Description:
The X and Y chromosomes are the human sex chromosomes. Chromosome X consists of about 153 million base pairs and nearly 1,000 genes. The combination of an X and Y chromosome lead to normal male development while two copies of X lead to normal female development. There are a number of conditions related to an unsual number and combination of sex chromosomes being inherited. More than one copy of the X chromosome with a Y chromosome causes Klinefelter's syndrome. A single copy of X alone leads to Turner's syndrome. More than 2 copies of the X chromosome, in the absence of a Y chromosome, is known as Triple X syndrome. Color blindness, hemophilia, and Duchenne muscular dystrophy are well known X chromosome-linked conditions which affect males more frequently as males carry a single X chromosome. The FAM104B gene product has been provisionally designated FAM104B pending further characterization.

Description:
The procedure for labeling of DNA probes with a polynucleotide 'tail' containing hapten-labeled nucleotides was developed by Enzo. In such terminal labeling reactions, terminal transferase catalyzes the addition of nucleotides to any 3'-OH terminus in a template independent manner. This rapid and convenient nonradioactive labeling procedure is free of any sequence bias that is normally observed in random priming or nick translation reactions. Terminal labeling is an ideal procedure for the labeling of oligonucleotides.

A wide variety of environmental and pathophysiological stressful conditions trigger the synthesis of a family of proteins known as heat shock proteins (hsp), more appropriately called as stress response proteins (srp). hsp60 is a potential antigen in a number of autoimmune diseases. In human arthritis and in experimentally induced arthritis in animals, disease development coincides with the development of immune reactivity directed against not only bacterial hsp60, but also against its mammalian homolog. Clone LK1, unlike LK2, recognizes only the mammalian (not bacterial) hsp60 and is useful in distinguishing hsp60 from mammals and bacteria.

Mammalian atonal homolog 2 (MATH-2) is a helix-loop-helix (HLH) transcription factor that is structurally homologous to the product of Drosophila atonal gene. MATH-2 is a 337 amino acid protein with an atonal-related basic HLH domain. In mice, expression of MATH-2 takes place by embryonic day 11.5 and initially localizes to the wall of brain vesicles and in the spinal cord. It is expressed in the cortical plate and the mantle layer in the developing central nervous system, and is limited to the nervous system in adults. Adult mouse cerebrums produce a high level of MATH-2 RNA with lower levels in other neuronal tissues. Research studies suggest that MATH-2 may function as a trans-acting factor involved in the development and maintenance of the mammalian nervous system.

The temporal genetic hierarchy influencing normal limb development can deregulate and mediate mammalian developmental syndromes. In mice, the limb deformity (ld) locus influences normal limb development and gives rise to alternative mRNAs that can translate into a family of proteins known as formins. Formins play a crucial role in cytoskeletal reorganization by influencing Actin filament assembly. Formins co-localize with the actin cytoskeleton and can translocate into the cell cytosol and into the nucleus in an HGF-dependent manner. Vertebrate nuclear formins can control polarizing activity in limb buds through establishment of a Sonic hedgehog/FGF-4 feedback loop. Deficiency mutations at the mammalian ld locus lead to profound developmental defects in limb and kidney formation. The human Formin 1 and 2 genes map to chromosome 15q13.3 and 1q43, respectively.

Description:
Calcium-dependent secretion activators (CAPS-1 and CAPS-2) are calcium-binding proteins that direct neurotransmitter and neuropeptide-filled vesicles to the cell membrane for secretory granule exocytosis. Both CAPS-1 and CAPS-2 are expressed primarily in the brain where they regulate the secretion of various substances. The CAPS proteins contain a PH domain that is essential for regulation of exocytosis, as well as regulation of phospholipid binding. Through their regulation of neurotrophin release from granule cells, CAPS proteins help to regulate cell fate during neuronal development. CAPS-1 is thought to regulate catecholamine release from neuronal cells, while CAPS-2 is thought to regulate release of both brain-derived neurotrophic factor and neurotrophin-3 from granule cells. Defects in the genes encoding CAPS-1 and CAPS-2 are implicated in impaired cerebral development and autism.

Additionally, Bcl-6 negatively regulates NFâ…¹B expression, thereby inhibiting NFâ…¹B-mediated cellular functions. Bcl-6b (B-cell CLL/lymphoma 6, member B), also known as ZNF62, BAZF or ZBTB28, is a 480 amino acid nuclear protein that contains one BTB (POZ) domain and five C2H2-type zinc fingers. Expressed ubiquitously with highest expression in placenta and heart, Bcl-6b associates with Bcl-6 and functions as a sequence-specific transcriptional repressor that is thought to be necessary for early B-cell development. The gene encoding Bcl-6b maps to human chromosome 17, which comprises over 2.5% of the human genome and encodes over 1,200 genes.
