Akirin2 Antibody: The highly conserved, nuclear-localized Akirin1 and Akirin2 proteins critically regulate the transcription of NF-kappa B-dependent genes and are required for defense against Gram-negative bacteria in the immune deficiency and NF-kappa B pathways. Akirin1 is dispensable in the mouse, and neither knockout mice nor cells derived from them have obvious distinctive phenotypes. In contrast, Akirin2 is required for development in the mouse and knockout of both Akirin homologs in mice show that Akirin2 is required downstream of toll-like receptor (TLR), TNF-alpha and IL-1 beta signaling, and for the production of IL-6. Akirin2 is functionally closer to the single gene in Drosophila, as the homozygous null D. melanogaster Akirin mutants show a similar, mid-to-early embryonic death.

Involved in DNA non-homologous end joining (NHEJ) required for double-strand break (DSB) repair and V(D)J recombination. Must be bound to DNA to express its catalytic properties. Promotes processing of hairpin DNA structures in V(D)J recombination by activation of the hairpin endonuclease artemis (DCLRE1C). The assembly of the DNA-PK complex at DNA ends is also required for the NHEJ ligation step. Required to protect and align broken ends of DNA. May also act as a scaffold protein to aid the localization of DNA repair proteins to the site of damage. Found at the ends of chromosomes, suggesting a further role in the maintenance of telomeric stability and the prevention of chromosomal end fusion. Also involved in modulation of transcription. Recognizes the substrate consensus sequence [ST]-Q. Phosphorylates 'Ser-139' of histone variant H2AX/H2AFX, thereby regulating DNA damage response mechanism. Phosphorylates DCLRE1C, c-Abl/ABL1, histone H1, HSPCA, c-jun/JUN, p53/TP53, PARP1, POU2F1, DHX9, SRF, XRCC1, XRCC1, XRCC4, XRCC5, XRCC6, WRN, MYC and RFA2. Can phosphorylate C1D not only in the presence of linear DNA but also in the presence of supercoiled DNA. Ability to phosphorylate p53/TP53 in the presence of supercoiled DNA is dependent on C1D. Contributes to the determination of the circadian period length by antagonizing phosphorylation of CRY1 'Ser-588' and increasing CRY1 protein stability, most likely through an indirect machanism. Interacts with CRY1 and CRY2; negatively regulates CRY1 phosphorylation.

R-spondin-1 is also known as Roof plate-specific Spondin 1 (RSPO1) and cysteinerich and single thrombospondin domain containing protein 3 (Cristin 3), which is a secreted protein which belongs to the R-Spondin family and encodes a secreted activator protein with two cystein-rich, furin-like domains and one thrombospondin type 1 domain. All Rspondins regulate Wnt/β-catenin signaling, but have distinct expression patterns. Like other R-Spondins, R-Spondin-1 contains two adjacent cysteinerich furinlike domains (aa 34-135) with one potential N-glycosylation site, followed by a thrombospondin (TSP1) motif (aa 147-207) and a region rich in basic residues (aa 211-263). Only the furinlike domains are needed for β-catenin stabilisation. A putative nuclear localisation signal at the C-terminus may allow some expression in the nucleus.

Description:
The RPA-T4 monoclonal antibody specifically binds to the CD4 receptor for the human immunodeficiency virus (HIV). CD4 is a 59 kDa single-chain transmembrane glycoprotein that expressed on the surface of most of the thymocytes, T-helper cells, and in low levels on monocytes and macrophages. CD4 is a co-receptor in the antigen-induced T cell activation, together with the MHC class II. The RPA-T4 antibody is capable of blocking HIV binding and inhibiting syncytium formation, by binding to the D1 domain of the CD4 antigen. The OKT4 and the RPA-T4 monoclonal antibodies recognize different epitopes of CD4 and they do not exhibit cross-block binding.

This gene encodes a member of the UDP-N-acetyl-alpha-D-galactosamine:polypeptide N-acetylgalactosaminyltransferase (GalNAc-T) family of enzymes. GalNAc-Ts initiate mucin-type O-linked glycosylation in the Golgi apparatus by catalyzing the transfer of GalNAc to serine and threonine residues on target proteins. They are characterized by an N-terminal transmembrane domain, a stem region, a lumenal catalytic domain containing a GT1 motif and Gal/GalNAc transferase motif, and a C-terminal ricin/lectin-like domain. GalNAc-Ts have different, but overlapping, substrate specificities and patterns of expression. In vitro, the encoded protein can complement other GalNAc-Ts in the complete O-glycosylation of the mucin-1 tandem repeat and can O-glycosylate the P-selectin glycoprotein ligand-1 molecule. The coding region of this gene is contained within a single exon.

RPA Interacting Protein Antibody: Replication protein A (RPA) is a single-stranded-DNA binding protein involved in numerous eukaryotic DNA processes including replication, repair and recombination. RPA interacting protein (RPA IP) has been identified as an adapter protein that is involved in RPA nuclear import instead of the prototypical importin proteins that normally mediate nuclear import. Multiple isoforms of RPA IP are known to exist, with the longest isoform localized to the cytoplasm. Isoform 2 is sumoylated and is located in the PML nuclear body within the nucleus. It has been suggested that this isoform mediates the localization of the RPA complex into the PML nuclear body, thereby participating in RPA function in DNA metabolism.

PIWI-L1 Antibody: PIWI-L1 is a member of the PIWI subfamily of Argonaute proteins, evolutionarily conserved proteins containing both PAZ and Piwi motifs that are crucial for the biogenesis and function of small non-coding RNAs and play important roles in stem cell self-renewal, RNA silencing, and translational regulation in diverse organisms. PIWI-L1 is thought to play a role as an intrinsic regulator of the self-renewal capacity of germline and hematopoietic stem cells as removing PIWI-L1 protein from single germline stem cells significantly decreases the rate of their division. Recent studies have shown that overexpression of PIWI-L1 (as well as other PIWI-like proteins) are potential biomarkers for colon and other cancers.

EGLN3 catalyzes the post-translational formation of 4-hydroxyproline in hypoxia-inducible factor (HIF) alpha proteins. It hydroxylates HIF-1 alpha at 'Pro-564', and HIF-2 alpha. EGLN3 functions as a cellular oxygen sensor and, under normoxic conditions, targets HIF through the hydroxylation for proteasomal degradation via the von Hippel-Lindau ubiquitination complex. It may play a role in cell growth regulation in muscle cells and in apoptosis in neuronal tissue. EGLN3 promotes cell death through a caspase-dependent mechanism.
