eNOS is a calcium/calmodulin dependent enzyme which undergoes several post translational modifications, including acylation with myristate and palmitate, and phosphorylation on numerous residues. As with the other members of the NOS family, eNOS derives the diffusible multifunctional second messenger NO from L arginine through a series of reactions in which L citrulline is an intermediate. eNOS plays an important role in controlling vascular tone, platelet aggregation, and cardiac myocyte function.

DMWD is a 674 amino acid protein containing five WD repeats. DMWD may play a role in the development of mental symptoms in severe cases of myotonic dystrophy, a chronic multisystemic disease characterized by wasting of the muscles, heart conduction defects, cataracts, endocrine changes and myotonia. The DMWD gene is located upstream of the DMPK gene and is prominently expressed in tissues affected in myotonic dystrophy patients. DMWD may also contribute to regulation in meiosis. DMWD is expressed in kidney and spleen, with strongest expression in brain, liver and testis. The gene encoding DMWD maps to human chromosome 19q13.32 and mouse chromosome 7 A3.

Rab17 belongs to the Rab family of small Ras-like GTPases. It is specifically expressed in epithelial cells and is upregulated during cell polarization. Immunofluorescence staining studies indicate that Rab17 is associated with the perinuclear recycling endosome in nonpolarized epithelial cells and with the apical recycling endosome in polarized epithelial cells. The function of Rab17 remains unclear. Reports of Rab17 colocalization with internalized IgA in the apical endosome suggest that it may regulate receptor-mediated transcytosis. Rab17 has also been shown to regulate melanocytic filopodia formation and melanosome trafficking. siRNA knockdown of Rab17 in melanoma cells induces melanosome accumulation in the cell periphery.

The Hox homeobox genes encode proteins that are transcriptional regulators with an established role in embryonic development. HoxA4 (homeobox A4), also known as HOX1D or HOX1, is a 320 amino acid protein that localizes to the nucleus and contains one homeobox DNA-binding domain. Expressed in the embryonic nervous system, HoxA4 functions as a sequence-specific DNA-binding transcription factor that is part of a regulatory mechanism that provides cells with positional identities during development. Via its ability to bind DNA, HoxA4 plays an important role in the regulation of gene expression, as well as morphogenesis and differentiation. The gene encoding HoxA4 maps to human chromosome 7, which houses over 1,000 genes and comprises nearly 5% of the human genome. Defects in some of the genes localized to chromosome 7 have been linked to Osteogenesis imperfecta, Williams-Beuren syndrome, Pendred syndrome, Lissencephaly, Citrullinemia and Shwachman-Diamond syndrome.

Description:
Ataxin-1, also designated spinocerebellar ataxia type 1 protein (Sca-1), is differentially expressed and localizes to both the cytoplasm and the nucleus. Mutations in Ataxin-1 are associated with the onset of the autosomal dominant neurodegenerative disorder spinocerebellar ataxia type 1 (SCA-1), which is characterised by progressive neuronal loss in the cerebellum, muscle wasting and ataxia. In Purkinje cells, where SCA-1 is predominantly observed, Ataxin-1 has been shown to directly associate with the Purkinje-enriched leucine-rich acidic nuclear protein (LANP) and the nuclear matrix-associated protein promyelocytic leukaemia protein PML. In SCA-1, Ataxin-1 is mutated to encode a polyglutamine protein that forms nuclear aggregates, which interact significantly more strongly with LANP and contribute to the pathogenesis of SCA-1.
