Description:
Serine protease which exhibits a preference for Arg over Lys in the substrate P1 position and for Ser or Pro in the P2 position. Shows activity against amyloid precursor protein, myelin basic protein, gelatin, casein and extracellular matrix proteins such as fibronectin, laminin, vitronectin and collagen. Degrades alpha-synuclein and prevents its polymerization, indicating that it may be involved in the pathogenesis of Parkinson disease and other synucleinopathies. May be involved in regulation of axon outgrowth following spinal cord injury. Tumor cells treated with a neutralizing KLK6 antibody migrate less than control cells, suggesting a role in invasion and metastasis.

Description:
Members of the myogenic determination family are basic helix-loop-helix (bHLH) proteins that can be separated into two classes, both of which work together to activate DNA transcription. Class A proteins include the ubiquitously expressed E-box binding factors, namely E2A, ITF-2 and HEB, while class B proteins, such as MyoD, myogenin and Neuro D (BETA2), are transiently expressed and exhibit a much more limited tissue distribution. Working in opposition to these positively acting factors are a specialized group of basic helix-loop-helix (bHLH) transcription factors that function as dominant negative regulators and are involved in cell lineage determination and differentiation. Neuro D2 (neurogenic differentiation 2), also known as NDRF, NEUROD2 or bHLHa1, is a 382 amino acid nuclear protein that contains one bHLH domain and functions to induce neurogenic differentiation, playing an important role in the maintenance and determination of cell fate.

Description:
Neugrin, also known as NGRN, mesenchymal stem cell protein DSC92, neurite outgrowth-associated protein or spinal cord-derived protein FI58G, is a 291 amino acid protein that plays a role in neuronal differentiation and belongs to the neugrin family. As both a secreted and nuclear protein, neugrin exists as two alternatively spliced isoforms and is highly expressed in skeletal muscle, brain and heart. Neugrin is upregulated in neuroblastostoma cells by retinoic acid treatment and is encoded by a gene that maps to human chromosome 15q26.1. Chromosome 15 houses over 700 genes and comprises nearly 3% of the human genome. Angelman syndrome, Prader-Willi syndrome, Tay-Sachs disease and Marfan syndrome are all associated with defects in chromosome 15-localized genes.

Description:
E3 ubiquitin-protein ligase that promotes the degradation of insoluble ubiquitinated proteins, including insoluble PAX6, poly-Gln repeat expanded HTT and poly-Ala repeat expanded ARX. Mediates PAX6 ubiquitination leading to proteasomal degradation, thereby modulating cortical neurogenesis. May also inhibit PAX6 transcriptional activity, possibly in part by preventing the binding of PAX6 to its consensus sequences. May contribute to the regulation of the intracellular level of HN (humanin) or HN-containing proteins through the proteasomal degradation pathway. Mediates MED15 ubiquitination leading to proteasomal degradation. May contribute to the innate restriction of retroviruses. Upon overexpression, reduces HIV-1 and murine leukemia virus infectivity, by suppressing viral gene expression. Antiviral activity depends on a functional E3 ubiquitin-protein ligase domain. May regulate TRIM5 turnover via the proteasome pathway, thus counteracting the TRIM5-mediated cross-species restriction of retroviral infection at early stages of the retroviral life cycle.

Description:
Fyb (Fyn binding protein) and the anchoring proteins SKAP55 and SKAP55-R (SKAP55-related protein) associate with the tyrosine kinase p59fyn. SKAP55 and SKAP55-R bind to Fyb through their SH3 domains and function as substrates for p59Fyn in resting T cells. SKAP55 contains an N-terminal pleckstrin homology domain and a C-terminal SH3 domain binding motif of adjacent arginine and lysine residues followed by tandem tyrosines (i.e. RKxxYxxY). SKAP55-R, similar in overall structure to SKAP55, contains a coiled-coil N-terminal domain. SKAP55 associates with SLAP-130, another component of the Fyn complex, which plays a role in the regulation of signaling events initiated by lymphocyte antigen receptors leading up to T cell activation. The human SKAP55 gene maps to chromosome 17q21.32 and encodes a 359 amino acid protein.

Description:
S100A8 is a calcium- and zinc-binding protein which plays a prominent role in the regulation of inflammatory processes and immune response. It can induce neutrophil chemotaxis and adhesion. Predominantly found as calprotectin (S100A8/A9) which has a wide plethora of intra- and extracellular functions. The intracellular functions include: facilitating leukocyte arachidonic acid trafficking and metabolism, modulation of the tubulin-dependent cytoskeleton during migration of phagocytes and activation of the neutrophilic NADPH-oxidase. Activates NADPH-oxidase by facilitating the enzyme complex assembly at the cell membrane, transferring arachidonic acid, an essential cofactor, to the enzyme complex and S100A8 contributes to the enzyme assembly by directly binding to NCF2/P67PHOX. The extracellular functions involve proinfammatory, antimicrobial, oxidant-scavenging and apoptosis-inducing activities. Its proinflammatory activity includes recruitment of leukocytes, promotion of cytokine and chemokine production, and regulation of leukocyte adhesion and migration. Acts as an alarmin or a danger associated molecular pattern (DAMP) molecule and stimulates innate immune cells via binding to pattern recognition receptors such as Toll-like receptor 4 (TLR4) and receptor for advanced glycation endproducts (AGER). Binding to TLR4 and AGER activates the MAP-kinase and NF-kappa-B signaling pathways resulting in the amplification of the proinflammatory cascade. Has antimicrobial activity towards bacteria and fungi and exerts its antimicrobial activity probably via chelation of Zn(2+) which is essential for microbial growth. Can induce cell death via autophagy and apoptosis and this occurs through the cross-talk of mitochondria and lysosomes via reactive oxygen species (ROS) and the process involves BNIP3. Can regulate neutrophil number and apoptosis by an anti-apoptotic effect; regulates cell survival via ITGAM/ITGB and TLR4 and a signaling mechanism involving MEK-ERK.

Description:
Unrip is part of the SMN complex that plays a role in snRNP assembly in the cytoplasm and pre mRNA splicing in the nucleus. Unrip interacts directly with Gemin 6 and Gemin 7 in the SMN complex. It is associated with the complex in the cytoplasm but not in the nucleus and thus is thought to play a role in it's subcellular localisation. It is a ubiquitously expressed protein that acts as an inhibitor of TGF-beta signaling and an important regulator of cell proliferation. Stable expression of STRAP results in activation of the mitogen-activated protein kinase/extracellular signal-regulated kinase pathway and in down-regulation of the cyclin-dependent kinase inhibitor p21 (Cip1), which results in retinoblastoma protein hyperphosphorylation. Upregulation of STRAP might play a role in tumor development as an early event for colorectal cancers. It is the first component of the U snRNP assembly machinery that associates with SMN complex in a compartment-specific way and plays a crucial role in the intracellular distribution of SMN1.

Description:
Sulfatases hydrolyze sulfate esters from sulfated steroids, carbohydrates, proteoglycans and glycolipids. They are involved in hormone biosynthesis, modulation of cell signaling and degradation of macromolecules. Arylsulfatase H, also known as ARSH, is a 562 amino acid protein that belongs to the sulfatase family of bone and cartilage matrix proteins. Localized to the plasma membrane, Arylsulfatase H uses calcium as a cofactor to hydrolyze sulfate esters. The gene encoding Arylsulfatase D maps to human chromosome X, which contains nearly 153 million base pairs and houses over 1,000 genes. In conjunction with chromosome Y, chromosome X is responsible for sex determination. There are a number of conditions related to an abnormal number and combination of sex chromosomes, some of which include Turner's syndrome, color blindness, hemophilia and Duchenne muscular dystrophy.

Description:
Chromosome 9 consists of about 145 million bases and 4% of the human genome and encodes nearly 900 genes. Considered to play a role in gender determination, deletion of the distal portion of 9p can lead to development of male to female sex reversal, the phenotype of a female with a male X,Y genotype. Hereditary hemorrhagic telangiectasia, which is characterised by harmful vascular defects, is associated with the chromosome 9 gene encoding endoglin protein, ENG. Familial dysautonomia is also associated with chromosome 9 though through the gene IKBKAP. Notably, chromosome 9 encompasses the largest interferon family gene cluster. Chromosome 9 is partnered with chromosome 22 in the translocation leading to the aberrant production of BCR-ABL fusion protein often found in leukemias. The C9orf93 gene product has been provisionally designated C9orf93 pending further characterisation. There are two isoforms of C9orf93 that are produced as a result of alternative splicing events.

Description:
Vasodilator-stimulated phosphoprotein (VASP) is a member of the Ena-VASP protein family. Ena-VASP family members contain an EHV1 N-terminal domain that binds proteins containing E/DFPPPPXD/E motifs and targets Ena-VASP proteins to focal adhesions. In the mid-region of the protein, family members have a proline-rich domain that binds SH3 and WW domain-containing proteins. Their C-terminal EVH2 domain mediates tetramerization and binds both G and F actin. VASP is associated with filamentous actin formation and likely plays a widespread role in cell adhesion and motility. VASP may also be involved in the intracellular signaling pathways that regulate integrin-extracellular matrix interactions. VASP is regulated by the cyclic nucleotide-dependent kinases PKA and PKG.
