Sphingomyelin synthases synthesize the sphingolipid, sphingomyelin, through transfer of the phosphatidyl head group, phosphatidylcholine, on to the primary hydroxyl of ceramide. The reaction is bidirectional depending on the respective levels of the sphingolipid and ceramide. Golgi apparatus SMS1 directly and specifically recognizes the choline head group on the substrate, requiring two fatty chains on the choline-P donor molecule in order to be recognized efficiently as a substrate. Major form in macrophages. Required for cell growth in certain cell types such as HeLa cells. Suppresses BAX-mediated apoptosis and also prevents cell death in response to stimuli such as hydrogen peroxide, osmotic stress, elevated temperature and exogenously supplied sphingolipids. May protect against cell death by reversing the stress-inducible increase in levels of proapoptotic ceramide.

S100 calcium-binding protein P (S100P),a member of the S-100 family, is also known as protein S100-P, migration-inducing gene 9 protein (MIG9) and protein S100-E,which contains two EF-hand domains. As for subunit structure,S100P is homodimer and heterodimer with S100A1. S100P may function as calcium sensor and contribute to cellular calcium signaling. EFCAB4B Antibody: EFCAB4B, also known as Calcium release-activated calcium channel regulator 2A, is a novel Ca2+-binding EF-hand protein that is thought to play a key role in store-operated Ca2+ entry in T-cells by regulating CRAC channel activation. EFCAB4B acts as a cytoplasmic calcium-sensor that forms a complex with ORAI1 and STIM1 at the junctional regions between the plasma membrane and the endoplasmic reticulum upon low Ca2+ concentration. A closely related protein, EFCAB4A, is likely to play a similar role as EFCAB4B, but the detailed function of EFCAB4A is still under investigation.

ORAI1 Antibody: Antigen stimulation of immune cells triggers Ca++ entry through Ca++ release-activated Ca++ (CRAC) channels. ORAI1 is a recently identified four-transmembrane spanning protein that is an essential component of CRAC. A missense mutation in this protein in humans is the cause of one form of hereditary severe combined immune deficiency (SCID) which results in ablated T-cell Ca++ entry. It has been suggested that ORAI1 functions as a highly selective Ca++ plasma membrane channel that is gated through interactions with STIM1, the store-activated endoplasmic reticulum Ca++ sensor.

The One Step RT-PCR Master Mix Kit allows rapid, sensitive analysis of gene expression from tissues and cells. It can replace methods for detecting and quantifying gene expression such as Northern blots,<i> in situ</i> hybridisation, dot blots, S nuclease assays and conventional two step RT-PCR. The kit utilises recombinant <i>Thermus thermophilus</i> (rTth) DNA Polymerase, which acts as both a thermostable RNA dependent DNA polymerase and a DNA dependent DNA polymerase. The rTth DNA Polymerase is provided in a 2X master mix with an antibody for antibody-mediated hot start, optimised buffer, and ultrapure deoxynucleotides. Antibody-mediated Hot Start enhances specificity of both reverse transcription and PCR. The kit enables cDNA synthesis from input RNA followed by PCR amplification of the cDNA in a single reaction, with no additional hands-on requirement for buffer changes or adding reagents. Typically, detection of a specific transcript requires only 2 hours.
