Omschrijving:   Cell adhesion molecule-related/down-regulated by oncogenes (CDO) and BOC (brother of CDO) are members of the immunoglobulin/fibronectin type III repeat family and act as cell surface receptors. CDO is a component of a cell-surface receptor complex which also contains BOC, NEO1, CTNNB1 and cadherins and which acts as a mediator of cell-cell interactions between muscle cells. CDO and BOC are single pass membrane proteins that play a role in myogenic cell differentiation. Together, CDO and BOC participate in a positive feedback loop with MyoD, a myogenic transcription factor. The 1,242 amino acid rat CDO protein has a 24 residue signal sequence, five Ig V-like repeats, a 25 residue membrane-spanning region, three FNIII-like repeats and a cytoplasmic region of 256 amino acids containing a proline-rich stretch. The human protein contains 1,225 amino acid residues and shares significant homology with the domain structures of the rat protein.

In human and mouse there are 5 protein-coding genes which comprise the nucleosome assembly protein (NAP) family. NAP1L1 (NAP1) and NAP1L4 (NAP2) are ubiquitously expressed family members which have been the most extensively studied. The remaining three family members, NAP1L2, NAP1L3 and NAP1L5 are neuron-specific nucleosome assembly proteins translated from intronless genes which are monoallelically expressed. NAP1L2 (nucleosome assembly protein 1-like 2), also known as BPX (brain specific protein, X-linked), is a 460 amino acid protein containing acidic domains which are thought to mediate histone interactions. NAP1L2 binds to chromatin and interacts with Histones H3 and H4. The function of NAP1L2 is not clearly defined although evidence suggests that NAP1L2 influences histone acetylation and therefore may play a significant role in regulating transcription in developing neurons.

DNA repair is initiated by MutS alpha (MSH2-MSH6) or MutS beta (MSH2-MSH6) binding to a dsDNA mismatch, then MutL alpha is recruited to the heteroduplex. Assembly of the MutL-MutS-heteroduplex ternary complex in presence of RFC and PCNA is sufficient to activate endonuclease activity of PMS2. It introduces single-strand breaks near the mismatch and thus generates new entry points for the exonuclease EXO1 to degrade the strand containing the mismatch. DNA methylation would prevent cleavage and therefore assure that only the newly mutated DNA strand is going to be corrected. MutL alpha (MLH1-PMS2) interacts physically with the clamp loader subunits of DNA polymerase III, suggesting that it may play a role to recruit the DNA polymerase III to the site of the MMR. Also implicated in DNA damage signaling, a process which induces cell cycle arrest and can lead to apoptosis in case of major DNA damages. Heterodimerizes with MLH3 to form MutL gamma which plays a role in meiosis.
