In mice, the Exo1 gene maps to distal chromosome 1, consistent with the recent mapping of the orthologous human HEX1/EXO1 gene to chromosome 1q43. Exo1 is expressed prominently in testis, an area of active homologous recombination, and spleen, a prominent lymphoid tissue. In both mammalian and yeast systems, Exo1 is a 5'-3' double stranded DNA exonuclease that has previously been implicated in DNA mismatch repair (MMR). The MMR system ensures genome integrity by removing mispaired and unpaired bases that originate during replication. In humans, Exo1 interacts with MSH2 and MLH1 and has been proposed to be a redundant exonuclease in MMR. Protocadherins are a large family of cadherin-like cell adhesion proteins that are involved in the establishment and maintenance of neuronal connections in the brain. There are three protocadherin gene clusters, designated alpha, beta and gamma, all of which contain multiple tandemly arranged genes. PCDHB6 (Protocadherin beta-6) is a 794 amino acid single pass transmembrane protein that is one of 16 proteins in the protocadherin beta cluster. Unlike the alpha and gamma gene clusters whose genes are spliced to downstream constant region exons during transcription, members of the beta cluster (such as PCDHB6) do not use constant-region exons to produce mRNAs. As a result, each protocadherin beta gene encodes the transmembrane, extracellular and short cytoplasmic domains of the protein. PCDHB6 is likely a calcium-dependent cell adhesion protein that is involved in the maintenance of neural connections in the brain. Unlike most protocadherin-beta proteins, PCDHB6 has not one but two PXXP motifs within its cytoplasmic domain, suggesting a role in signal transduction cascade events.

Dpy-19 (dumpy-19), is a 683 amino acid C. elegans protein that is required to orient the neuroblasts QL and QR correctly on the anterior/posterior axis. Dpy-19 is expressed highly in dorsal hyp7 cells, ventral P cells and lateral V cells, and dorsal and ventral body muscle cells. DPY19L1 (Dpy-19-like protein 1), also known as KIAA0877, is a 675 amino acid multi-pass membrane protein that belongs to the Dpy-19 family. DPY19L1 is expressed as two isoforms produced by alternative splicing and is encoded by a gene mapping to human chromosome 7, which encodes over 1,000 genes and makes up about 5% of the human genome. Diseases associated with chromosome 7 include Osteogenesis imperfecta, Pendred syndrome, Lissencephaly, Citrullinemia and Shwachman-Diamond syndrome. The deletion of a portion of the q arm of chromosome 7 is associated with Williams-Beuren syndrome, a condition characterized by mild mental retardation, an unusual comfort and friendliness with strangers and an elfin appearance. Deletions of portions of the q arm of chromosome 7 are also seen in a number of myeloid disorders including cases of acute myelogenous leukemia and myelodysplasia.

Gem belongs to the Rad/Gem/Kir (RGK) subfamily of Ras-related GTPases, which lack typical C-terminal amino acid motifs for isoprenylation. Rad and Gem bind calmodulin in a Ca2+-dependent manner via this C-terminal extension, involving residues 278–297 in human Rad. High intracellular Gem levels, which interact with intact microtubules and microfilaments, promote profound changes in cell morphology. Ectopic Gem expression is sufficient to stimulate cell flattening and neurite extension in N1E-115 and SH-SY5Y neuroblastoma cells, suggesting a role for Gem in cytoskeletal rearrangement and/or morphological differentiation of neurons. Gem was also observed in developing trigeminal nerve ganglia in 12.5 day mouse embryos, demonstrating that Gem expression is a property of normal ganglionic development. The interaction of Gem with beta-subunits regulates Ca2+ channel expression at the cell surface. The human Gem gene maps to chromosome 8q22.1.

The protein encoded by this gene is a lysosomal cysteine proteinase that plays a major role in intracellular protein catabolism. Its substrates include collagen and elastin, as well as alpha-1 protease inhibitor, a major controlling element of neutrophil elastase activity. The encoded protein has been implicated in several pathologic processes, including myofibril necrosis in myopathies and in myocardial ischemia, and in the renal tubular response to proteinuria. This protein, which is a member of the peptidase C1 family, is a dimer composed of disulfide-linked heavy and light chains, both produced from a single protein precursor. At least two transcript variants encoding the same protein have been found for this gene.
