Description:   Human Vitronectin/VTN is a cell adhesion and spreading factor. It can be found in the blood and the extracellular matrix (ECM). Vitronectin interacts with glycosaminoglycans and proteoglycans. The multimeric Vitronectin can efficiently bind to and incorporate into the ECM; Vitronectin can support cell adhesion through binding to various integrins and other proteoglycans. Vitronectin can be recognized by certain members of the integrin family and serves as a cell-to-substrate adhesion molecular. It can as a inhibitor of the membrane-damaging effect of the terminal cytolytic complement pathway. Vitronectin contains an endogenous cleavage site, plus cleavage sites for elastase, thrombin, and plasmin.

Description:   Dickkopf-related protein 3 (DKK3) belongs to the DKK protein family including Dkk-1, 2, 3 and -4. DKK3 is a 350 amino acid secreted glycoprotein which is comprised of an N-terminal signal peptide and 2 conserved cysteine-rich domains that are separated by a 12 amino acid linker region. Dkk-3 also have one prokineticin domain. DKK3 is involved in embryonic development through its inhibition of the WNT signaling pathway. The Dkk family also includes Soggy, which is homologous to Dkk-3 but not to the other family members. Soggy has not been shown to inhibit Wnt signaling, and its role in the pathway is unclear.

Description:   Interleukin-1 receptor antagonist protein (Il1rn), also known as IL-1ra, IRAP or IL1 inhibitor, is a member of the interleukin 1 cytokine family. This protein inhibits the activities of interleukin 1 alpha (IL1A) and interleukin 1 beta (IL1B), and modulates a variety of interleukin 1 related immune and inflammatory responses. The mouse Il1rn gene encodes a 178 amino acids (aa) protein with a 26 aa signal peptide. Mouse Il1rn protein shares 26% and 19% identity with its homologues IL-1 beta and IL-1 alpha, respectively. Il1rn can Inhibits the activity of interleukin-1 by binding to receptor IL1R1 and preventing its association with the coreceptor IL1RAP for signalling, but has no interleukin-1 like activity. Recently, an recombinant human Il1rn protein is used in the treatment of rheumatoid arthritis, an autoimmune disease in which IL-1 plays a key role.

Description:   Mouse interleukin 13 (mIL-13) is a pleiotropic cytokine produced by activated Th2 cells. IL-13 induces B cell proliferation and immunoglobin production. It contains a four helical bundle with two internal disulfide bonds. Mouse IL13 shares 58% sequence identity with human protein and exhibits cross-species activity. IL13 signals via receptor IL13R (type2, IL4R) and activates STAT-6. IL13 initially binds IL-13R alpha1 with low affinity and triggers association of IL4R alpha, generating a high affinity heterodimeric receptor IL13R and eliciting downstream signals. Description:   Vascular endothelial growth factor (VEGF/VEGF-A ) is originally known as vascular permeability factor (VPF). It belongs to the PDGF family with a cysteine-knot structure comprised of eight conserved cysteine residues, and reckoned as a potent mediator in the process of angiogenesis and vasculogenesis in either fetus or adult. VEGF is particularly expressed in supraoptic , paraventricular nuclei and the choroid plexus of the pituitary, and abundant in the corpus luteum of the ovary and in kidney glomeruli. The rat VEGF protein contains a putative 20 amino acids (aa) signal peptide, and alternative splicing of rat VEGF gene produces isoforms of 120, 144, 164 and 188 aa. Rat VEGF164 respectively displays 97% and 88% aa identity with that regions of mouse and human VEGF. VEGF can bind to the FLT1/VEGFR1 and KDR/VEGFR2 receptors, heparan sulfate and heparin, and play important roles in inducing endothelial cell proliferation, promoting cell migration, inhibiting apoptosis and inducing permeabilisation of blood vessels.

Description:   Programmed Death-1 (PD-1), firstly cloned from mouse T cell hybridoma 2B4.11, is one member of CD28/CTLA-4 superfamily. PD-1 belongs to type I transmembrane protein and acts as an important immunosuppressive molecule. This family also include members of CD28, CTLA-4 and ICOS.The mouse Programmed Death-1 protein, encoded by PD-1 gene, comprises four parts including a putative 20 aa signal peptide, a 149 aa extracellular region, a 21 aa transmembrane domain and a 98 aa cytoplasmic region. The cytoplamsic tail of PD-1 contains two structural motifs, an immunoreceptor tyrosine-based inhibitory motif (ITIM) and an immunoreceptor tyrosine-based switch motif (ITSM) formed by two tyrosine residues which make the difference in PD-1 signal mediating. Mouse PD-1 is expressed in thymus and shares about 69% aa sequence identity with human PD-1. Recently, programmed death-1 (PD-1) with its ligands, programmed death ligand B7H1 (PD-L1) and B7DC (PD-L2), was found to regulate T-cell activation and tolerance, upon ligand binding, inhibiting T-cell effector functions in an antigen-specific manner. PD-1 gene knocked out mice would induce some autoimmune diseases, which suggests that PD-1 acts as a co-inhibitory molecule actively participating in maintaining peripheral tolerance. Thus, PD-1 may be a useful target for the immunologic therapy of carcinoma,infection,autoimmune diseases as well as organ transplantation.

Description:   CCL5, also known as RANTES (Regulated upon Activation Normal T cell Expressed and Secreted), is an 8kDa β-chemokine that plays a primary role in the inflammatory immune response by means of its ability to attract and activate leukocytes. Human and mouse RANTES exhibit cross-species activity on human and mouse cells. CCL5 is secreted by many cell types at inflammatory sites and it exerts a wide range of activities through the receptors CCR1, CCR3, CCR4 and CCR5. In humans, CCR5 binding to CCL5 inhibits the infectivity of R5 (M-tropic) but not X4 (T-tropic) strains of HIV-1. The deposition of CCL5 on activated vascular endothelial cells is crucial for monocyte adhesion to damaged vasculature, but CCL5 oligomerisation is not required for the extravasation of adherent leukocytes. CCL5 is upregulated in breast cancer and promotes tumour progression through the attraction of proinflammatory macrophages in addition to its actions on tumour cells, stromal cells and the vasculature.

Description:   CCL22, also named stimulated T cell chemotactic protein (STCP1), is a CC chemokine initially isolated from clones of monocyte-derived macrophages and binds to the receptor CCR4. Human CCL22 is highly expressed in macrophage and in monocyte-derived dendritic cells and thymus. It is also found in lymph node, appendix, activated monocytes, resting and activated macrophages. May play a role in the trafficking of activated/effector T-lymphocytes to inflammatory sites and other aspects of activated T-lymphocyte physiology. Description:   CD112 is a type I transmembrane glycoprotein belonging to the Immunoglobulin superfamily. It comprises one Ig-like V-type domain and two Ig-like C2-type domains in the extracellular region. The V domain is believed to mediate nectin binding to its ligands. Nectin2 is known to bind the pseudorabies virus, and herpes simplex virus2 (HSV2), involving in cell to cell spreading of these viruses. It does not bind poliovirus. As a homophilic adhesion molecule, CD112 is found concentrated in adherens junctions, and exists on neurons, endothelial cells,epithelial cells and fibroblasts. CD112 has been identified as the ligand for DNAM-1 (CD226), and the interaction of CD226/CD112 mediates cytotoxicity and cytokine secretion by T and NK cells. The costimulatory responses may be a critical component in allergic reactions and may therefore become targets for anti-allergic therapy.

Description:   Mouse CD83 is a 30-35kDa member of the Siglec (or sialic-acid-binding immunoglobulin-like lectin) family of transmembrane proteins. CD83 is a primary marker for dendritic cells. It is also found on B cells, neutrophils, monocytes and macrophages. Except for dendritic cells, CD83 expression is often transient. CD83 binds to sialic acids on target cells. Membrane CD83 promotes T cell proliferation, particularly of CD8+ cytotoxic T cells. Soluble CD83 is immunosuppressive and blocks T cell activation. On monocytes, CD83 is suggested to drive monocytes into a fibrocyte phenotype. A lack of membrane-expressed CD83 leads to an unusual IL-4/ IL-10 producing CD4+ T cell phenotype.

ICOSLG contains Ig-like C2-type (immunoglobulin-like) domain and 1 Ig-like V-type (immunoglobulin-like) domain. ICOSLG acts as a costimulatory signal for T-cell proliferation and cytokine secretion, it also induces B-cell proliferation and differentiation into plasma cells. It could play an important role in mediating local tissue responses to inflammatory conditions, as well as in modulating the secondary immune response by co-stimulating memory T-cell function.
