Description:
The Notch signaling pathway is an evolutionary conserved system that is involved in intracellular communication. Notch receptors play an important role in development and cell-fate decisions. Notchless is a loss-of-function mutant allele that encodes for protein NLE1 (notchless homolog 1). NLE1 is a 485 amino acid WD40-repeat protein that binds to the cytoplasmic domain of Notch, regulating its signaling activity in Drosophila melanogaster and in mice. Deletion of the NLE1 gene in mice during the early stages of development results in embryonic death, while gene deletion in the late stages of development leads to activation of a caspase-3-dependent apoptotic pathway. In plants, NLE1 is crucial for normal cellular growth and development. Under-expression during shoot proliferation causes pleiotropic defects such as delayed flowering and abnormal organ maturation. It may also play a role in 60S ribosomal subunit biogenesis in yeast. NLE1 contains eight WD40 domains and produces one isoform due to alternative splicing.

DOCK 5 is a 1,870 amino acid protein belonging to the DOCK family of cytokinesis-regulating proteins. This cytoplasmic peripheral membrane protein activates Rac 1 and Rac 2 small GTPases, while presumably acting as a guanine nucleotide exchange factor (GEF), which exchanges bound GDP for free GTP. DOCK 5 contains one DHR-1 (CZH-1) domain, one DHR-2 (CZH-2) domain and one SH3 domain. The DHR-2 domain is a putative GEF activity mediator. In mice, spontaneous mutation of the gene encoding DOCK 5 leads to deletion of the DHR-1 domain, which functions to bind phospholipids and assists in protein-protein interactions, resulting in rupture of lens cataract (RLC). Due to siRNA knockdown studies, it is suspected that DOCK 5 may also be an important mediator of CrkII/CrkL regulation of Caco-2 migration and spreading on COL4. There are two isoforms of DOCK 5 that exist as a result of alternative splicing events.

FANK1 (fibronectin type III and ankyrin repeat domains 1), also known as HSD13, is a 345 amino acid nuclear and cytoplasmic testis-specific protein found primarily in pachytene spermatocytes and round spermatids. Containing six ANK repeats and a single fibronectin type-III domain, FANK1 undergoes alternative splicing events to form three isoforms. Possessing DNA binding activity, FANK1 is suggested to act as a transcription factor and may regulate gene expression during spermatogenesis. The gene encoding FANK1 maps to human chromosome 10, which contains over 800 genes and 135 million nucleotides, making up nearly 4.5% of the human genome. PTEN is an important tumor suppressor gene located on chromosome 10 and, when defective, causes a genetic predisposition to cancer development known as Cowden syndrome.

Protein kinase C (PKC) is a family of serine and threonine specific protein kinases that can be activated by calcium and the second messenger diacylglycerol. PKC family members phosphorylate a wide variety of protein targets and are known to be involved in diverse cellular signaling pathways. PKC family members also serve as major receptors for phorbol esters, a class of tumor promoters. Each member of the PKC family has a specific expression profile and is believed to play a distinct role. The protein encoded by this gene is one of the PKC family members. This kinase can be activated rapidly by the agonists of G protein coupled receptors. It resides in both cytoplasm and nucleus, and its nuclear accumulation is found to be dramatically enhanced in response to its activation. This kinase can also be activated after B cell antigen receptor (BCR) engagement, which requires intact phopholipase C gamma and the involvement of other PKC family members.
