GIOT-1 is a 563 amino acid protein belonging to the Krüppel C2H2-type zinc-finger protein family. Localized to the nucleus, GIOT-1 is widely expressed in tissues, with highest levels in liver, kidney, small intestine, pancreas and thymus. GIOT-1 contains 12 C2H2-type zinc fincers and one KRAB domain. Because the KRAB domain functions as a transcriptional repressor when attached to the template DNA, GIOT-1 is thought to be involved in transcriptional regulation. The gene encoding GIOT-1 is localized to chromosome 19q13.12 and two isoforms of GIOT-1 exist as a result of alternative splicing events.

HP1 alpha is a component of heterochromatin. It recognizes and binds histone H3 tails methylated at Lys-9, leading to epigenetic repression. HP1 alpha may interact with lamin B receptor (LBR). This interaction can contribute to the association of the heterochromatin with the inner nuclear membrane. HP1 proteins are relatively small proteins (~25 kDa) with a conserved amino-terminal chromo domain and a structurally related carboxy-terminal motif, the chromo shadow domain. Both domains of HP1 are requiredfor binding to native chromatin in vivo, but they contribute differentially to binding in euchromatin and heterochromatin.

Expressed predominantly in cardiac and skeletal muscle, beta-taxilin binds to the coiled coil region of the syntaxin family members STX1A, STX3A, and STX4A. Beta- and gamma-taxilins, bind to the alpha subunit of the nascent polypeptide-associated complex (NAC) and affect its nuclear distribution, suggesting that the taxilin family is involved not only in the translational process through its interaction with NAC but also in the transcriptional process through its interaction with alphaNAC alone.

ZBED5 contains 1 BED type zinc finger and it may be derived from an ancient transposon that has lost its ability to translocate. This gene is unusual in that its coding sequence is mostly derived from Charlie like DNA transposon. There is mRNA and EST evidence to suggest that this gene is transcribed. The encoded protein shares 70% identity with Charlie 1 transposase, however, this gene does not appear to be an active DNA transposon as it is not flanked by terminal inverted repeats. The exact function of this gene product is not known.

CYP2R1 is a member of the cytochrome P450 superfamily of enzymes. The cytochrome P450 proteins are mono-oxygenases which catalyze many reactions involved in drug metabolism and synthesis of cholesterol, steroids and other lipids. This enzyme is a microsomal vitamin D hydroxylase that converts vitamin D into the active ligand for the vitamin D receptor.Defects in CYP2R1 are a cause of 25-hydroxyvitamin D(3) deficiency, also known as pseudovitamin D(3) deficiency rickets due to 25-hydroxylase deficiency. First described in patients who had rickets at a young age despite a history of adequate vitamin D intake. The patients sera had low calcium concentrations, low phosphate concentrations, elevated alkaline phosphatase activity and low levels of 25-hydroxyvitamin D.

In omnivorous mammals, the short chain fatty acids acetate, propionate and butyrate are produced primarily by the gut microbiome that metabolizes dietary fibers. SCFAs serve as a source of energy but also act as signaling molecules. That G protein-coupled receptor is probably coupled to the pertussis toxin-sensitive, G(i/o)-alpha family of G proteins but also to the Gq family (PubMed:12496283, PubMed:12711604, PubMed:23589301). Its activation results in the formation of inositol 1,4,5-trisphosphate, the mobilization of intracellular calcium, the phosphorylation of the MAPK3/ERK1 and MAPK1/ERK2 kinases and the inhibition of intracellular cAMP accumulation. May play a role in glucose homeostasis by regulating the secretion of GLP-1, in response to short-chain fatty acids accumulating in the intestine. May also regulate the production of LEP/Leptin, a hormone acting on the central nervous system to inhibit food intake. Finally, may also regulate whole-body energy homeostasis through adipogenesis regulating both differentiation and lipid storage of adipocytes. In parallel to its role in energy homeostasis, may also mediate the activation of the inflammatory and immune responses by SCFA in the intestine, regulating the rapid production of chemokines and cytokines. May also play a role in the resolution of the inflammatory response and control chemotaxis in neutrophils. In addition to SCFAs, may also be activated by the extracellular lectin FCN1 in a process leading to activation of monocytes and inducing the secretion of interleukin-8/IL-8 in response to the presence of microbes (PubMed:21037097).

Synapsin I is a member of the synapsin family. Synapsins are neuronal phosphoproteins which associate with the cytoplasmic surface of synaptic vesicles. Family members are characterized by common protein domains, and they are implicated in synaptogenesis and the modulation of neurotransmitter release, suggesting a potential role in several neuropsychiatric diseases. This member of the synapsin family plays a role in regulation of axonogenesis and synaptogenesis. The protein serves as a substrate for several different protein kinases and phosphorylation may function in the regulation of this protein in the nerve terminal. Mutations of the Synapsin I gene may be associated with X linked disorders with primary neuronal degeneration such as Rett syndrome.

Transportin 1 is the beta subunit of the karyopherin receptor complex which interacts with nuclear localization signals to target proteins to the nucleus. The karyopherin receptor complex is a heterodimer of an alpha subunit which recognizes the nuclear localization signal and a beta subunit which docks the complex at nucleoporins. Alternate splicing of this gene results in two transcript variants encoding different proteins. In cases of HIV-1 infections, transportin 1 is known to bind and mediate the nuclear import of HIV-1 Rev.
