Description:   Purines are critical for energy metabolism, cell signaling and cell reproduction and also function as precursors for coenzymes, energy transfer molecules, regulatory factors and proteins involved in RNA and DNA synthesis. GART (GAR transformylase), also referred to as AIRS, GARS, PAIS, PGFT, PRGS or GARTF, is 1,010 amino acids in length and is a key folate-dependent trifunctional enzyme with phosphoribosylglycinamide formyltransferase, phosphoribosylglycinamide synthetase and AICAR (phosphoribosylaminoimidazole synthetase) activity required for de novo purine biosynthesis. Cancer cells require considerable amounts of purines to sustain their accelerated growth and GART is, therefore, a target for cancer chemotherapy. GART is highly conserved in vertebrates. Two isoforms of GART are expressed due to alternative splicing events.

Site-specific RNA editing of transcripts encoding these proteins results in amino acid substitutions which consequently alters their functional activities. Exhibits low-level editing at the GRIA2 Q/R site, but edits efficiently at the R/G site and HOTSPOT1. Its viral RNA substrates include: hepatitis C virus (HCV), vesicular stomatitis virus (VSV), measles virus (MV), hepatitis delta virus (HDV), and human immunodeficiency virus type 1 (HIV-1). Exhibits either a proviral (HDV, MV, VSV and HIV-1) or an antiviral effect (HCV) and this can be editing-dependent (HDV and HCV), editing-independent (VSV and MV) or both (HIV-1). Impairs HCV replication via RNA editing at multiple sites. Description:   Growth/differentiation factors (GDFs) are members of the TGF superfamily (1,2). Members of the TGF superfamily are involved in embryonic development and adult tissue homeostasis (1). GDF-1 expression is almost exclusively restricted to the central nervous system and mediates cell differentiation events during embryonic development (3). Neither GDF-3 (Vgr-2) nor GDF-9 contains the conserved cysteine residue which is found in most other TGF superfamily members. GDF-3 is detectable in bone marrow, spleen, thymus and adipose tissue, whereas GDF-9 has only been detected in ovary (4). GDF-5 (also designated CDMP-1) has been shown to induce activation of plasminogen activator, thereby inducing angiogenesis. It is predominantly expressed in long bones during fetal embryonic development and is involved in bone formation. (5). GDF-5 mutations have been identified in mice with the mutation brachypodism (bp), a mutation which affects the length and number of bones in limbs (6). Description:   The members of the murine Cdx family (Cdx1, Cdx2, and Cdx4) are members of the caudal-type homeobox family of genes, which are homologues of the Drosophila caudal? gene required for anterior-posterior regional identity. The intestine-specific transcription factors Cdx1 and Cdx2 are candidate genes for directing intestinal development, differentiation, proliferation and maintenance of the intestinal phenotype. The relative expression of Cdx1 to Cdx2 protein may be important in the anterior to posterior patterning of the intestinal epithelium and in defining patterns of proliferation and differentiation along the crypt-villus axis. Expression of the Cdx1 homeobox gene in epithelial intestinal cells promotes cellular growth and differentiation. Cdx1 positively regulates its own expression. Cdx1 and Cdx2 are expressed in the small intestine and colon of fetus and adult. A decrease in human Cdx1 and/or Cdx2 expression is associated with colorectal tumorigenesis. Both Cdx1 and Cdx2 genes must be expressed to reduce tumorigenic potential, to increase sensitivity to apoptosis and to reduce cell migration, suggesting that the two genes control the normal phenotype by independent pathways. The human Cdx1 gene maps to chromosome 5q31-q33 and encodes a 265-amino acid protein.

Description:   Inositol polyphosphate multikinase (IPMK) belongs to the inositol phosphokinase (IPK) family and is characterized as having a broad substrate specificity. However, IPMK displays a preference for inositol-1,4,5-trisphosphate (Ins(1,4,5)P3) and inositol 1,3,4,6-tetrakisphosphate (Ins(1,3,4,6)P4). IPMK is ubiquitously expressed with the highest expression in skeletal muscle, liver, placenta, lung, peripheral blood leukocytes, kidney, spleen and colon. IPMK is localized to the nucleus, where it may play a role in the regulation of calcium release from intracellular stores and has been implicated as a drug target for cancer therapies. The gene encoding IPMK maps to human chromosome 10, which contains over 800 genes. Notably, disorders linked to genes on chromosome 10 include Cowden syndrome, Cockayne syndrome and Tetrahydrobiopterin deficiency.
