Description:
This gene encodes a nuclear phosphoprotein that plays a role in maintaining genomic stability, and it also acts as a tumor suppressor. The encoded protein combines with other tumor suppressors, DNA damage sensors, and signal transducers to form a large multi-subunit protein complex known as the BRCA1-associated genome surveillance complex (BASC). This gene product associates with RNA polymerase II, and through the C-terminal domain, also interacts with histone deacetylase complexes. This protein thus plays a role in transcription, DNA repair of double-stranded breaks, and recombination. Mutations in this gene are responsible for approximately 40% of inherited breast cancers and more than 80% of inherited breast and ovarian cancers. Alternative splicing plays a role in modulating the subcellular localization and physiological function of this gene. Many alternatively spliced transcript variants, some of which are disease-associated mutations, have been described for this gene, but the full-length natures of only some of these variants has been described. A related pseudogene, which is also located on chromosome 17, has been identified. [provided by RefSeq, May 2009].

This gene is one of several genes encoding pulmonary-surfactant associated proteins (SFTPA) located on chromosome 10. Mutations in this gene and a highly similar gene located nearby, which affect the highly conserved carbohydrate recognition domain, are associated with idiopathic pulmonary fibrosis. The current version of the assembly displays only a single centromeric SFTPA gene pair rather than the two gene pairs shown in the previous assembly which were thought to have resulted from a duplication.

Reacts with CD95 (Fas/APO-1), a 46 kDa single chain type I glycoprotein of the tumour necrosis factor/nerve growth factor (TNF/NGF) receptor superfamily, expressed on a variety of normal and neoplastic cells. It seems that the antibody LT95 does not induce Fas mediated apoptosis, although it cross-blocks anti-Fas DX2 antibody that recognizes a functional epitope of Fas molecule.

CD300C is a single-pass type I membrane protein which belongs to the immunoregulatory signaling (IRS) family. CD300C contains one Ig-like V-type domain and is present on the surface of natural killer cells, granulocytes, most myeloid cells, dendritic cells, and a subpopulation of T and B lymphocytes. The CD300C (CMRF-35A) and CD300A (CMRF-35H) molecules are homologous leukocyte surface proteins. CD300a and CD300C play an important role in the cross-regulation of TNF-alpha and IFN-alpha secretion from pDCs. CD300A and CD300C are indistinguishable on the surface of NK cells. The ligand for CD300C is presently unknown.

TGFBR2 is a single-pass type I membrane protein and contains one protein kinase domain. TGFBR2 exsits as a heterodimeric complex with another receptor protein and binds TGF-beta. Signals triggered through the TGF-beta receptor complex prompt various responses by the cell. One such response is to inhibit cell growth and division. Based on this action, the TGF-beta receptor type 2 is sometimes called a tumor suppressor. Defects in TGFBR2 have been associated with Marfan syndrome, Loeys-Deitz aortic aneurysm syndrome, Osler-Weber-Rendu syndrome and the development of various types of tumors.

Poliovirus Receptor (PVR) is a 70 kDa type I transmembrane single-span glycoprotein that belongs to the nectin-like (Necl) family and was originally identified based on its ability to mediate the cell attachment and entry of poliovirus (PV), an etiologic agent of the central nervous system disease poliomyelitis. PVR contains three Ig-like extracellular domains, a transmembrane segment, and a cytoplasmic tail. The normal cellular function of PVR maybe the involvement of intercellular adhension between epithelial cells. Alternate splicing of the PVR mRNA yields four different isoforms ( alpha, beta , gamma , and delta) with identical extracellular domains.
