Description:
Interferon (IFN) Inducible 16 (IFI16) protein belongs to a family of HIN 200 human and mouse proteins. IFI16 is a nuclear protein comtaining regulatory domains such as DNA binding domain, transcriptional regulatory domain and DAPIN/PAAD domain. IFI16 has three isotypes A, B, and C (85-95 kDa), which arise as a result of mRNA alternative splicing. All are phosphorylated on serine and threonine residues and can homo and heterodimerize. Expression is restricted to the nuclei of hematopoietic cells, fibroblasts and epithelial cells. IFI16 expression in hematopoietic cells of myeloid lineage is tightly regulated and highly induced in the differentiation and proliferation of the cell. Due to its localisation in the nucleus, regulation of protein expression, and ability to bind DNA, it is assumed that IFI16 has a role in transcription regulation of cell differentiation. In addition, it was found that IFI16 can act as a transcriptional repressor and is involved in regulation and activation of p53 in cancer cells.

Proliferation Marker Ki67 antigen is the prototypic cell cycle related nuclear protein, expressed by proliferating cells in all phases of the active cell cycle (G1, S, G2 and M phase). It is absent in resting (G0) cells. Ki67 are useful in establishing the cell growing fraction in neoplasms (immunohistochemically quantified by determining the number of Ki67 positive cells among the total number of resting cells = Ki67 index). In neoplastic tissues the prognostic value is comparable to the tritiated thymidine labelling index. The correlation between low Ki67 index and histologically low grade tumours is strong. Ki67 is routinely used as a neuronal marker of cell cycling and proliferation.

Recognizes a 40-43 kDa transmembrane epithelial glycoprotein, identified as epithelial specific antigen (ESA), or epithelial cellular adhesion molecule (Ep-CAM). Ep-CAM is expressed on baso-lateral cell surface in most simple epithelia and a vast majority of carcinomas. This antibody has been used to distinguish adenocarcinoma from pleural mesothelioma and hepatocellular carcinoma. It is also useful in distinguishing serous carcinomas of the ovary from mesothelioma. This epithelial antigen plays an important role as a tumor-cell marker in lymph nodes from patients with esophageal carcinoma otherwise classified as node-negative. Epithelial antigen has also been suggested as a discriminator between basal cell and baso-squamous carcinomas, and squamous cell carcinoma of the skin.

Recognizes a protein of 80 kDa-90 kDa, identified as CD36. It is expressed on platelets, monocytes and macrophages, microvascular endothelial cells, erythrocyte precursors, mammary epithelial cells, and some macrophage derived dendritic cells. CD36 acts as a receptor for thrombospondin (TSP), collagen types I, IV and V, P. falciparum malaria-infected erythrocytes, and sickle erythrocytes. It also functions as a scavenger receptor, mediating macrophage uptake of oxidized low-density lipoprotein (LDL) and recognition of apoptotic polymorphonuclear leukocytes (PMN). CD36 plays a role in platelet aggregation, macrophage foam cell development, inflammation, and the tissue ischemia observed in sickle cell disease and cerebral malaria. Note that 1-4% of Japanese and East Asia population lack CD36.

VpreB is expressed in pre-B lymphocytes, but not in mature B cells or in other blood cell lineages. The gene which encodes VpreB maps to human chromosome 22q11.2. The VpreB and Lambda 5 genes encode the Iota and Omega polypeptide chains, respectively, which associate with the Ig-Mu chain to form a molecular complex that is expressed on the surface of pre-B cells. This complex presumably regulates Ig gene rearrangements in the early steps of B cell differentiation. In the mouse the two genes are simultaneously expressed in pre-B cells and belong to the same transcription unit. A primary transcript is synthesized from which the pre-B and Lambda 5 mRNAs are derived by alternative splicing. In the human, however, the two genes are separate and do not belong to the same transcription unit.
