This antibody recognises a protein of 145 kDa, identified as CD117, or p145kit. It is found on a wide variety of tumor cells including follicular and papillary carcinoma of thyroid, adenocarcinomas from endometrium, lung, ovary, pancreas, and breast as well as malignant melanoma, endodermal sinus tumor, and small cell carcinoma. However, CD117 antibody has been particularly useful in differentiating gastrointestinal stromal tumors from Kaposi’s sarcoma, tumors of smooth muscle origin, fibromatosis, and neural tumors of the GI tract. CD117 antibody is also useful in recognizing myeloblasts in bone marrow biopsy and clot section.

This cytokeratin 18 antibody reacts with a wide variety of simple epithelia. It does not react with stratified squamous epithelia. It reacts with epithelial tumors of the gastrointestinal tract, lung, breast, pancreas, ovary, and thyroid. Cytokeratin 18, which belongs to the type A (acidic) subfamily of low molecular weight keratins, exists in combination with cytokeratin 8. It is reported that tissues from gastrointestinal tract are positive for both cytokeratin 18 and 8 but do not contain cytokeratin 14. Tissues from gastrointestinal tract, respiratory tract and urogenital tract, as well as endocrine and exocrine tissues and mesothelial cells are positive for cytokeratin 18.

This antibody is specific to the kappa light chain of immunoglobulin and shows no cross-reaction with lambda light chain or any of the five heavy chains. In mammals, the two light chains in an antibody are always identical, with only one type of light chain, kappa or lambda. In general, the ratio of Kappa to Lambda is 3:1. However, with the occurrence of multiple myeloma or other B-cell malignancies this ratio is disturbed. Antibody to the kappa light chain is reportedly useful in the identification of leukemias, plasmacytomas, and certain non-Hodgkin's lymphomas. Demonstration of clonality in lymphoid infiltrates indicates that the infiltrate is malignant.

<p>Immunoglobulin gamma (IgG) is the most common class of antibody in blood and extracellular fluid. Approximately 75% of serum antibodies in humans are IgG. There are four immunoglobulin gamma subclasses: one, two, three and four. IgG1 is the most common, with 68% of all gamma class antibodies being G1, and G4 is the least common at 4%. Gamma class antibodies are found primarily in the secondary immune response, class switching from IgM and IgD. They are the only class of antibody that can cross the placenta, and along with IgA secreted in breast milk, provide the neonate with humoral immunity before immune system development occurs.<p>This antibody recognises a protein of 75 kDa identified as the gamma heavy chain of human immunoglobulins. It does not cross-react with alpha, mu, epsilon, or delta heavy chains, T-cells, monocytes, granulocytes, or erythrocytes. The IgG antibody is useful in the identification of leukemias, plasmacytomas, and certain non-Hodgkin's lymphomas. The most common feature of these malignancies is the restricted expression of a single heavy chain class. Demonstration of clonality in lymphoid infiltrates indicates that the infiltrate is clonal and therefore malignant.

This antibody recognises a polypeptide which is identified as insulin, a 51-amino acid polypeptide composed of A and B chains connected through the C-peptide. Proinsulin, which has very little biological activity, is cleaved by proteases within its cell of origin into the insulin molecule and the C-terminal basic residue. Insulin enhances membrane transport of glucose, amino acids, and certain ions. It also promotes glycogen storage, formation of triglycerides, and synthesis of proteins and nucleic acids. Deficiency of insulin results in diabetes mellitus. The main storage site for insulin is the pancreatic islets. antibody to insulin is an important beta-cell and insulinoma marker.
