Description:   Promotes a prolonged MAP-kinase signaling by neurotrophins through activation of a Rap1-dependent mechanism. Provides a docking site for the CRKL-C3G complex, resulting in Rap1-dependent sustained ERK activation. May play an important role in regulating postsynaptic signal transduction through the syntrophin-mediated localization of receptor tyrosine kinases such as EPHA4. In cooperation with SNTA1 can enhance EPHA4-induced JAK/STAT activation. May play a role in neurotrophin- and ephrin-mediated neuronal outgrowth and in axon guidance during neural development and in neuronal regeneration (By similarity). Modulates stress-induced apoptosis of melanoma cells via regulation of the MEK/ERK signaling pathway.

Description:   STK31 (Serine/threonine protein kinase 31) is similar to a mouse gene that encodes a putative protein kinase with a tudor domain, and shows testis specific expression. Alternative splicing results in multiple transcript variants encoding different isoforms. Protein kinases are enzymes that transfer a phosphate group from a phosphate donor, generally the g phosphate of ATP, onto an acceptor amino acid in a substrate protein. By this basic mechanism, protein kinases mediate most of the signal transduction in eukaryotic cells, regulating cellular metabolism, transcription, cell cycle progression, cytoskeletal rearrangement and cell movement, apoptosis, and differentiation. With more than 500 gene products, the protein kinase family is one of the largest families of proteins in eukaryotes.

Description:   Syntaxins, a family of proteins involved in the fusion of synaptic vesicles with the plasma membrane, display broad tissue distribution and contain carboxy-terminal hydrophobic domains that direct themselves to their respective intracellular compartments. Synaptin 6 is a 255 amino acid protein that is widely expressed, with higher expression levels in brain, lung and kidney. This synaptin co-localizes with vesicle associated membrane protein (VAMP) 4 to tubular and vesicular membranes of the Golgi apparatus. The cytosolic domain of Syntaxin 6 reduces the rate on Glut4 reinternalization upon insulin withdrawl and is involved in a memrane-trafficking process that removes Glut4 from traffic directed to the plasma membrane. Syntaxin 6 is upregulated in activated macrophages in conjunction with an increase in the secretion of cytokines. The delivery of microdomain-associated lipids and proteins to the cell surface is regulated by Syntaxin 6.

Description:   In eukaryotes, the phosphorylation and dephosphorylation of proteins on serine and threonine residues is an essential means of regulating a broad range of cellular functions including division, homeostasis and apoptosis. A group of proteins that are intimately involved in this process are the protein phosphatases. In general, the protein phosphatase (PP) holoenzyme is a trimeric complex composed of a regulatory subunit, a variable subunit and a catalytic subunit. Four major families of protein phosphatase catalytic subunit have been identified, designated PP1, PP2A, PP2B and PP2C. An additional protein phosphatase catalytic subunit, PPX (also known as PP4), is a putative member of a novel PP family. The PP2B family comprises subfamily members PP2B-A alpha, PP2B-A Beta and PP2B-A Gamma. Two additional regulatory subunits been identified, designated PP2B-B1 and PP2B-B2.

Description:   RecQ helicases are implicated in DNA recombination and repair helicases and are critical in genome maintenance. RecQL5 is one of five known RecQ homologues in humans, and defects in three of these (BLM, WRN and RecQ4)are associated with autosomal recessive disorders that involve genomic instability and a predisposition to cancer. The exact role of ReqL5 is unclear. It has been shown in mouse embryonic stem (ES) cells that mutations in the Recql5 genes result in a significant increase in the frequency of sister chromatid exchange (SCE), whereas deleting Recql5 lead to an even higher frequency of SCE. There are three different RecQL5 isoforms - alpha, beta and gamma, produced by alternative splicing.

Description:   Adaptins are heterotetrameric subunits of adaptors, which are complexes involved in the formation of Clathrin-coated pits for vesicle-mediated endocytosis. Clathrin and its associated heterotetrameric protein complexes make up the main protein components of the coat surrounding the cytoplasmic face of coated vesicles. The Adaptin family, comprising a, b, and g classes, is also responsible for the transport of ligand-receptor complexes from plasma membranes and the trans-Golgi network to lysosomes. Two main types of adaptor proteins (APs), AP-1 and AP-2, are found in Clathrin-coated structures located at the Golgi complex and the plasma membrane of mammalian cells, respectively. Adaptor protein complex 2 (AP-2) is composed of two large Adaptins (a1A/AP2A1 and b1/AP2B1), a medium Adaptin (m2/AP-2m1) and a small Adaptin (s2 long/AP2S1). AP-2m1, a 435 amino acid protein, links Clathrin to receptors in coated vesicles.

Description:   MP21 is a member of the EMP24/GP25L/p24 cargo family of proteins that regulates vesicular trafficking in the early secretory pathway. TMP21 is a ubiquitously expressed single-pass type I membrane protein localizing to the Golgi cisternae and the plasma membrane. It contains one GOLD (Golgi dynamics) domain and participates in protein transport and quality control between the endoplasmic reticulum (ER) and the Golgi complex. In addition, TMP21 is a component of the heteromeric secretase complex (or Presenilin complex) and functions to regulate the ?secretase activity. Two isoforms, namely TMP21-I and TMP21-II, exist for this protein.

Description:   NEPH2 is a 778 amino acid single-pass type I membrane protein that belongs to the nephrin-like protein family and immunoglobulin superfamily. Expressed in both fetal and adult brain, as well as podocytes of kidney glomeruli, NEPH2 contains five Ig-like C2-type (immunoglobulin-like) domains and is thought to plaly a role in the hematopoetic supportive capacity of stroma cells. NEPH2 undergoes alternative splicing to produce two isoforms and contains a C-terminal cytoplasmic domain which it uses to interact with Podocin, a podocyte protein involved in ultrafiltration. Defects in the gene encoding NEPH2 are associated with mental retardation autosomal dominant type 4 (MRD4).

Description:   The isthmic organizer signals at the mid/hindbrain boundary (MHB) regulate the development and differentiation of the vertebrate caudal midbrain and the anterior hindbrain. The MHB forms at the boundary of expression between homeobox genes Gbx2 and Otx2. Gbx2 and Otx2 play distinct, essential roles in MHB positioning and development. During development, the GBX2 gene is expressed in the anterior hindbrain. Specifically, Gbx2 negatively regulates Otx2 expression along the anterior-posterior axis; Gbx2(-) mutants demonstrate an expanded Otx2 domain. During development, the GBX2 gene is expressed in the anterior hindbrain. Gbx2 is expressed in the adult brain, spleen and female genital tract. The GBX2 gene is over-expressed in human prostate cancer cell lines (TSU-prl, PC3, DU145 and LNCaP). Furthermore, downregulation of Gbx2 expression restricts tumorigenicity in human prostate cancer cell lines, which suggests that Gbx2 expression may be required for growth of malignant prostate cells.
