Description:
Programmed cell death protein 1(PDCD1) is a single-pass type I membrane protein and contains 1 Ig-like V-type domain. PD-1 is a member of the extended CD28/CTLA-4 family of T cell regulators. PDCD1 inhibits the T-cell proliferation and production of related cytokines including IL-1, IL-4, IL-10 and IFN- gamma by suppressing the activation and transduction of PI3K/AKT pathway. In addition, coligation of PDCD1 inhibits BCR-mediating signal by dephosphorylating key signal transducer. PDCD1 has been suggested to be involved in lymphocyte clonal selection and peripheral tolerance, and thus contributes to the prevention of autoimmune diseases. As a cell surface molecule, PDCD1 regulates the adaptive immune response. Engagement of PD-1 by its ligands PD-L1 or PD-L2 transduces a signal that inhibits T-cell proliferation, cytokine production, and cytolytic function.

Description:
T cell immunoreceptor with Ig and ITIM domains (TIGIT), also called VSIG9 and Vstm3, is a member of the CD28 family within the Ig superfamily of proteins. TIGIT contains an immunoglobulin variable domain, a transmembrane domain and an immunoreceptor tyrosine-based inhibitory motif (ITIM), and is expressed on regulatory, memory, activated T cells and NK cells. TIGIT binds to CD155(PVR) that appear on dendritic cells (DC), macrophages and endothelium with high affinity, and CD112(PVRL2) with lower affinity, but not CD113 (PVRL3). TIGIT-Fc fusion protein could interact with PVR on DC and enhance the secretion of IL-10, but inhibit the macrophage activation. Mice lacking TIGIT show increased T cell responses and susceptibility to autoimmune challenges, while knockdown of TIGIT with siRNA in human memory T cells did not affect T cell responses.

Complement is defined as key part of innate immunity and as the first line of defence in the fight against invading pathogens. Complement 3 (C3) is the most abundant component of the complement cascade and the convergent point for all three major complement activation pathways: namely classical, alternative and mannose-binding lectin pathways. Complement activation leads to the formation of the C3 convertase, which cleaves C3 into the key effector molecules, C3a (anaphylatoxin) and C3b (opsonin) which then drive microbe removal. By binding to C3a receptor (C3aR), C3a exhibits potent anaphylatoxin activity, including increased vascular permeability, triggering degranulation of mast cells, inflammation, and activating leukocytes.

CD300A consists of a 163 amino acid (aa) extracellular domain (ECD) with one Ig-like V- type domain, a 21 amino acid transmembrane segment, and a 98 amino acid cytoplasmic domain with tyrosine residues. CD300A is expressed not only by natural killer (NK) cells but also by T-cell subsets, B-cells, dendritic cells, mast cells, granulocytes and monocytes. CD300A is an inhibitory receptor which may contribute to the down-regulation of cytolytic activity in natural killer (NK) cells, and to the down-regulation of mast cell degranulation.

Amphoterin-Induced Protein 2 (AMIGO2) is a single-pass type I membrane protein which belongs to the AMIGO family of immunoglobulin superfamily. Mature AMIGO2 contains an Ig-like C2-type (immunoglobulin-like) domain, 6 LRR (leucine-rich) repeats, a LRRCT domain, as well as a LRRNT domain. AMIGO2 is mainly expressed in in breast, ovary, cervix, and uterus, although lower in lung, colon, and rectum. AMIGO2 required for depolarization-dependent survival of cultured cerebellar granule neurons. AMIGO2 may mediate homophilic as well as heterophilic cell-cell interaction with AMIGO1 or AMIGO3. Cytotoxic T-lymphocyte protein 4, is a single-pass type I membrane protein. It is widely expressed with highest levels in lymphoid tissues. CD28 and CTLA-4, together with their ligands, B7-1 and B7-2, constitute one of the dominant costimulatory pathways that regulate T and B cell responses. CD28 and CTLA-4 are structurally homologous molecules that are members of the immunoglobulin (Ig) gene superfamily. CTLA4 transmits an inhibitory signal to T cells, whereas CD28 transmits a stimulatory signal. Intracellular CTLA4 is also found in regulatory T Cells and may play an important role in their functions. Tcell activation through the Tcell receptor and CD28 leads to increased expression of CTLA4.

Retinoic acid receptor responder protein 2(RARRES2) is a secreted protein that in humans is encoded by the RARRES2 gene. It is highly expressed in skin, also found in pancreas, liver, spleen, prostate, ovary, small intestine and colon. It is a chemoattractant protein that acts as a ligand for the G protein-coupled receptor CMKLR1. RARRES2 is secreted in an inactive form as prochemerin and is activated through cleavage of the C-terminus by inflammatory and coagulation serine proteases. It is thought to act as a cell surface receptor, found to stimulate chemotaxis of dendritic cells and macrophages to the site of inflammation. RARRES2 is inhibited in psoriatic lesions,it is activated by tazarotene in skin rafts and in the epidermis of psoriatic lesions.

It contains an ephrin RBD (ephrin receptor-binding) domain, and expressed in heart, placenta, lung, liver, skeletal muscle, kidney and pancreas. Ephrin-B1 is cell surface transmembrane ligand for Eph receptors, a family of receptor tyrosine kinases which are crucial for migration, repulsion and adhesion during neuronal, vascular and epithelial development. It binds promiscuously Eph receptors residing on adjacent cells, leading to contact-dependent bidirectional signalling into neighboring cells. Ubiquitin Carboxyl-Terminal Hydrolase 14 (USP14) belongs to the ubiquitin-specific processing (USP) family which is a deubiquitinating enzyme (DUB) with His and Cys domains. USP14 located in the cytoplasm is a proteasome-associated deubiquitinase which releases ubiquitin from the proteasome targeted ubiquitinated proteins. USP14 acts also as a physiological inhibitor of endoplasmic reticulum-associated degradation (ERAD) under the non-stressed condition by inhibiting the degradation of unfolded endoplasmic reticulum proteins via interaction with ERN1. In addition, USP14 is indispensable for synaptic development and function at neuromuscular junctions, required for the degradation of the chemokine receptor CXCR4 which is critical for CXCL12-induced cell chemotaxis.

CD152 and CD28, together with their ligands B7-1 and B7-2, constitute one of the dominant costimulatory pathways that regulate T and B cell responses. CD152 and CD28 are structurally homologous molecules that are members of the immunoglobulin (Ig) gene superfamily. Both CD152 and CD28 are composed of a single Ig V-like extracellular domain, a transmembrane domain and an intracellular domain. CD152 and CD28 are both expressed on the cell surface as disulphide-linked homodimers or as monomers. CD152 was originally identified as a gene that was specifically expressed by cytotoxic T lymphocytes. However, CD152 transcripts have since been found in both Th1 and Th2, and CD4+ and CD8+ T cell clones. Whereas, CD28 expression is constitutive on the surfaces of 95% of CD4+ T cells and 50% of CD8+ T cells and is down regulated upon T cell activation, CD152 expression is upregulated rapidly following T cell activation and peaks approximately 24 hours following activation. Although both CD152 and CD28 can bind to the same ligands, CD152 binds to B71 and B72 with 20-100-fold higher affinity than CD28.

Description:
CD134 (OX40) is a T cell activation antigen structurally belonging to a lymphocyte-specific subgroup of the nerve growth factor and tumour necrosis factor receptor superfamily, which also includes the T cell antigen CD27, B cell antigen CD40, FAS antigen and the T cell activation antigen 4-1BB. The human CD134 protein is expressed only on activated CD4+ T blasts, and its ligand has been identified as gp34. Interactions between CD134 and its ligand in vivo are necessary for the differentiation of activated B cells into highly immunoglobulin-producing cells, however not involved in other pathways of antigen-driven differentiation of B cells such as development of memory cells in the germinal centers. In addition, the CD134 and gp34 system directly mediate adhesion of activated T cells to vascular endothelial cells, and contribute to growth stimulation of the virus-infected T cells.
