Recognizes a protein of 33-55 kDa, identified as CD37 (Workshop V; Code CD37.7). CD37 is strongly expressed on normal and neoplastic mature (sIg ) B-lymphocytes. In B-cell ontogeny, CD37 appears after the pre-B-cell stage, is maintained during peripheral B-cell development and is lost upon terminal differentiation into plasma cells.1 CD37 is also present, at low densities, on resting and activated T cells, neutrophils, monocytes, and some myelomonocytic leukemia cells. It is absent from platelets, erythrocytes. CD37 is a member of a family of tetraspan transmembrane proteins, including CD9, CD53, CD63, CD81, and CD82. It associates other tetraspan transmembrane proteins and MHC class II molecules to form a large complex at the surface of B cells and play a role in signal transduction. CD37 is a valuable and stable marker for peripheral mature B-cells and corresponding malignancies like B-cell chronic lymphocytic leukemia (B-CLL), hairy cell leukemia (HCL), and all types of B-cell non-Hodgkin'' lymphoma (B-NHL).

Human reproduction is controlled by the hypothalaic-pituitary gonadal axis laid down early in fetal development. Gonadotropin releasing hormone (GnRH), also known as GnRH-associated peptide, luteinizing hormone releasing hormone (LHRH), luliberin or gonadorelin, is a decapeptide that is an important molecule in the hypothalamic-pituitary-gonadal axis control circuit. GnRH is produced by hypothalamic neurons and secreted in a pulsatile manner into the capillary plexus of the medianeminence. GnRH affects the release of lutenizing hormone and follicle stimulating hormone from gonadotropic cells in the anterior pituitary. In addition to hypothalamic GnRH (GnRH I), a second GnRH form (GnRH II) functions primarily in the midbrain. GnRH is expressed in the acrosomal region of human sperm and in the anterior pituitary tissue and cancer cells. Unlike GnRH I, GnRH II is highly expressed outside the brain, particularly in the kidney, bone marrow and prostate, suggesting that it may have multiple functions. GnRH binds to a specific G protein-coupled receptor in the pituitary to regulate synthesis and secretion of gonadotropins.

C9orf96, also known as Protein kinase-like protein SgK071, is a 680 amino acid protein that belongs to the Ser/Thr protein kinase family of the protein kinase superfamily. There are three isoforms of C9orf96 that are produced as a result of alternative splicing events. The gene encoding C9orf96 maps to human chromosome 9, which consists of about 145 million bases and 4% of the human genome and encodes nearly 900 genes. Considered to play a role in gender determination, deletion of the distal portion of 9p can lead to development of male to female sex reversal, the phenotype of a female with a male X,Y genotype. Hereditary hemorrhagic telangiectasia, which is characterised by harmful vascular defects, is associated with the chromosome 9 gene encoding endoglin protein, ENG. Familial dysautonomia is also associated with chromosome 9 though through the gene IKBKAP. Notably, chromosome 9 encompasses the largest interferon family gene cluster. Elongation is thought to be a critical stage for the regulation of gene expression. ELL (11-19 lysine-rich leukemia protein), also designated MEN, functions as an RNA polymerase II elongation factor that increases the rate of transcription by suppressing transient pausing by RNA polymerase II. It is also thought to regulate cellular proliferation. ELL is abundantly expressed in peripheral blood leukocytes, skeletal muscle, placenta and testis, with lower expression in spleen, thymus, heart, brain, lung, kidney, liver and ovary. The gene encoding human ELL, which maps to chromosome 19p13.1, is one of several genes that undergo translocation with the MLL gene on chromosome 11q23 in acute myeloid leukemia. The majority of mitochondrial-directed proteins are encoded by the nuclear genome and are transported to the mitochondria via regulated processes involving the mitochondrial Tom and Tim proteins. The mitochondrial Tim protein family is comprised of a large group of evolutionarily conserved proteins that are found in most eukaryotes. Import of nuclear-encoded precursor proteins into and across the mitochondrial inner membrane is mediated by two distinct complexes, the Tim23 complex and the Tim22 complex, which differ in their substrate specificity. Defects in Tim proteins are implicated in several neuro-degenerative diseases, suggesting important roles for Tim proteins in development and health. Tim8A and Tim8B, which map to human chromosomes Xq22.1 and 11q23.1-q23.2, respectively, are conserved proteins of the mitochondrial intermembrane space, which are organized in hetero-oligomeric complex with Tim13. Tim8A is highly expressed in fetal and adult brain. Smad2 is a 58 kDa member of a family of proteins involved in cell proliferation, differentiation and development. The Smad family is divided into three subclasses: receptor-regulated Smad's, activin/TGF alpha receptor-regulated (Smad2 and 3) or BMP receptor regulated (Smad1, 5, and 8); the common partner, (Smad4) that functions via its interaction to the various Smad's; and the inhibitory Smad's, (Smad6 and Smad7). Smad2 consists of two highly conserved domains, the N terminal Mad homology (MH1) and the C-terminal Mad homology 2 (MH2) domains. The MH1 domain binds DNA and regulates nuclear import and transcription while the MH2 domain conserved among all the Smad's regulates Smad2 oligomerization and binding to cytoplasmic adaptors and transcription factors. Activated Smad2 associates with Smad4 and translocates as a complex into the nucleus, allowing its binding to DNA and transcription factors. This translocation of Smad2 (as well as Smad3) into the nucleus is a central event in TGF beta signaling. Phosphorylation of threonine 8 in the calmodulin binding region of the MH1 domain by extracellular signal regulated kinase 1(ERK 1) enhances Smad2 transcriptional activity, which is negatively regulated by calmodulin. The regulation of Smad2 phosphorylation on threonine 8 by ERK 1 and calmodulin is critical for Smad2 mediated signaling.
