Angiotensin-Converting Enzyme 2 (ACE-2) is an integral membrane protein and a zinc metalloprotease of the ACE family, the ACE family includes somatic and germinal ACE. ACE-2 cleaves angiotensins I and II as a carboxypeptidase, ACE-2 converts angiotensin I to angiotensin 1-9, and angiotensin II to angiotensin 1-7. ACE-2 is also able to hydrolyze apelin-13 and dynorphin-13 with high efficiency. ACE-2 can be high expressed in testis, kidney and heart, in colon, small intestine and ovary at moderate levels. Captopril and lisinopril as the classical ACE inhibitor don’t inhibit ACE-2 activity. ACE-2 may play an important role in regulating the heart function.

emaphorin-3A is a secreted protein which belongs to the semaphorin family. Semaphorins are a family of secreted and cell-bound signaling molecules defined by the presence of a common 500 aa Sema domain.This protein can function as either a chemorepulsive agent, inhibiting axonal outgrowth, or as a chemoattractive agent, stimulating the growth of apical dendrites. In both cases, the protein is vital for normal neuronal pattern development. Increased expression of this protein is associated with schizophrenia and is seen in a variety of human tumor cell lines.

Mouse Betacellulin is a single type I membrane protein which belongs to the EGF family of cytokines. EGF family has many members including EGF, TGF-a, Amphiregulin, HB-EGF, Epiregulin, Tomoregulin and the Neuregulins. Betacellulin is characterised by a six-cysteine consensus motif that forms three intra-molecular disulfide bonds crucial for binding the ErbB receptor family. Betacellulin is expressed in several tissues and tumor cells including kidney, uterus, liver, pancreas and small intestine. Betacellulin binds and activates ErbB-1 and ErbB-4 homodimers. Betacellulin is thought to play a role in the differentiation of pancreatic beta cells.Human and mouse mature BTC protein are 80% identical at the amino acid sequence level. Betacellulin is involved in many biological processes such as stimulating gastrointestinal growth. It is proteolytically processed from a larger membrane-anchored precursor and is a potent mitogen for a wide variety of cell types.

Interleukin-2 receptor subunit alpha (IL2RA) is also known as IL-2R subunit alpha, IL-2-RA, IL2-RA, TAC antigen, p55, CD antigen CD25, is a type I transmembrane glycoprotein. IL2RA is expressed on activated T cells and regulatory T cells, and is capable of binding IL2 with low affinity by itself. However, a ligand-induced high affinity heterotrimeric receptor complex is produced when IL2RA is associated non-covelently with the IL2 receptor beta and gamma chain, and subsequently initiates the intacellular signal pathways such as MAPK or JAK/STAT. On dendritic cells (DC), CD25 has been previously regarded as an activation marker, while both murine and human DC can express CD25, they do not express the beta-chain of the IL-2 receptor, which is indispensable for the execution of IL-2 signaling.

CD152 and CD28, together with their ligands B7-1 and B7-2, constitute one of the dominant costimulatory pathways that regulate T and B cell responses. CD152 and CD28 are structurally homologous molecules that are members of the immunoglobulin (Ig) gene superfamily. Both CD152 and CD28 are composed of a single Ig V-like extracellular domain, a transmembrane domain and an intracellular domain. CD152 and CD28 are both expressed on the cell surface as disulphide-linked homodimers or as monomers. CD152 was originally identified as a gene that was specifically expressed by cytotoxic T lymphocytes. However, CD152 transcripts have since been found in both Th1 and Th2, and CD4+ and CD8+ T cell clones. Whereas, CD28 expression is constitutive on the surfaces of 95% of CD4+ T cells and 50% of CD8+ T cells and is down regulated upon T cell activation, CD152 expression is upregulated rapidly following T cell activation and peaks approximately 24 hours following activation. Although both CD152 and CD28 can bind to the same ligands, CD152 binds to B71 and B72 with 20-100-fold higher affinity than CD28.

CD134 (OX40) is a T cell activation antigen structurally belonging to a lymphocyte-specific subgroup of the nerve growth factor and tumour necrosis factor receptor superfamily, which also includes the T cell antigen CD27, B cell antigen CD40, FAS antigen and the T cell activation antigen 4-1BB. The human CD134 protein is expressed only on activated CD4 + T blasts, and its ligand has been identified as gp34. Interactions between CD134 and its ligand in vivo are necessary for the differentiation of activated B cells into highly immunoglobulin-producing cells, however not involved in other pathways of antigen-driven differentiation of B cells such as development of memory cells in the germinal centers. In addition, the CD134 and gp34 system directly mediate adhesion of activated T cells to vascular endothelial cells, and contribute to growth stimulation of the virus-infected T cells.

CD83 is expressed by activated lymphocytes, Langerhans cells and interdigitating reticulum cells. It contains one Ig-like V-type (immunoglobulin-like) domain., the soluble CD83 has the opposite effect and has an immune inhibitory capacity. T cell immunoglobulin and mucin domain-3 (TIM3), also called hepatitis A virus cellular receptor 2 (HAVCR2), is a transmembrane glycoprotein of the TIM family of immune regulating molecules and plays an important role in the Th1-mediated immune response. TIM3 is expressed on the Th1 cells, CD8 T-cells, monocytes, and dendritic cells, but not on Th2 cells. TIM3 expressed by monocytes and dendritic cells facilitates phagocytosis of apoptotic cells and up-regulates cross-presentation of apoptotic cell-associated antigens through interaction with phosphatidylserine. Engagement of TIM3 by its ligand galectin-9 induces a range of immunosuppressive functions which enhance immune tolerance and inhibit anti-tumour immunity. Stimulation of TIM3 with an agonistic antibody promotes inflammation through the activation of innate immune cells. TIM3 is also regarded as a potential target molecule for immunotherapy. TIM3 and programmed cell death 1 (PD-1) as two important coinhibitory regulators of T cell responses, have been implicated with the T-cell dysfunction or exhaustion associated with chronic HBV infection including HBV-related HCC.

Amiloride-sensitive amine oxidase (AOC1) belongs to the copper/topaquinone oxidase family. The protein exists as homodimer by disulfide and mainly located in placenta and kidney. AOC1 catalyses the degradation of compounds such as putrescine, histamine, spermine, and spermidine, substances involved in allergic and immune responses, cell proliferation, tissue differentiation, tumour formation, and possibly apoptosis. Placental DAO is thought to play a role in the regulation of the female reproductive function. The activity of this protein can be inhibited by amiloride in a competitive manner. It is inhibited by amiloride, a diuretic that acts by closing epithelial sodium ion channels.
