Description:   NCAM2 is an 837 amino acid protein encoded by the human gene NCAM2. NCAM2 contains five immunoglobulin-like domains, two Fibronectin type III domains, a transmembrane domain and a cytoplasmic domain. The gene is expressed most strongly in human adult and fetal brain. NCAM2 is a member of the neural cell adhesion molecule (NCAM) family. NCAMs are closely related cell surface glycoproteins involved in cell to cell interactions during growth and are thought to play an important role in embryogenesis and development. NCAM2 is a considered a good candidate for involvement in certain Down syndrome phenotypes because a slight overexpression of NCAMs increases many-fold the homotypic adhesion properties of cells. Stat5 regulates NCAM2 in vivo by binding to the NCAM2 intron in the NKL natural killer cell line; this binding is induced by cytokines that activate Stat5. Neither Stat1 nor Stat3 bind to this region, despite sharing a consensus binding sequence with Stat5.

Description:   Cell adhesion molecule-related/down-regulated by oncogenes (CDO) and BOC (brother of CDO) are members of the immunoglobulin/fibronectin type III repeat family and act as cell surface receptors. CDO is a component of a cell-surface receptor complex which also contains BOC, NEO1, CTNNB1 and cadherins and which acts as a mediator of cell-cell interactions between muscle cells. CDO and BOC are single pass membrane proteins that play a role in myogenic cell differentiation. Together, CDO and BOC participate in a positive feedback loop with MyoD, a myogenic transcription factor. The 1,242 amino acid rat CDO protein has a 24 residue signal sequence, five Ig V-like repeats, a 25 residue membrane-spanning region, three FNIII-like repeats and a cytoplasmic region of 256 amino acids containing a proline-rich stretch. The human protein contains 1,225 amino acid residues and shares significant homology with the domain structures of the rat protein.

Description:   Vinexin is a 671 amino acid protein that is expressed as two isoforms, designated Vinexin alpha and Vinexin beta. Localized to cell junctions in both the cytoplasm and the cytoskeleton, Vinexin alpha functions to promote Actin stress fiber formation, playing an important role in modification of the Actin cytoskeleton. Like Vinexin alpha, Vinexin beta is localized to cell junctions in the cytoplasm, but is also found in the nucleus where it plays an important role in cell spreading and in activation of the JNK pathway in response to EGF stimulation. Although Vinexin alpha and Vinexin beta have different roles within the cell, both proteins contain three SH3 domains in their carboxy terminus and are expressed in a variety of tissues, including placenta, heart, liver, brain, pancreas and skeletal muscle. Description:   PCDHB11 is a member of the protocadherin beta gene cluster, one of three related gene clusters tandemly linked on chromosome five. PCDHB11 clusters demonstrate an unusual genomic organization similar to that of B-cell and T-cell receptor gene clusters. The beta cluster contains 16 genes and 3 pseudogenes, each encoding 6 extracellular cadherin domains and a cytoplasmic tail that deviates from others in the cadherin superfamily. The extracellular domains interact in a homophilic manner to specify differential cell-cell connections. Description:   G2A (for G2 accumulation) is a seven transmembrane G protein-coupled receptor that is upregulated in response to DNA damage and stress (1). G2A is predominantly expressed in hematopoietic tissues and in hematopoietic stem cells, and it is more highly detected in pro-B cells, while lower expression is observed in immature B cells and pre-B cells (1,2). Ectopic expression of a G2A fusion protein in NIH/3T3 fibroblasts induces a cell cycle arrest that is consistent with a block at the G2/M transition (1,4). G2A is also able to attenuate the proliferative effects of Bcr-Abl, a chimeric tyrosine kinase oncogene, suggesting that G2A possesses anti-oncogenic properties (5). The amino acid sequence of G2A contains a destruction box motif that is consistently observed in cyclins, where it is required for ubiquitination and proteolytic degradation (6).

Inhibits proliferation, migration, and tubule formation of endothelial cells and reduces vascular leakage. May exert a protective function on endothelial cells through an endocrine action. It is directly involved in regulating glucose homeostasis, lipid metabolism, and insulin sensitivity. In response to hypoxia, the unprocessed form of the protein accumulates in the subendothelial extracellular matrix (ECM). The matrix-associated and immobilized unprocessed form limits the formation of actin stress fibers and focal contacts in the adhering endothelial cells and inhibits their adhesion. It also decreases motility of endothelial cells and inhibits the sprouting and tube formation.
