Description:   FIG4 is a 907 amino acid protein that contains one SAC domain through which it is thought to function as a phosphoinositide phosphatase that may play an important role in signal transduction and vesicle trafficking. Defects in the gene encoding FIG4 are the cause of Charcot-Marie-Tooth disease type 4J (CMT4J) and amyotrophic lateral sclerosis type 11 (ALS11), both of which negatively affect the nervous system. CMT4J is a recessive demyelinating disorder of the peripheral nervous system and is characterised by reduced motor nerve conduction velocities and axonal degeneration. Unlike CMT4J, ALS11 is characterised by the degeneration of upper motor neurons in the brain and lower neurons in the spinal cord, causing paralysis and, ultimately, death.

Description:   Enzymes known as phosphatidylserine decarboxylases (PSDs) catalyze the formation of phosphatidylethanolamine from phosphatidylserine via phosphatidylserine decarboxylation. Type I PSDs contain LGST motifs and are found in bacteria and eukaryotic mitochondria, whereas type II PSDs contain GGST motifs and are found in eukaryotic endomembrane systems. PISD (phosphatidylserine decarboxylase), also known as phosphatidylserine decarboxylase proenzyme, PSDC, PSD, PSSC, DJ858B16, dJ858B16.2 or DKFZp566G2246, is a 408 amino acid a type I phosphatidylserine decarboxylase that localizes to the inner mitochondrial membrane. PISD contains a conserved LGST motif which is cleaved to produce two isoforms known as PISD α and PISD β. PISD is capable of forming a heterodimer and is highly expressed in liver and testis. The gene encoding PISD maps to human chromosome 22q12.2.

Description:   Zinc is an essential cofactor that is involved in cell growth and development, as well as in protein, nucleic acid and lipid metabolism. The transport of zinc across the cell membrane is crucial for correct enzyme and overall cell function. SLC39A11 (solute carrier family 39 (metal ion transporter), member 11), also known as ZIP11 (Zrt- and Irt-like protein 11), is a 342 amino acid multi-pass membrane protein belonging to the ZIP transporter family. Expressed as multiple alternatively spliced isoforms, SLC39A11 acts as a zinc-influx transporter and is encoded by a gene located on human chromosome 17, which comprises over 2.5% of the human genome and encodes over 1,200 genes, some of which are involved in tumor suppression and in the pathogenesis of Li-Fraumeni syndrome, early onset breast cancer and a predisposition to cancers of the ovary, colon, prostate gland and fallopian tubes.

Description:   The ABL1 protooncogene encodes a cytoplasmic and nuclear protein tyrosine kinase that has been implicated in processes of cell differentiation, cell division, cell adhesion, and stress response. Activity of c-Abl protein is negatively regulated by its SH3 domain, and deletion of the SH3 domain turns ABL1 into an oncogene. The t(9;22) translocation results in the head-to-tail fusion of the BCR (MIM:151410) and ABL1 genes present in many cases of chronic myelogeneous leukemia. The DNA-binding activity of the ubiquitously expressed ABL1 tyrosine kinase is regulated by CDC2-mediated phosphorylation, suggesting a cell cycle function for ABL1. The ABL1 gene is expressed as either a 6- or 7-kb mRNA transcript, with alternatively spliced first exons spliced to the common exons 2-11. [provided by RefSeq].

Description:   Protein kinases are enzymes that transfer a phosphate group from a phosphate donor onto an acceptor amino acid in a substrate protein. By this basic mechanism, protein kinases mediate most of the signal transduction in eukaryotic cells, regulating cellular metabolism, transcription, cell cycle progression, cytoskeletal rearrangement and cell movement, apoptosis, and differentiation. The protein kinase family is one of the largest families of proteins in eukaryotes, classified in 8 major groups based on sequence comparison of their tyrosine (PTK) or serine/threonine (STK) kinase catalytic domains. Epidermal Growth factor receptor (EGFR) is the prototype member of the type 1 receptor tyrosine kinases. EGFR overexpression in tumors indicates poor prognosis and is observed in tumors of the head and neck, brain, bladder, stomach, breast, lung, endometrium, cervix, vulva, ovary, esophagus, stomach and in squamous cell carcinoma.

Description:   Polyglutamine(Q) tract binding protein-1 (PQBP-1) is a transcription repressor that associates with polyglutamine tract-containing transcription regulators and causative genes for neurodegenerative disorders. Hepta- and di-amino acid repeat sequences rich in polar residues are essential for PQBP-1 to interact with polyglutamine tract-containing proteins (i.e. huntingtin, androgen receptor and Brain-2). PQBP-1 contains a WWP/WW domain that binds proline-rich motifs and a C2 domain that can influence Ca2+-dependent phospholipid signaling. PQBP-1 localizes to the nucleus and is present in neurons throughout the brain, with abundant levels in hippocampus, cerebellar cortex and olfactory bulb. The human PQBP-1 gene maps to chromosome Xp11.23.

Description:   Neurexophilin family (Neurexophilin-1-4) of neuropeptide-like glycoproteins that are proteolytically processed after synthesis. Neurexophilin-1-3 are secreted proteins that are thought to function as signaling molecules which specifically bind to target proteins, such as neurexin I?(a protein that promotes adhesion between dendrites and axons), and are essential for proper neurotransmitter release. While Neurexophilin-1 is located primarily in spleen tissue, Neurexophilin-2 is expressed primarily in kidney and both Neurexophilin-2 and Neurexophilin-3 are highly expressed in brain. Defects in the gene encoding Neurexophilin-1 may be associated with schizophrenia, a mental disorder characterized by an abnormal perception of reality.

Description:   Chromosome 9 consists of about 145 million bases and 4% of the human genome and encodes nearly 900 genes. Considered to play a role in gender determination, deletion of the distal portion of 9p can lead to development of male to female sex reversal, the phenotype of a female with a male X,Y genotype. Hereditary hemorrhagic telangiectasia, which is characterised by harmful vascular defects, is associated with the chromosome 9 gene encoding endoglin protein, ENG. Familial dysautonomia is also associated with chromosome 9 though through the gene IKBKAP. Notably, chromosome 9 encompasses the largest interferon family gene cluster. Chromosome 9 is partnered with chromosome 22 in the translocation leading to the aberrant production of BCR-ABL fusion protein often found in leukemias. The C9orf71 gene product has been provisionally designated C9orf71 pending further characterisation.

Description:   C8orf76 (chromosome 8 open reading frame 76) is a 380 amino acid protein encoded by a gene that maps to human chromosome 8q24.13. Consisting of nearly 146 million bases, chromosome 8 encodes about 800 genes. Translocation of portions of chromosome 8 with amplifications of the c-Myc gene are found in some leukemias and lymphomas, and are typically associated with a poor prognosis. Portions of chromosome 8 have been linked to schizophrenia and bipolar disorder. Trisomy 8, also known as Warkany syndrome 2, most often results in early miscarriage but is occasionally seen in a mosaic form in surviving patients who suffer to a varying degree from a number of symptoms including retarded mental and motor development, and certain facial and developmental defects. WRN is a DNA helicase encoded by chromosome 8 and shown defective in those with the early aging disorder Werner syndrome. Chromosome 8 is also associated with Pfeiffer syndrome, congenital hypothyroidism and Waardenburg syndrome.

Description:   A novel gene, Genes associated with Retinoid IFN induced Mortality (GRIM) GRIM19 gene was identified. Antisense expression of GRIM19 confers a strong resistance against IFN/RA induced death by reducing the intracellular levels of GRIM19 protein. Overexpression of GRIM19 enhances cell death in response to IFN/RA. GRIM19 is primarily a nuclear protein whose expression is induced by the IFN/RA combination. These data indicate that GRIM19 is a novel cell death regulatory molecule.
