Description:
Interleukin-35 is a newest and potent suppressive cytokine of the IL-12 family. Unlike other members of the IL-12 family which are produced mainly by antigen-presenting cells (APCs), IL-35 has been identified as a potentially immunosuppressive cytokine primarily produced by regulatory T (Treg) cells, and to a lesser extent by activated dendritic cells and macrophages. IL-35 is composed of the IL-12 p35 subunit and the IL-27 Epstein–Barr virus-induced protein 3 (EBI3) subunit. IL-35 plays an important role in many disease models and has been recently shown to have additional functions aside from inhibition of proliferation, including inducing its own expression in non-Treg cells.

CD274, also known as B7-H1 or programmed death ligand 1 (PD-L1), is a 40 kD type I transmembrane protein and a member of the B7 family within the immunoglobulin receptor superfamily. Programmed death-1 ligand-1 (PD-L1, CD274, B7-H1) has been identified as the ligand for the immunoinhibitory receptor programmed death-1(PD1/PDCD1) and has been demonstrated to play a role in the regulation of immune responses and peripheral tolerance. By binding to PD1 on activated T-cells and B-cells, PD-L1 may inhibit ongoing T-cell responses by inducing apoptosis and arresting cell-cycle progression. Accordingly, it leads to growth of immunogenic tumour growth by increasing apoptosis of antigen specific T cells and may contribute to immune evasion by cancers. PD-L1 thus is regarded as promising therapeutic target for human autoimmune disease and malignant cancers.

erebral Dopamine Neurotrophic Factor (CDNF), also known as ARMETL1 (ARMET-like protein 1), is a secreted protein with eight conserved cysteine residues.It is belongs to the ARMET family. CDNF/ARMETL1 is a evolutionary conserved protein which can protect and restore the function of dopaminergic neurons in the rat model of Parkinson's disease, suggesting that CDNF might be beneficial for the treatment of Parkinson's disease. CDNF is widely expressed in neurons in several brain regions including cerebral cortex, hippocampus, substantia nigra, striatum and cerebellum. Human CDNF is glycosylated and secreted from transiently transfected cells. CDNF promotes the survival, growth, and function of dopamine-specific neurons and is expressed in brain regions that undergo cocaine-induced neuroplasticity.

Coronin 6, a newly identified member of the coronin family, is highly enriched at adult NMJs and regulates AChR clustering via modulating the interaction between receptors and the actin cytoskeletal network. Coronins are a family of conserved actin-binding proteins originally identified in the actin-rich structure of the amoeba Dictyostelium discoideum . To date, seven members of coronins have been identified in mammals, and most exhibit tissue-specific distribution patterns. Coronin 6 is prominently expressed in adult muscle and enriched at the NMJ. Studies with cultured myotubes reveal that Coronin 6 regulates both agrin- and laminin-induced AChR clustering and is important for anchoring AChRs onto the actin cytoskeleton. Also, both the C-terminal region and a conserved Arg29 residue at the N terminus of Coronin 6 are essential for its actin-binding activity and stabilisation of AChR–cytoskeleton linkage. Importantly, in vivo knockdown of Coronin 6 in mouse skeletal muscle fibers leads to destabilisation of AChR clusters, which demonstrates that Coronin 6 is a critical regulator of AChR clustering at the postsynaptic region of the NMJs through modulating the receptor-anchored actin cytoskeleton. The human Coronin 6 has five isoforms produced by alternative splicing, and tissue-specific expression of these isoforms are unclear.

Neutrophil cytosol factor 1( NCF1) is a 47 kDa cytosolic subunit of neutrophil NADPH oxidase. This oxidase is characterised as a multicomponent enzyme which is activated to produce superoxide anion. NCF2, NCF1, and a membrane bound cytochrome b558 are required for the activation of the latent NADPH oxidase. The human NCF1 gene encodes a 390 amino acids protein without a signal peptide. The NCF1 gene interacts with other subunits of nicotinamide adenine dinucleotide phosphate-oxidase (NADPH) and plays an important role in innate immunity, producing reactive oxygen species and reducing the severity and duration of parasitic infection and autoimmune disease. NCF1 also has a role in T cell activation.

Endothelial protein C receptor (PROCR) is also known as Activated protein C receptor (APC receptor), Endothelial cell protein C receptor, CD antigen CD201. PROCR / EPCR / CD201 is a N-glycosylated protein, with a soluble form exists. PROCR is probably released by a metalloprotease. PROCR seems to have the same activity as the membrane-bound form. PROCR / EPCR / CD201 is expressed strongly in the endothelial cells of arteries and veins in heart and lung. PROCR binds activated protein C and enhances protein C activation by the thrombin - thrombomodulin complex. PROCR plays a role in the protein C pathway controlling blood coagulation.

Cluster of differentiation 14 (CD14), is a cell surface glycoprotein, and is a is a component of the innate immune system. CD14 is a myelomonocytic differentiation antigen preferentially expressed on monocytes, macrophages, and activated granulocytes. CD14 exists in two forms. Either it is anchored into the membrane by a glycosylphosphatidylinositol tail (mCD14) or it appears in a soluble form (sCD14). Soluble CD14 either appears after shedding of mCD14 (48 kDa) or is directly secreted from intracellular vesicles (56 kDa). CD14 acts as a co-receptor (along with the Toll-like receptor TLR 4 and MD-2) for the detection of bacterial lipopolysaccharide (LPS). CD14 can bind LPS only in the presence of lipopolysaccharide-binding protein (LBP). CD14 has been proposed to be involved in various biological processes, including transportation of other lipids, cell-cell interaction during different immune responses, as well as recognition of apoptotic cells. Although LPS is considered its main ligand, CD14 also recognizes other pathogen-associated molecular patterns. CD14+ cells are monocytes that can differentiate into a host of different cells. CD14 has been shown to interact with Lipopolysaccharide-binding protein.
