Protocadherins are a large family of cadherin-like cell adhesion proteins that are involved in the establishment and maintenance of neuronal connections in the brain. There are three protocadherin gene clusters designated alpha, beta and gamma, all of which contain multiple tandemly arranged genes. The protocadherein gamma cluster consists of three subfamilies (A, B and C). As a member of the gamma subfamily B, PCDHGB1 (Protocadherin gamma B1) is a 927 amino acid protein that is one of 22 proteins encoded by the protocadherin gamma cluster. Typical of gamma protocadherins, PCDHGB1 contains six cadherin motifs and is a type I transmembrane receptor expressed in the central nervous system. With localization to synapses, members of the gamma cluster of protocadherins are essential for neuronal survival. There are two isoforms of PCDHGB1 that are produced as a result of alternative splicing events.

Plays an important role in the cascades of cellular responses evoked by changes in the environment. Once activated, acts as an upstream activator of the MKK/JNK signal transduction cascade and the p38 MAPK signal transduction cascade through the phosphorylation and activation of several MAP kinase kinases like MAP2K1/MEK1, MAP2K3/MKK3, MAP2K6/MKK6 and MAP2K7/MKK7. These MAP2Ks in turn activate p38 MAPKs, c-jun N-terminal kinases (JNKs) and I-kappa-B kinase complex (IKK). Both p38 MAPK and JNK pathways control the transcription factors activator protein-1 (AP-1), while nuclear factor-kappa B is activated by IKK. MAP3K7 activates also IKBKB and MAPK8/JNK1 in response to TRAF6 signaling and mediates BMP2-induced apoptosis. In osmotic stress signaling, plays a major role in the activation of MAPK8/JNK1, but not that of NF-kappa-B. Promotes TRIM5 capsid-specific restriction activity.

MLC1 is a 377 amino acid multi-pass membrane protein that may serve as a non-selective neuronal cation channel in brain. Mutant MLC1 proteins that show impaired folding have been corrected <i>in vitro</i> with the addition of a Ca(2+)-ATPase inhibitor, curcumin. Mutations in the gene encoding MLC1 is the cause of megalencephalic leukoencephalopathy with subcortical cysts, also known as van der Knaap disease, a rare syndrome characterised early in life by progressive brain destruction causing mental retardation and incoordination. Single nucleotide polymorphisms within the MLC1 gene may be associated with periodic catatonia, but there seems to be conflicting evidence on whether or not the gene is implicated in general schizophrenia.

Atm mutation leads to the disorder known as ataxia-telangiectasia. The blood disorders Sickle cell anemia and β thalassemia are caused by HBB gene mutations. Wilms' tumors, WAGR syndrome and Denys-Drash syndrome are associated with mutations of the WT1 gene. Jervell and Lange-Nielsen syndrome, Jacobsen syndrome, Niemann-Pick disease, hereditary angioedema and Smith-Lemli-Opitz syndrome are also associated with defects in chromosome 11. Small leucine-rich proteoglycans (SLRPs) such as Decorin, Biglycan, Fibromod-ulin, Keratocan, Lumican and Osteoglycin mediate extracellular matrix organization and are binding partners of TGF Beta. The Decorin core protein binds to growth factors, intercellular matrix molecules such as Fibronectin and Throm-bospondin, and to the Decorin endocytosis receptor. Fibromodulin is a collagen-binding keratan sulphate proteoglycan that influences adhesion processes of connective tissue and plays a role in fibrillogenesis by regulating collagen fibril spacing and thickness. Keratocan (KTN) develops corneal transparency and maintains the stromal matrix structure. Keratocan is a secreted protein in the extracellular matrix that binds to keratan sulfate chains. Keratocan is mainly detected in the cornea, but can also be expressed in trachea, intestine, ovary, lung and skeletal muscle.
