AADACL4 is a 407 amino acid single-pass type II membrane protein belonging to the 'GDXG' lipolytic enzyme family. Integral to the cell membrane, AADACL4 participates in carboxylesterase and hydrolase activities and is encoded by a gene that maps to human chromosome 1p36.22. Chromosome 1, the largest human chromosome, makes up 8% of the human genome and contains about 260 million base pairs, which encode 3,000 genes. Chromosome 1 houses a large number of disease-associated genes, including those that are involved in familial adenomatous polyposis, Stickler syndrome, Parkinson’s disease, Gaucher disease, schizophrenia and Usher syndrome. Aberrations in chromosome 1 are found in a variety of cancers, including head and neck cancer, malignant melanoma and multiple myeloma.

Description:
Shootin1 is a 631 amino acid protein that belongs to the shootin family. The shootin1 protein contains three coiled-coil domains, a proline-rich region and interacts with RUFY3. Shootin1 is involved in the generation of internal asymmetric signals required for neuronal polarization. The shootin1 protein acts upstream of PI3K (phosphoinositide 3-kinase), by being required for spatially localized PI3K activity. By accumulating asymmetrically in a single neurite before polarization, shootin1 leads to axon induction for polarization, additionally the absence of shootin1 from the nascent axon's siblings by competition prevents the formation of surplus axons. Existing as seven alternatively spliced isoforms, the shootin1 gene is conserved in chimpanzee, dog, mouse, rat, chicken and zebrafish, and maps to human chromosome 10q25.3.

ADCK2 (aarF domain containing kinase 2), also known as AARF, is a 626 amino acid single-pass membrane protein belonging to the protein kinase superfamily and the ADCK protein kinase family. The ADCK family consists of five paralogs in human (ADCK1-5). Encoded by a gene that maps to human chromosome 7q34, ADCK2 contains one protein kinase domain. ADCK2 participates in ATP and nucleotide binding, transferase functions and protein serine/threonine kinase activities. Expression of ADCK2 inversely correlates with cellular viability, suggesting elevated expression of ADCK2 may be essential for tumour survival. ADCK2 is necessary for cell proliferation of glioblastoma multiforme (GBM), a fatal primary brain tumor containing countless genetic and epigenetic alterations.

Involved in a variety of functions, protocadherins help to regulate neural development and synapse formation. PCDH1 is a 1,026 amino acid single-pass type I membrane protein that contains seven cadherin domains and is a member of the protocadherin family. Localized to cell-cell and cell-matrix boundaries and expressed at high levels in brain and neuro-glial cells, PCDH1 is thought to be involved in cell adhesion and cell-cell interactions and may play a role in neuronal development. PCDH1 contains a C-terminal cytoplasmic region, an extracellular region and a transmembrane region, and is expressed as two isoforms due to alternative splicing events.

Expressed as two alternatively spliced isoforms, PCDHGC4 functions as a potential calcium-dependent cell adhesion protein that is thought to be involved in the establishment and maintenance of neuronal connections within the brain. Syntaxins were originally thought to be docking proteins, but have more recently been categorized as anchoring proteins that anchor themselves to the cytoplasmic surfaces of cellular membranes. Syntaxins bind to various proteins involved in exocytosis, including VAMPs (vesicle-associated membrane proteins), NSF (N-ethylmaleimide-sensitive factor), SNAPs (soluble NSF attachment proteins) and Synaptotagmin. Endobrevin, also designated VAMP-8 or ED, is a 100 amino acid single-pass type IV membrane protein that belongs to the synaptobrevin family. Similar in sequence to the synaptobrevins, endobrevin is abundantly expressed in kidney, moderately expressed in heart and spleen, and slightly expressed in brain, thymus and liver. Endobrevin interacts specifically with the SNAPs, most likely through an endobrevin-containing SNARE complex.
