Septin 14, also known as SEPT14, is a 432 amino acid protein that belongs to the septin family. The highly conserved septin family of GTP-binding cytoskeletal proteins is implicated in membrane transport, apoptosis, cell polarity, cell cycle regulation, cytokinesis and other cellular functions. Septins polymerize into heterooligomeric protein complexes that form filaments, and can associate with cellular membranes, actin filaments and microtubules. Septin 14 has a GTPase domain followed by a C-terminal coiled-coil domain characteristic of group II septins. Septin 14 interacted with all septins except those that are members of its phylogenetic cluster, including itself. Septin 14 shares highest identity (67%) with Septin 10. Expressed in testis and more weakly in fetal liver, tonsil and thymus, the Septin 14 protein is not detected in testicular cancer or other normal or cancer cell lines. The Septin 14 gene is conserved in chimpanzee, dog, cow, mouse and rat, and maps to human chromosome 7p11.2.

Ro autoantigens are of clinical significance because directed against them are found in most patients with primary Sjqgren syndrome, subacute cutaneous lupus erythematosus (SLE), neonatal lupus erythematosus, ANA-negative lupus erythematosus, and systemic lupus erythematosus-like disease secondary to homozygous C2 or C4 complement deficiency (1). Ro/SSA is a ribonucleoprotein that binds to auto in 35 to 50% of patients with SLE and in up to 97% of patients with Sjqgren syndrome (2). The Ro/SSA particle consists of a single immunoreactive protein noncovalently bound with one of four small RNA molecules (2). Most anti-Ro/SSA-positive sera detect not only the main protein, but also a smaller Ro/SSA protein (2). The genes which encode the smaller and larger proteins map to human chromosomes 11p15.5 and 1q31, respectively (3?). This MAb is very special because it reacts only with the intact-HCG and not with either free alpha- or free beta-chain of HCG. HCG is a glycoprotein and is composed of two non-identical, non-covalently linked polypeptide chains designated as the alpha and beta subunits. The alpha subunit is identical to that of thyroid stimulating hormone (TSH), follicle stimulating hormone (FSH), and luteinizing hormone (LH). HCG is secreted in large quantities by normal trophoblasts. It is present only in trace amounts in non-pregnant urine and sera but rises sharply during pregnancy. HCG MAb detects cells and tumors of trophoblastic origin such as choriocarcinoma. Large cell carcinoma and adenocarcinoma of the lung demonstrate anti-hCG positivity in 90% and 60% of cases respectively. 20% of lung squamous cell carcinomas are positive. HCG expression by non-trophoblastic tumors may indicate aggressive behavior.

The tumor suppressor PTEN plays an essential role in regulating signaling pathways involved in cell growth and apoptosis and is inactivated in a wide variety of tumors. Protein interacting with PTEN carboxyl terminus 1 (PICT-1), also designated p60 or Glioma tumor suppressor candidate region gene 2 protein, binds to the C-terminus of PTEN and regulates its turnover. Five Ser/Thr residues within the C-terminal segment of PTEN, including Ser-380, are phosphorylated upon binding of PTEN to PICT-1 and may contribute to the stabilization of PTEN. PICT-1 is localized to the nucleus and/or nucleolus and is highly expressed in pancreas and heart, but can also be detected in liver, skeletal muscle, placenta and kidney. PICT-1 also interacts with herpes simplex virus 1 regulatory proteins ICP22 and ICP0. The tumor suppressor GLTSCR2 gene, which encodes PICT-1, is located in a 150-kb minimal common deletion region for human gliomas, especially oligodendrogliomas, and maps to human chromosome 19q13.3.

It is a non-Ig differentiation antigen of B-cells and its expression is restricted to normal and neoplastic B-cells, being absent from all other leukocytes and tissues. CD20 is expressed by pre B-cells and persists during all stages of B-cell maturation but is lost upon terminal differentiation into plasma cells. This MAb can be used for immunophenotyping of leukemia and malignant cells, B lymphocyte detection in peripheral blood and B cell localization in tissues. It reacts with the majority of B-cells present in peripheral blood and lymphoid tissues and their derived lymphomas. In lymphoid tissue, germinal center blasts and B-immunoblasts are particularly reactive. It is a reliable antibody for ascribing a B-cell phenotype in known lymphoid tissues. Rarely, CD20-positive T-cell lymphomas have been reported. Reactivity has also been noted with Reed-Sternberg cells in cases of Hodgkin's disease, particularly of lymphocyte predominant type.

Reacts with viral glycoprotein of rabies virus strains SAD-Vnukovo and Pitman-Moore. This MAb is useful in detecting rabies virus by ELISA and Western. It is capable of neutralizing rabies virus. Rabies virus (Neurotropic virus) is a member of the Rhabdoviridae family. Rabies is a single stranded, neurotropic, negative sense RNA virus which encodes 5 proteins: a glycoprotein, a nucleoprotein, and three others. The mature virus has a bullet shape, a protein coat, and a lipid envelope. The outer surface of the virus is covered with thumb like glycoprotein projections 5-10 nm long and 3 nm in diameter. The virus averages approximately 780 nm in length. Lipid solvents destroy virus infectivity. Rabies virus is a very successful virus, with a very wide range of hosts. It causes an acute, central nervous system infection, characterized by CNS irritation, followed by paralysis and death. Approximately 50,000 human deaths each year are caused by rabies.

This MAb recognizes granulocyte-colony stimulating factor (G-CSF) in the cytoplasm of mature granulocytes. It shows no reactivity with any other cell types. Markers of myeloid cells are useful in the identification of different levels of cellular differentiation. It reacts with early precursor and mature forms of myeloid cells. It is useful for the detection of myeloid leukemias and granulocytic sarcomas. It can be used as a marker of granulocytes in normal tissues or inflammatory processes.G-CSF is a pleiotropic cytokine that influences differentiation, proliferation and activation of the neutrophilic granulocyte lineage. The human G-CSF cDNA encodes a 207 amino acid precursor containing a 29 amino acid signal peptide that is proteolytically cleaved to form a 178 amino acid residue mature protein. Two G-CSF's, which are identical except for a three amino acid deletion in the amino-terminus of one form of the protein have been isolated from human cells. Murine and human G-CSF's share 73% sequence identity at the amino acid level.
