Syntaxins were originally thought to be docking proteins, but have more recently been categorized as anchoring proteins that anchor themselves to the cytoplasmic surfaces of cellular membranes. Syntaxins bind to various proteins involved in exocytosis, including VAMPs (vesicle-associated membrane proteins), NSF (N-ethylmaleimide-sensitive factor), SNAPs (soluble NSF attachment proteins) and Synaptotagmin. Endobrevin, also designated VAMP-8 or ED, is a 100 amino acid single-pass type IV membrane protein that belongs to the synaptobrevin family. Similar in sequence to the synaptobrevins, endobrevin is abundantly expressed in kidney, moderately expressed in heart and spleen, and slightly expressed in brain, thymus and liver. Endobrevin interacts specifically with the SNAPs, most likely through an endobrevin-containing SNARE complex.

Ligand binding at the cell surface induces dimerization and autophosphorylation of AXL. Other downstream substrate candidates for AXL are CBL, NCK2, SOCS1 and TENC1. Recruitment of GRB2 and phosphatidylinositol 3 kinase regulatory subunits by AXL leads to the downstream activation of the AKT kinase. GAS6/AXL signaling plays a role in various processes such as endothelial cell survival during acidification by preventing apoptosis, optimal cytokine signaling during human natural killer cell development, hepatic regeneration, gonadotropin-releasing hormone neuron survival and migration, platelet activation, or regulation of thrombotic responses. Plays also an important role in inhibition of Toll-like receptors (TLRs)-mediated innate immune response. In case of filovirus infection, seems to function as a cell entry factor.

Possessing glycerophosphoinositol inositolphosphodiesterase activity, GDE3 hydrolyzes glycerophosphoinositol to form inositol 1-phosphate and glycerol, and is suggested to play a role in Actin cytoskeleton remodeling and osteoblast differentiation and growth. A multi-pass membrane protein, GDE3 localizes to cell membrane and cytoplasm, and colocalizes with actin in the cytoskeleton. GDE3 contains one GDPD domain, binds calcium as a cofactor and is encoded by a gene mapping to human chromosome Xq13.1.

Localized to the membrane of the endoplasmic reticulum (ER), UBE2J2 catalyzes the attachment of ubiquitin to misfolded membrane proteins, thereby targeting them for proteasomal destruction. This ATP-dependent reaction yields AMP, a diphosphate and a ubiquitin-tagged protein and may be a method of quality control within the ER. Two isoforms of UBE2J2 exist due to alternative splicing events.

The Src family of protein tyrosine kinases act in signal transduction pathways (2-4). The BUB1-BUB3 complex plays a role in the inhibition of APC/C when spindle-assembly checkpoint is activated and inhibits the ubiquitin ligase activity of APC/C by phosphorylating its activator CDC20. This complex can also phosphorylate MAD1L1. Kinase activity is essential for inhibition of APC/CCDC20 and for chromosome alignment but does not play a major role in the spindle-assembly checkpoint activity. Mediates cell death in response to chromosome missegregation and acts to suppress spontaneous tumorigenesis.

Siah2 is a E3 Ubiquitin ligase protein that mediates ubiquitination and subsequent proteasomal degradation of target proteins. E3 ubiquitin ligases accept ubiquitin from an E2 ubiquitin-conjugating enzyme in the form of a thioester and then directly transfer the ubiquitin to targets. Siah2 triggers the ubiquitin-mediated degradation of many proteins, including transcription regulators (e.g. POU2AF1, PML, NCOR1), cell surface receptors (e.g. DCC), antiapoptotic proteins (e.g. BAG1), and proteins involved in synaptic vesicle function in neurons (e.g. SYP). Siah2 has some overlapping function with SIAH1 but is able to trigger the ubiquitin-mediated degradation of TRAF2, whereas SIAH1 can not.
