Description:   This product is prepared by labeling highly cross-adsorbed donkey anti-rat IgG (H+L) with CF™405S. CF™405S is a blue fluorescent dye with an absorption peak wavelength that nearly coincides with the 405 nm blue diode laser line. In addition, the emission peak wavelength of the dye well centers within the blue detection window of BD flow cytometers. As a result, flow cytometry analysis using CF™405S dye results in much brighter signal than that using Alexa Fluor® 405.   To minimize cross-reactivity, the antibody has been adsorbed against bovine, chicken, goat, guinea pig, Syrian hamster, horse, human, mouse, rabbit, and sheep serum proteins .

Description:   This MAb is specific to progesterone receptor and shows minimal cross-reaction with other members of the family. Progesterone receptor is expressed as two major isoforms, PR-A (81 kDa) and PR-B (116 kDa). Expression of PgR has been suggested to reflect a intact estrogen regulatory machinery and therefore, predict better clinical response to endocrine therapy than ER alone. It is excellent for immunohistochemical staining of formalin/paraffin tissues.

Description:   Recognizes a 53 kDa protein, which is identified as p53 suppressor gene product. It reacts with the mutant as well as the wild form of p53 under denaturing and non-denaturing conditions. p53 is a tumor suppressor gene expressed in a wide variety of tissue types and is involved in regulating cell growth, replication, and apoptosis. It binds to MDM2, SV40 T antigen and human papilloma virus E6 protein. Positive nuclear staining with p53 antibody has been reported to be a negative prognostic factor in breast carcinoma, lung carcinoma, colorectal, and urothelial carcinoma. Anti-p53 positivity has also been used to differentiate uterine serous carcinoma from endometrioid carcinoma as well as to detect intratubular germ cell neoplasia. Mutations involving p53 are found in a wide variety of malignant tumors, including breast, ovarian, bladder, colon, lung, and melanoma.

Description:   This MAb reacts with a protein of 20-30 kDa, identified as PGP9.5, also known as ubiquitin carboxyl-terminal hydrolase-1 (UchL1). Initially, PGP9.5 expression in normal tissues was reported in neurons and neuroendocrine cells but later it was found in distal renal tubular epithelium, spermatogonia, Leydig cells, oocytes, melanocytes, prostatic secretory epithelium, ejaculatory duct cells, epididymis, mammary epithelial cells, Merkel cells, and dermal fibroblasts. Furthermore, immunostaining for PGP9.5 has been shown in a wide variety of mesenchymal neoplasms as well. A mutation in PGP9.5 gene is believed to cause a form of Parkinson's disease.

Description:   Recognizes a glycoprotein of 110 kDa, identified as CD26 (Workshop VI; Code: N-L039). It is an atypical serine protease belonging to the prolyl oligopeptidase family. It is expressed on lymphocyte cells and is upregulated during T-cell activation. CD26 is also expressed on activated B cells and natural killer cells and abundantly on epithelia. CD26 is implicated in a variety of biological functions including T-cell activation, cell adhesion with extracellular matrix such as fibronectin or collagens, and in HIV infection. Cross-linking of CD26 using this antibody dramatically enhances the anti-CD3-induced IL-2 production. In Western blotting, this MAb reacts with only glycosylated CD26, but not with the deglycosylated form. It does not prevent ADA binding to CD26.

Description:   The human CD146 gene maps to chromosome 11q23 and encodes a trans-membrane glycoprotein, also designated MCAM, MUC 18, that belongs to the immunoglobulin superfamily and functions as a Ca2 -independent cell adhesion molecule. CD146 expression is restricted to advanced primary and metastatic melanomas and to cell lines of the neuroectodermal lineage, but not normal melanocytes. CD146 is found on 80% of advanced primary human melanomas and correlates well with development of metastatic disease.

Description:   This antibody recognizes a transcription factor of 64-67 kDa, identified as c-myc. This MAb shows no cross-reaction with v-myc. c-myc is involved in the control of cell proliferation and differentiation and is amplified and/or over-expressed in a variety of tumors. Over-expression of c-myc protein occurs frequently in luminal cells of prostate intraepithelial neoplasia as well as in most primary carcinomas and metastatic disease. Rearrangement of the MYC gene is found in 3% to 16% of diffuse large B-cell lymphoma (DLBCL's) and in nearly 100% of Burkitt lymphomas (BL). Identifying MYC status is important in establishing final diagnosis of DLBCL, BL, or B-cell lymphoma, with features intermediate between DLBCL and BL as well as in differential diagnoses of the lymphomas.

Description:   This MAb recognizes full-length MUC1 in a glycosylation-independent manner and can bind to the fully glycosylated protein. The dominant epitope of this MAb is APDTR in the VNTR region. It reacts with the core peptide of the MUC1 protein, which is a member of a family of mucin glycoproteins that are characterized by high carbohydrate content, O-linked oligosaccharides, high molecular weight (>200 kDa) and an amino acid composition rich in serine, threonine, proline and glycine. The core protein contains a domain of 20 amino-acid tandem repeats that functions as multiple epitopes for the MAb. Incomplete glycosylation of some tumor-associated mucins may lead to variable unmasking of the multiple peptide epitopes leading to the observed differences in staining intensity between normal and malignant tissues. This MAb reacts with both normal and malignant epithelia of various tissues including breast and colon.

Description:   This antibody recognizes actin of skeletal, cardiac, and smooth muscle cells. It is not reactive with other mesenchymal cells except for myoepithelium. Actin can be resolved on the basis of its isoelectric points into three distinctive components: alpha, beta, and gamma in order of increasing isoelectric point. Anti-muscle specific actin recognizes alpha and gamma isotypes of all muscle groups. Non-muscle cells such as vascular endothelial cells and connective tissues are non-reactive. Also, neoplastic cells of non-muscle-derived tissue such as carcinomas, melanomas, and lymphomas are negative.It stains tumors of smooth muscle (leiomyomas and leiomyosarcomas) as well as skeletal muscle (rhabdomyomas and rhabdomyosarcomas).

Description:   CD31 (PECAM-1) is a transmembrane glycoprotein member of the immunoglobulin supergene family of adhesion molecules. CD31 is expressed by stem cells of the hematopoietic system and is primarily used to identify and concentrate these cells for experimental studies as well as for bone marrow transplantation. Anti-CD31 has shown to be highly specific and sensitive for vascular endothelial cells. Staining of nonvascular tumors (excluding hematopoietic neoplasms) is rare. CD31 MAb reacts with normal, benign, and malignant endothelial cells which make up blood vessel lining. The level of CD31 expression can help to determine the degree of tumor angiogenesis, and a high level of CD31 expression may imply a rapidly growing tumor and potentially a predictor of tumor recurrence.

Description:   This MAb recognizes a single chain, transmembrane, heavily glycosylated protein of 90-120 kDa, which is identified as CD34. Its expression is a hallmark for identifying pluripotent hematopoietic stem or progenitor cells. Its expression is gradually lost as lineage committed progenitors differentiate. CD34 is a marker of choice for staining blasts in acute myeloid leukemia. In addition, CD34 is expressed by soft tissue tumors, such as solitary fibrous tumor and gastrointestinal stromal tumor. Its expression is also found in vascular endothelium. Additionally, it appears that proliferating endothelial cells express this molecule more than the non-proliferating endothelial cells. Anti-CD34 labels > 85% of angiosarcoma and Kaposi's sarcoma, but with a lower specificity.

Description:   This antibody recognizes CD16 (FcγRIII), the low-affinity receptor for IgG with an apparent molecular weight of 50-80 kDa. Two similar genes represent CD16, CD16A (FcγRIIIA), which exists as a hetero-oligomeric polypeptide-anchored form in macrophages and NK cells and CD16B (FcγRIIIB), which exist as a monomeric GPI-anchored form in neutrophils. Furthermore, there are two known polymorphisms of CD16B, NA-1 and NA-2. Individuals homozygous for NA-2 show a lower phagocytic capacity compared with NA-1. CD16 binds IgG in the form of immune complexes and shows preferential binding of IgG1 and IgG3 isotypes and minimal binding of IgG2 and IgG4. Upon IgG binding, both CD16 isoforms initiate signal transduction cascades that lead to a variety of responses including antibody-dependent cell-mediated cytotoxicity (ADCC), phagocytosis, degranulation and proliferation.

Description:   Recognizes a protein of 50-65 kDa, identified as CD16 (Workshop IV; Code N39 ) (also known low affinity Fc receptor III for IgG (FcRIII) or Leu 11). CD16 exists as a polypepetide-anchored from (FCRIIIA or CD16A) on human natural killer (NK) cells and monocytes/ macrophages and as a glycosylphosphatidylinositol (GPI)-anchored form (FcRIIIB or CD16B) on neutrophils. CD16B is polymorphic and the two alleles are termed NA1 and NA2.3 CD16 plays a role in signal transduction, NK cell activation and antibody-dependent cellular cytotoxicity. This MAb has been showed to inhibit the binding of immune complex to NK cells, inhibit cytotoxicity of NK cells, and induce calcium fluxes in NK cells and neutrophils.
