Description:
Tissue inhibitor of metalloproteinases 1 (TIMP1) and Tissue inhibitor of metalloproteinases 2 (TIMP 2) have similar properties, specifically in inhibiting enzymes of the matrix metalloproteinase family, and are thought to be of great importance in the maintenance of connective tissue integrity. TIMP1 forms a complex of 1:1 stoichiometry with activated interstitial collagenases, activated stromelysin, active form of 72kDa Type IV collagenase (also known as MMP2 or gelatinase A), and latent and active forms of 92kDa Type IV collagenase (also known as MMP0 or gelatinase B). TIMPs inhibit the proteolytic invasiveness of tumour cells and normal placental trophoblast cells. TIMP1 is produced in low (pg/mL) levels by most cell types. Treatment of cells with the phorbol ester TPA stimulates production of TIMP1 in some cell types, but the low protein levels produced often require concentration of cell culture media to visualize the bands by Western Blotting.

Betacellulin is a member of the epidermal growth factor (EGF) family, and signals through EGF receptor and ERBB4. It activates ERK and AKT pathways, which induces neural stem cell proliferation and prevents spontaneous differentiation in culture. Betacellulin stimulates the expansion of neural stem cells, transit-amplifying cells, and neuroblasts derived from subventricular zone and dentate gyrus (Gómez-Gaviro <i>et al.</i>). It is a potent mitogen for retinal pigment epithelial cells and vascular smooth muscle cells. Betacellulin down-regulates E-cadherin expression in ovarian cancer cell lines via MEK/ERK1/2 and PI3K/AKT signaling pathways, thus increasing cell migration (Zhao <i>et al.</i>). It is a modulator of interferon (IFN) response and enhances anti-viral effects of IFN (Al-Yahya <i>et al.</i>). Betacellulin is expressed in pancreatic α cells, β cells, and duct cells. It induces the proliferation of pancreatic cancer cell lines, inhibits apoptosis, promotes the neogenesis of β cells, and converts non-β cells into insulin-producing cells (Kawaguchi <i>et al.</i>; Miyagawa al.; Saito <i>et al.</i>).

PRKCDBP Antibody: The protein kinase C delta (PKC-δ) binding protein (PRKCDBP), also known as cavin-3, is a member of the cavin family of proteins that are involved in caveolin formation and regulation. PRKCDBP was initially identified in a screen of cultured cell lines for proteins that were strongly induced by serum starvation. Studies indicate that PRKCDBP binds not only to PKC-δ but also to caveolin-1 and helps regulate caveolin traffic and function. Similar to other members of the cavin family, the expression of PRKCDBP was found to be down-regulated in various cancer cell lines, suggesting a possible tumor suppressor function of PRKCDBP.

Ambra1 Antibody: Autophagy, the process of bulk degradation of cellular proteins through an autophagosomic-lysosomal pathway is important for normal growth control and may be defective in tumor cells. It is involved in the preservation of cellular nutrients under starvation conditions as well as the normal turnover of cytosolic components. Beclin-1, a principal regulator of autophagosome formation, is in turn regulated by Ambra1. Ambra1 associates with Beclin-1 through a region near its center as determined by yeast two-hybrid assay. Null mutations in this gene in mice resulted in embryonic lethality with severe neural tube defects associated with autophagy impairment, accumulation of ubiquitinated proteins, unbalanced cell proliferation and excessive apoptotic death. Furthermore, down-regulation of Ambra1 in cultured cells though RNA interference decreased the level of rapamycin- and nutrient starvation-induced autophagy. Multiple isoforms of Ambra1 are known to exist.

Granulocyte-macrophage colony-stimulating factor (GM-CSF) promotes the proliferation and differentiation of hematopoietic progenitor cells and the generation of neutrophils, eosinophils, and macrophages. In synergy with other cytokines such as stem cell factor, IL-3, erythropoietin, and thrombopoietin, it also stimulates erythroid and megakaryocyte progenitor cells (Barreda <i>et al.</i>). GM-CSF was first purified from the culture of mouse lung tissue after lipopolysaccharide treatment. GM-CSF is produced by multiple cell types, including stromal cells, Paneth cells, macrophages, dendritic cells (DCs), endothelial cells, smooth muscle cells, fibroblasts, chondrocytes, and Th1 and Th17 cells (Francisco-Cruz <i>et al.</i>). The receptor for GM-CSF (GM-CSFR) is composed of two subunits: the cytokine-specific α subunit (GMRα; CD116) and the common subunit βc (CD131) shared with IL-3 and IL-5 receptors (Broughton <i>et al.</i>). GM-CSFR is expressed on hematopoietic cells, including progenitor cells and immune cells, as well as non-hematopoietic cells. GM-CSF is able to stimulate the development of DCs that ingest, process, and present antigens to the immune system (Francisco-Cruz <i>et al.</i>).

Nk1.1 is an antigen encoded by the Klrb1c/NKR-P1C gene expressed by the natural killer cells of some selected strains of mice (C57BL, FVB/N, NZB) and encoded by the Klrb1b/NKR-P1B gene expressed on Swiss NIH and SJL mice. Pk136 binds to an epitope common to NKR-P1B and NKR-P1C.The Klrb1 is a family of type II transmembrane C-type lectin receptors. Klrb1c activates the NK-cell cytotoxicity, while Klrb1b inhibits it. PK136 is useful in defining the NK cells and the rare population of NK-T lymphocytes and specific cultured monocytes.

NALP1 Antibody: NALP1 belongs to a family of cytoplasmic proteins that have been implicated in cell responses to apoptotic and inflammatory stimuli. It contains a caspase recruitment domain (CARD) in addition to a NACHT domain and leucine rich repeat (LRR). This protein interacts strongly with caspase 2 and weakly with caspase 9, as well as with Apaf-1. Transient overexpression of this gene in cultured cells was sufficient to induce apoptosis. NALP1 mRNA and protein expression levels in myeloid leukemia cells are increased following CREB (cAMP-response element binding protein) activation, suggesting that NALP1 may contribute to modulate the response of these cells to pro-inflammatory stimuli. Multiple alternatively spliced transcript variants encoding distinct isoforms have been found for NALP1.

Neurofilaments are the 10nm or intermediate filament proteins found specifically in neurons, and are composed predominantly of three major proteins called NF-L, NF-M and NF-H. NF-M is the neurofilament middle or medium molecular weight polypeptide and runs on SDS-PAGE gels at 145-160 kDa, with some variability across species boundaries. Antibodies to NF-M are useful for identifying neuronal cells and their processes in tissue sections and in tissue culture. NF-M antibodies can also be useful to visualize neurofilament accumulations seen in many neurological diseases, such as Amyotrophic Lateral Sclerosis (Lou Gehrig's disease) and Alzheimer's disease.
