D-myo-Inositol is a component of membrane phospholipids, glycosylphophatidylinositol anchors that bind glycoproteins to cell membranes, and inositol phosphate second messengers. It is a growth factor for animals as well as microorganisms. Myo-Inositol is the most abundant form of polyols that serves as a structural element of secondary messengers in eukaryotic cells. Inositol or cyclohexane-1,2,3,4,5,6-hexol is a chemical compound, a sixfold alcohol (polyol) of cyclohexane. It exists in nine possible stereoisomers, of which the most prominent form, widely occurring in nature, is cis-1,2,3,5-trans-4,6-cyclohexanehexol, or myo-inositol (former name meso-inositol). Inositol is a carbohydrate, though not a classical sugar. It has a taste which has been assayed at half the sweetness of table sugar. Used as a lipotropic agent. A component of membrane phospholipids, glycosylphosphatidylinositol anchors that bind glycoproteins to cell membranes, and inositol phosphate second messengers. In addition, inositol serves as an important component of the structural lipids phosphatidylinositol (PI) and its various phosphates, the phosphatidylinositol phosphate (PIP) lipids.

ASAH1 Antibody: Sphingolipids are hydrolyzed by ceramidases to yield sphingosine and fatty acids. These ceramidases are classified according to the pH range that supports their optimal activity. ASAH1 is an acid ceramidase and key regulator of ceramide metabolism. Mutations in this gene results in Farber Lipogranulomatosis, a fatal human genetic disorder that results in the painful swelling of the joints and tendons and pulminary insufficiency, while a complete knockout of its expression is lethal in mice. Recent studies have shown elevated levels of ASAH1 in Alzheimer's disease (AD) patients correlating with a reduction in sphingomyelin and elevation of ceramide. Pretreatment of cultured neurons with recombinant AHAH1 prevented the cells from undergoing A-beta (Ab)-induced apoptosis. Multiple isoforms of this protein are known to exist.

Glutathione is the major low molecular weight thiol compound of the living plant or animal cell. It is a tripeptide with a gamma peptide linkage between the amine group of cysteine (which is attached by normal peptide linkage to a glycine) and the carboxyl group of the glutamate side-chain. It is an antioxidant, preventing damage to important cellular components caused by reactive oxygen species such as free radicals and peroxides. The sulfhydryl (thiol) group (SH) of cysteine serves as a proton donor and is responsible for the biological activity of glutathione. Glutathione suppresses human immunodeficiency virus expression in chronically infected monocytic cells. It is a useful tripeptide involved in many aspects of metabolism, including transport of g-glutanyl amino acids and reductive cleavage of disulfide bonds. Endogenous antioxidant that plays a major role in reducing reactive oxygen species formed during cellular metabolism and the respiratory burst. Glutathione-S-transferase catalyzes the formation of glutathione thioethers with xenobiotics, leukotrienes, and other molecules that have an electrophilic center. Glutathione also forms disulfide bonds with cysteine residues in proteins. Via these mechanisms, it can have the paradoxical effect of reducing the efficacy of anti-cancer agents.

Iterleukin 7 is a lymphoid cell growth factor that affects pre-B, pro-B, and early T cells. IL-7 was previously known as pre-B cell growth factor and lymphopoietin 1. IL-7 supports the growth of early B cells from long-term lymphoid bone marrow cultures. It is mitogenic to thymocytes and enhances the response of cells to other stimuli such as polyhydroxyalkanoate (PHA) and concanavalin A (ConA). IL-7 stimulates the proliferation of CD4+/CD8+ cells. The proliferative response of thymocytes to IL-7 is not affected by antibodies to the T cell growth factors such as IL-2, IL-4 and IL-6, suggesting that IL-7 is capable of stimulating T cell proliferation through a pathway independent of the known T cell growth factors. Mature T cells respond to IL-7 and Con A, but not to IL-7 alone. In myeloid lineage cells, IL-7 upregulates the production of pro-inflammatory cytokines and stimulates the tumoricidal activity of monocytes/macrophages. IL-7 is expressed by adherent stromal cells from various tissues.

Description:
Tumor Necrosis Factor Alpha (TNF alpha) is a protein secreted by lipopolysaccharide-stimulated macrophages, and causes tumor necrosis when injected into tumor bearing mice. TNF alpha is believed to mediate pathogenic shock and tissue injury associated with endotoxemia. TNF alpha exists as a multimer of two, three, or five non-covalently linked units, but shows a single 17 kDa band following SDS PAGE under non-reducing conditions. TNF alpha is closely related to the 25 kDa protein Tumor Necrosis Factor beta (lymphotoxin), sharing the same receptors and cellular actions. TNF alpha causes cytolysis of certain transformed cells, being synergistic with interferon gamma in its cytotoxicity. Although it has little effect on many cultured normal human cells, TNF alpha appears to be directly toxic to vascular endothelial cells. Other actions of TNF alpha include stimulating growth of human fibroblasts and other cell lines, activating polymorphonuclear neutrophils and osteoclasts, and induction of interleukin 1, prostaglandin E2 and collagenase production. TNF alpha is currently being evaluated in treatment of certain cancers and AIDS Related Complex.

Noggin is also known as NOG, SYM1, SYNS1 and is a secreted homodimeric glycoprotein whose scaffold contains a cystine-knot topology similar to that of BMPs.Secreted Noggin probably remains close to the cell surface due to its binding of heparincontaining proteoglycans.Noggin inhibits TGF-β signal transduction by binding to TGF-β family ligands and preventing them from binding to their corresponding receptors. Noggin plays a key role in neural induction by inhibiting BMP4, along with other TGF-β signaling inhibitors such as chordin and follistatin. Mouse knockout experiments have demonstrated that noggin also plays a crucial role in bone development, joint formation, and neural tube fusion. During embryogenesis, Noggin antagonizes specific BMPs at defined times, for example, during neural tube, somite and cardiomyocyte growth and patterning. During skeletal development, Noggin prevents chondrocyte hyperplasia, thus allowing proper formation of joints. During culture of human embryonic stem cells (hESC) or neural stem cells under certain conditions, addition of Noggin to antagonize BMP activity may allow stem cells to proliferate while maintaining their undifferentiated state, or alternatively, to differentiate into dopaminergic neurons Noggin also appears to maintain adult stem cell populations in vivo, for example, maintaining neural stem cells within the hippocampus.

HIG2 Antibody: HIG1 and HIG2 (Hypoxia-inducible gene 1 and 2, respectively) are known to be induced by hypoxic conditions. HIG2 is induced by hypoxia and by glucose deprivation in cultured cells. In addition, tumor xenografts derived from human cervical cancer cells display increased expression of HIG1 and HIG2 when they are deprived of oxygen. Unlike HIG2, which is ubiquitously expressed and might be an activator and target of the canonical Wnt pathway, the function and the mechanisms underlying its regulation of HIG1 still remained unknown. The putative link between hypoxia and an oncogenic signaling pathway might play an important role in tumorigenesis.
