Description:
Ubiquitin: Exists either covalently attached to another protein, or free (unanchored). When covalently bound, it is conjugated to target proteins via an isopeptide bond either as a monomer (monoubiquitin), a polymer linked via different Lys residues of the ubiquitin (polyubiquitin chains) or a linear polymer linked via the initiator Met of the ubiquitin (linear polyubiquitin chains). Polyubiquitin chains, when attached to a target protein, have different functions depending on the Lys residue of the ubiquitin that is linked: Lys-6-linked may be involved in DNA repair; Lys-11-linked is involved in ERAD (endoplasmic reticulum-associated degradation) and in cell-cycle regulation; Lys-29-linked is involved in lysosomal degradation; Lys-33-linked is involved in kinase modification; Lys-48-linked is involved in protein degradation via the proteasome; Lys-63-linked is involved in endocytosis, DNA-damage responses as well as in signaling processes leading to activation of the transcription factor NF-kappa-B. Linear polymer chains formed via attachment by the initiator Met lead to cell signaling. Ubiquitin is usually conjugated to Lys residues of target proteins, however, in rare cases, conjugation to Cys or Ser residues has been observed. When polyubiquitin is free (unanchored-polyubiquitin), it also has distinct roles, such as in activation of protein kinases, and in signaling.

BAG5 is a member of the BAG1 related protein family. BAG1 is an anti apoptotic protein that functions through interactions with a variety of cell apoptosis and growth related proteins including BCL 2, Raf protein kinase, steroid hormone receptors, growth factor receptors and members of the heat shock protein 70 kDa family. A BAG domain near the C terminus, may bind and inhibit the chaperone activity of Hsc70/Hsp70. It has been hypothesized that the BAG5 protein will induce the death of nigral neurons through its predicted interaction with hsp70, which will cause increased protein aggregation and cell death by disinhibition of hsp70’s anti apoptotic function. It is believed that BAG5 will play an important role in the mechanisms of neuronal death. BAG5 may also be of interest due to its possible role as a modulator of the hsp70/hsp40 chaperone axis or its possible interaction and coordination of localization/modulation of other BAG containing proteins via BAG-BAGheterodimerization.

Chromosome 22 contains over 500 genes and about 49 million bases. Being the second smallest human chromosome, 22 contains a surprising variety of interesting genes. Phelan-McDermid syndrome, neurofibromatosis type 2 and autism are associated with chromosome 22. A schizophrenia susceptibility locus has been identified on chromosome 22 and studies show that 22q11 deletion symptoms include a high incidence of schizophrenia. Translo-acations between chromosomes 9 and 22 may lead to the formation of the Philadelphia chromosome and the subsequent production of the novel fusion protein, Bcr-Abl, a potent cell proliferation activator found in several types of leukemia. The C22orf26 gene product has been provisionally designated C22orf26 pending further characterization.
