3 (SH2 and SH3) domains. The SHC gene encodes 2 widely expressed overlapping proteins of 46 and 52 kD, both containing a C-terminal SH2 domain. Adjacent to the SH2 region is a glycine and proline rich region. These 2 proteins differ in their N terminals. SHC proteins are involved in mitogenic signal transduction and act by coupling growth factor receptors to the RAS signaling pathway. The protein encoded by the SHC1 gene is thought to act as an adaptor in many signal transduction pathways.
