Inositol 1,4,5-triphosphate (IP3) functions as a second messenger for a myriad of extracellular stimuli including hormones, growth factors and neurotransmitters. Receptor tyrosine kinases indirectly increase the intracellular levels of IP3 through the activation of phospholipases such as phospholipase C (PLC), which convert phosphatidylinositol-4,5 bisphosphate into IP3 and diacylglycerol (DAG). The inositol 1,4,5-triphosphate receptor, IP3R, acts as an inositol triphosphate (IP3)-gated calcium release channel in a variety of cell types. Three IP3 receptor subtypes have been described and are designated IP3R-I, IP3R-II and IP3R-III. IP3R-I is the predominant IP3R subtype expressed in neuronal tissues and the central nervous system, but is also expressed at high levels in the liver.

Serum response factor (SRF) is a transcription factor that binds the serum response element (SRE), a sequence that mediates the transient response of many cellular genes to growth stimulation. SRF-binding sites are also constitutive promoter elements in many muscle-specific promoters. At the c-Fos SRE, formation of a ternary complex containing SRF and its accessory protein p62TCF appears to be important for signal transduction. Two related Ets domain proteins, Elk-1 and SRF accessory protein-1 (SAP-1), have DNA binding properties identical to that of p62TCF. Elk-1 and SAP-1 contain two homologous regions of which the two amino terminal regions, the Ets domain (box A) and the B box, mediate ternary complex formation with SRF. The third homologous region, the C box located toward the C-terminus of the proteins, contains conserved consensus phosphorylation sites for MAP kinases.

This protein is found to possess 3' to 5' exonuclease activity, which may contribute to its role in sensing and repairing DNA damage. It forms a checkpoint protein complex with RAD1 and HUS1. This complex is recruited by checkpoint protein RAD17 to the sites of DNA damage, which is thought to be important for triggering the checkpoint-signaling cascade. Binds to the Q promoter (Qp) of EBV nuclear antigen 1 a (EBNA1) and may play a role in the regulation of EBV latency. Chromosome 1 is the largest human chromosome spanning about 260 million base pairs and making up 8% of the human genome. There are about 3,000 genes on chromosome 1, and considering the great number of genes there are also a large number of diseases associated with chromosome 1. Notably, the rare aging disease Hutchinson-Gilford progeria is associated with the LMNA gene which encodes lamin A. When defective, the LMNA gene product can build up in the nucleus and cause characteristic nuclear blebs. The mechanism of rapidly enhanced aging is unclear and is a topic of continuing exploration. The MUTYH gene is located on chromosome 1 and is partially responsible for familial adenomatous polyposis. Stickler syndrome, Parkinsons, Gaucher disease and Usher syndrome are also associated with chromosome 1. A breakpoint has been identified in 1q which disrupts the DISC1 gene and is linked to schizophrenia. Aberrations in chromosome 1 are found in a variety of cancers including head and neck cancer, malignant melanoma and multiple myeloma. The C1orf173 gene product has been provisionally designated C1orf173 pending further characterization. There are three isoforms of C1orf173 that are produced as a result of alternative splicing events.

MSK1 is a mitogen and stress activated protein kinase 1 which belongs to the AGC family of kinases and is related in structure to the ribosomal p70 S6 kinase subfamily. MSK1 can be activated by ERK1/2 and SAPK2/p38 MAP kinase. It is also known to be required for the phosphorylation of CREB, ATF1 H3 and HMG14 in response to mitogen and stress. Similar to RSK, MSK1 contains two kinase domains (N term and a C term). Once phosphorylated on Thr581 and Ser360 by ERK1/2 and SAPK2/p38, MSK1 autophosphorylate on at least 5 sites. Of these autophosphorylation sites Ser212 and Ser376 get phosphorylated by the C terminal kinase domain of MSK1 which is essential for the catalytic activity of the N terminal kinase domain. MSK2 plays an essential role in the control of RELA transcriptional activity in response to TNF. Phosphorylates 'Ser-10' of histone H3 in response to mitogenics, stress stimuli and EGF, which results in the transcriptional activation of several immediate early genes, including proto-oncogenes c-fos/FOS and c-jun/JUN. May also phosphorylate 'Ser-28' of histone H3. Mediates the mitogen- and stress-induced phosphorylation of high mobility group protein 1 (HMGN1/HMG14). In lipopolysaccharide-stimulated primary macrophages, acts downstream of the Toll-like receptor TLR4 to limit the production of pro-inflammatory cytokines. Functions probably by inducing transcription of the MAP kinase phosphatase DUSP1 and the anti-inflammatory cytokine interleukin 10 (IL10), via CREB1 and ATF1 transcription factors.

May counteract a default induction of apoptosis in G2/M phase. The acetylated form represses STAT3 transactivation of target gene promoters. May play a role in neoplasia. Isoform 3 shows a marked reduction in its anti-apoptotic effects when compared with the displayed wild-type isoform.

The PSD-95/SAP 90 family of proteins, which are known to bind to and cluster various membrane proteins, are involved in the organization of synaptic structure. These proteins are physically and functionally linked to cytoskeletal and/or signaling proteins. CRIPT (for cysteine-rich interactor of PDZ three), a novel postsynaptic protein, binds specifically to the PDZ3 domain of PSD-95/SAP 90. CRIPT induces the recruitment of PSD-95/SAP 90 to microtubules, and it has been shown to bind directly to microtubules, indicating that it may be responsible for cytoskeletal anchoring of PSD-95/SAP 90. CRIPT is widely expressed outside of the brain and is highly conserved from animals to plants suggesting a wider role in regulating cytoskeleton-membrane associations.
