Description:
IL-2R is a heterotrimeric protein binds and responds to the cytokine IL-2. Three distinct chains of IL-2R, termed as α, β and γ, which are non-covalently associated are identified. The α and β chains are involved in binding IL-2, while signal transduction following cytokine interaction is carried out by the γ chain, along with the β subunit. The γ chain of the IL-2R can bind to the β chain before receptor interaction with IL-2. The γ chain alone has a very weak affinity for IL-2, but after the ligand is bound to the α/β heterodimer, the γ chain becomes recruited to the complex to form a very stable macromolecular quaternary ligand/receptor complex.
Interleukin-2 receptor subunit gamma (IL2RG), also known as cytokine receptor common subunit gamma, CD antigen CD132, gammaC, p64, which belongs to the type I cytokine receptor family or type 5 subfamily. IL2RG is located on the surface of immature blood-forming cells in bone marrow. Defects in IL2RG are the cause of severe combined immunodeficiency X-linked T-cell-negative/B-cell-positive/NK-cell-negative (XSCID).

Description:
Mesothelin is a cell surface glycoprotein whose expression is limited to mesothelial cells of the serosa (pleura, pericardium, and peritoneum) and epithelial cells of the trachea, tonsils, fallopian tube, and kidneys. Mesothelin plays an important role in cell survival, proliferation, migration, invasion, tumour progression, and resistance to chemotherapy. The overexpression of mesothelin can activate NF-κB and signal transducer and activator of transcription 3 (Stat3), inhibit apoptotic signalling and TNF- alpha-induced apoptosis, and accelerate the G1–S transition. Mesothelin is also found overexpressed in various cancers, including malignant mesothelioma, pancreatic or ovarian carcinoma, sarcomas and in some gastrointestinal or pulmonary carcinomas. As a result of its limited expression in normal tissues, mesothelin has been reported as an ideal tumour-associated marker for the development of targeted therapy.

Description:
Interleukin-27 (IL-27) is a heterodimeric group 2 receptor ligand molecule that belongs to the IL-6/IL-12 family of long type I cytokines. It is composed of EBI3 (EBV-induced gene 3), a 34 kDa glycoprotein that is related to the p40 subunit of IL-12 and IL-23, and p28, the cloned 28 kDa glycoprotein that is related to the p35 chain of IL-12. IL-27 is expressed by monocytes, endothelial cells and dendritic cells. IL-27 binds to and signals through a heterodimeric receptor complex composed of WSX1 (TCCR) and gp130. Evidence suggests IL-27 interacts only with WSX-1. IL-27 has both anti- and proinflammatory properties. As an antiinflammatory, IL-27 seems to induce a general negative feedback program that limits T and NK-T cell activity. At the onset of infection, IL-27 induces an IL-12 receptor on naie CD4+ T cells, making them susceptible to subsequent IL-12 activity (and possible Th1 development). Notably, IL-12 family cytokines are both induced and inhibited by bacterial products. Microbes promote IL-27 secretion through TLR4, and also block IL-27 production via C5a induction.

CD40 is a Type I Transmembrane Glycoprotein that belongs to the TNF Receptor Superfamily. CD40 is expressed in B cells, follicular dendritic cells, dendritic cells, activated monocytes, macrophages, endothelial cells, vascular smooth muscle cells, and several tumor cell lines. The extracellular domain of CD40 is characterised by Cysteine rich repeat regions. Interaction of CD40 with its ligand (CD40L) leads to aggregation of CD40 molecules, which in turn interact with cytoplasmic components to initiate signalling pathways. Several different TRAF proteins (adaptor proteins) have been identified to serves as mediators of the signal transduction. CD40 plays an essential role in mediating a broad variety of immune and inflammatory responses including T cell-dependent immunoglobulin class switching, memory B cell development, and germinal center formation.

ICAM-2 is a 55-65 kD transmembrane glycoprotein possessing 2 extracellular Ig domains, a single transmembrane domain, and a short 26-amino acid cytoplasmic domain. ICAM-2 is expressed on most leukocytes, and is strongly expressed on vascular endothelial cells. Interactions of ICAM-2 and the integrin receptors mediate cell adhesion in a wide range of lymphocyte, monocyte, natural killer cell, and granulocyte with other cells, and play important roles in many adhesion-dependent immune and inflammation responses, such as T cell aggregation, NK-cell cytotoxicity and migration, lymphocyte recirculation, etc. Serum levels of ICAM-2 correlated significantly with the inflammatory and course sequences of trichinosis in mice and had a similar relation with blood eosinophilia. So, estimation of ICAM-2 serum levels may prove useful in diagnosis of trichinosis recent infections, and in monitoring the prognosis and response to treatment.

Interleukin-33 (IL-33; HF-NEV; IL-1F11), a member of the IL-1 family of cytokines, is expressed by many cell types following pro-inflammatory stimulation and is thought to be released on cell lysis. The 30kDa human IL33 is converted by CASP1 to a 18kDa protein. IL33 binds to and signals through ST2 (IL1R1) and its stimulation recruits MYD88, IRAK, IRAK4, and TRAF6, followed by phosphorylation of ERK1 (MAPK3)/ERK2 (MAPK1), p38 (MAPK14), and JNK. Chemokine (C-X-C Motif) Ligand 9 (CXCL9) belongs to the intercrine alpha (chemokine CXC) family. It is secreted by interferon stimulated monocytes, macrophages and endothelial cells, which elicits chemotactic functions by interacting with the chemokine receptor CXCR3. CXCL9 acts as a Th1 (type 1 helper T) cell chemoattractant and plays a role in the growth, activation and movement of cells associated with immune and inflammatory responses, and in tumour growth inhibition.
