Description:
High density lipoproteins (HDLs) play a critical role in cholesterol metabolism and their plasma concentrations are inversely correlated with risk for atherosclerosis. SR-BI and SR-BII (previously known as SR-BI.2) are the alternatively spliced products of a single gene. SR-BII and SR-BI are identical except for the encoded c-terminal cytoplasmic domain. Both SR-BI and SR-BII bind HDL and mediates selective uptake of HDL cholesteryl ester, but with SR-BII having an approximately 4-fold lower efficiency than SR-BI. SR-BI and SR-BII are expressed primarily in liver and non-placental steroidgenic tissues. Although the role of these scavenger receptors is not completely clear, SR-BII mRNA results from the alternative splicing of SR-BI precursor transcripts with both isoforms mediating selective transfer of lipid between HDL and cells. Therefore, the relative expression and functional activities of these two isoforms create a potential means of regulating selective lipid transfer between HDL and cells.

The cadherins represent a family of Ca2+-dependent adhesion molecules that function to mediate cell to cell binding that is critical for the maintenance of structure and morphogenesis. Cadherins each contain a large extracellular domain at the N-terminus, which is characterized by a series of five homologous repeats, the most distal of which is thought to be responsible for binding specificity. Containing thirty-three cadherin domains, four EGF-like domains and one laminin G-like domain, FAT3 may participate in the interactions between neurites derived from specific subsets of neurons during development.

Orai2 is a 254 amino acid multi-pass membrane protein that belongs to the orai family of proteins. Localizing to the plasma membrane, Orai2 plays an important role in store-operated calcium (SOC) entry, a process involving Ca2+ influx and replenishment of Ca2+ stores formerly emptied through the action of inositol 1,4,5-trisphosphate production and other Ca2+ mobilizing agents. CRAC channels are responsible for medi-ating calcium influx in T-cells and play an important role in the immune response. Orai2 specifically increases the Ca2+-selective current through coaction with the Ca2+ sensor Stim1.

The protein encoded by this gene belongs to the ser/thr protein kinase family. It is the catalytic subunit of the 5'-prime-AMP-activated protein kinase (AMPK). AMPK is a cellular energy sensor conserved in all eukaryotic cells. The kinase activity of AMPK is activated by the stimuli that increase the cellular AMP/ATP ratio. AMPK regulates the activities of a number of key metabolic enzymes through phosphorylation. It protects cells from stresses that cause ATP depletion by switching off ATP-consuming biosynthetic pathways. Alternatively spliced transcript variants encoding distinct isoforms have been observed. Synaptotagmins are a large family of synaptic vesicle type III integral membrane proteins that function as regulators of both exocytosis and endocytosis and are involved in neurotransmitter secretion from small secretory vesicles. Synaptotagmin XI, also known as SYT11 (Synaptotagmin-11), is a 431 amino acid protein that localizes to the membrane and is expressed ubiquitously with highest expression in brain and lung. Like other Synaptotagmin proteins, Synaptotagmin XI is involved in the calcium-dependent exocytosis of secretory vesicles and is thought to act as a calcium sensor during vesicular trafficking. Synaptotagmin XI contains two C2 domains through which it can bind either three calcium ions or the zinc-finger protein Parkin (a juvenile Parkinson's disease gene product), the latter of which causes the polyubiquitination and subsequent degradation of Synaptotagmin XI by the proteasome complex. Defects in the gene encoding Synaptotagmin XI are implicated in a number of neurological disorders, including schizophrenia and Parkinson's disease.
