ABT1 (activator of basal transcription 1) is a nuclear protein that associates with the TATA-binding protein (TBP) and enhances basal transcription activity of class II promoters. ABT1 associates with TBP in HeLa nuclear extracts in vitro. Another protein, designated ERF, is a member of the Ets family of transcription factors. The members of the Ets family are grouped because they share a highly conserved DNA binding domain. These factors are involved in growth factor pathways and regulate both proliferation and differentiation. ERF (Ets-2 repressor factor) is a ubiquitously expressed Ets-domain protein that exhibits strong transcriptional repressor activity, suppresses Ets-induced transformation and is regulated by MAPK phosphorylation. ERF transcription may be regulated by Ets-domain proteins. Additionally, modulation of ERF activity is involved in the transcriptional regulation of genes activated during entry into G1 phase.

Hsp20 is a small heat shock protein related to Hsp25, Hsp27 and may form different hetercomplexes with these proteins. The specific physiological function of Hsp20 is not yet known. It is distributed ubiquitously in tissues, but is found in higher levels in skeletal, smooth and heart muscle. Under normal conditions, Hsp20 is diffusely distributed in the cytosol, but under heat stress conditions, it translocates to the nucleus. Unlike other heat shock proteins the amount of Hsp20 does not increase after heat shock. The Hsp20 was demonstrated to constitute up to 1.3% of the total cellular protein in vertebrate tissues, especially in muscle, and its expression is related to muscle contraction, specifically in slow-twitch muscle. Hsp20 may form different heterocomplexes with other Hsp's, such as alpha-crystalline and Hsp25. Phosphorylated form of Hsp20 is proposed to interact with monomeric actin whereas dephosphorylated form binds polymeric actin filaments. In normal conditions Hsp20 is diffusely disturbed in cytosol but under the heat stress it undergoes translocation to membrane fraction.

GCAPs are calcium-The binding proteins belonging to the calmodulin superfamily. GCAP1 is a calcium-binding protein that stimulates synthesis of c-GMP in photoreceptors. GCAP1 is present in rod and cone photoreceptor outer segments where phototransduction occurs. In contrast to other calcium-binding proteins from the calmodulin superfamily, the calcium-free form of GCAP1 stimulates the effector enzyme. By molecular cloning of human and mouse GCAP cDNA, the known mammalian GCAPs are found to be more than 90% similar, consisting of 201 to 205 amino acids, and containing three identically conserved calcium-binding sites. A related protein, GCAP2, is detectable only in the retina and results from a gene duplication event.

The UNC5H family of proteins act as transmembrane receptors for netrin-1 and play a crucial role in axon guidance and migration of neural cells. Additionally, when cleaved by a caspase to produce an intracellular fragment containing a death domain,UNC5H receptors induce apoptosis. This activity is blocked by the binding of netrin-1. In the absence of netrin-1, UNC5H receptors act as tumor suppressors by inhibiting anchorage-independent growth and invasion, but mutation of these receptors provides a potential mechanism for tumorigenicity. Nudel is important for normal cortical development. It is invovled in microtubule organization, nuclear translocation, and neuronal positioning in concert with various other factors (including Lis1, Pafah1b1, Pahfah1b2, dynein, dynorphin A and cdk5). Western blot analysis of mouse tissues shows abundant expression of Nudel in brain and testis, and much lower expression in heart, liver, kidney, and skeletal muscle. In fractionated rat brain, Nudel and Lis1 are both found in fractions enriched for postsynaptic density proteins. Immunostaining of embryonic day 18 mouse brain sections revealed staining of migrating neurons and thalamocortical axons of the intermediate zone of the developing cerebral cortex, as well as several other developing brain regions. The deduced protein contains 345 amino acids and has a calculated molecular mass of 38.4 kDa. It has a coiled coil motif (residues 19 to 201), followed by several potential phosphorylation sites for casein kinase II, protein kinase C or CDK5. Nudel shares about 50% identity with mouse and human NUDE proteins.
