This gene encodes a bifunctional enzyme containing phosphoribosylaminoimidazole carboxylase activity in its N-terminal region and phosphoribosylaminoimidazole succinocarboxamide synthetase in its C-terminal region. It catalyzes steps 6 and 7 of purine biosynthesis. The gene is closely linked and divergently transcribed with a locus that encodes an enzyme in the same pathway, and transcription of the two genes is coordinately regulated. LAP3 (leucine aminopeptidase 3), also known as LAPEP or PEPS, is a 519 amino acid protein that localizes to the cytoplasm and belongs to the peptidase M17 family. Existing as a homohexamer, LAP3 uses zinc as a cofactor to catalyze the release of an N-terminal proline from a target peptide and is, therefore, involved in the processing and turnover of intracellular proteins. Multiple isoforms of LAP3 exist due to alternative splicing events. The gene encoding LAP3 maps to human chromosome 4, which houses nearly 6% of the human genome and has the largest gene deserts (regions of the genome with no protein encoding genes) of all of the human chromosomes. Defects in some of the genes located on chromosome 4 are associated with Huntington's disease, Ellis-van Creveld syndrome, methylmalonic acidemia and polycystic kidney disease.

CRTAC1 is a 661 amino acid secreted protein. CRTAC1 is O-glycosylated and contains one EGF-like domain and four FG-GAP repeats. Up-regulated in mesenchymal stem cells undergoing chondrogenic differentiation and also by growth factor BMP-4, CRTAC1 is found in the interterritorial matrix of articular deep zone cartilage. CRTAC1 is expressed as three isoforms produced by alternative splicing events. All isoforms are expressed in bone, cartilage and lung, while isoforms 1 and 2 are expressed in the brain. The gene that encodes CRTAC1 maps to human chromosome 10, which contains over 800 genes and 135 million nucleotides, making up nearly 4.5% of the human genome.

Glutamate receptors mediate most excitatory neurotransmission in the brain and play an important role in neural plasticity, neural development and neurodegeneration. Ionotropic glutamate receptors are categorized into NMDA receptors and kainate/AMPA receptors, both of which contain glutamate-gated, cation-specific ion channels. Kainate/AMPA receptors are co-localized with NMDA receptors in many synapses and consist of seven structurally related subunits designated GluR-1 to -7. The kainate/AMPA receptors are primarily responsible for the fast excitatory neuro-transmission by glutamate, whereas the NMDA receptors are functionally characterized by a slow kinetic and a high permeability for Ca2+ ions. The NMDA receptors consist of five subunits: epsilion 1, 2, 3, 4 and one zeta subunit. The zeta subunit is expressed throughout the brainstem, whereas the four epsilon subunits display limited distribution.

Description:
Crystallins are separated into two classes: taxon-specific, or enzyme, and ubiquitous. The latter class constitutes the major proteins of vertebrate eye lens and maintains the transparency and refractive index of the lens. Since lens central fiber cells lose their nuclei during development, these crystallins are made and then retained throughout life, making them extremely stable proteins. Mammalian lens crystallins are divided into alpha, beta, and gamma families; beta and gamma crystallins are also considered as a superfamily. Alpha and beta families are further divided into acidic and basic groups. Seven protein regions exist in crystallins: four homologous motifs, a connecting peptide, and N- and C-terminal extensions. Alpha crystallins are composed of two gene products: alpha-A and alpha-B, for acidic and basic, respectively. Alpha crystallins can be induced by heat shock and are members of the small heat shock protein (sHSP also known as the HSP20) family. They act as molecular chaperones although they do not renature proteins and release them in the fashion of a true chaperone; instead they hold them in large soluble aggregates. Post-translational modifications decrease the ability to chaperone. These heterogeneous aggregates consist of 30-40 subunits; the alpha-A and alpha-B subunits have a 3:1 ratio, respectively. Two additional functions of alpha crystallins are an autokinase activity and participation in the intracellular architecture. Alpha-A and alpha-B gene products are differentially expressed; alpha-A is preferentially restricted to the lens and alpha-B is expressed widely in many tissues and organs. Impairment of DDAH causes ADMA accumulation and a reduction in cGMP generation (3). DDAH II, the predominant DDAH isoform in endothelial cells, facilitates the induction of nitric oxide synthesis by all-trans-Retinoic acid (atRA) (4). MAP (mitogen-activated protein) kinases play a significant role in many biological processes, including cell adhesion and spreading, cell differentiation and apoptosis. p38 alpha, p38 beta and p38 gamma, also known as MAPK14, MAPK11 and MAPK12, respectively, each contain one protein kinase domain and belong to the MAP kinase family. Expressed in different areas throughout the body with common expression patterns in heart, p38 proteins use magnesium as a cofactor to catalyze the ATP-dependent phosphorylation of target proteins. Via their catalytic activity, p38 alpha, p38 beta and p38 gamma are involved in a variety of events throughout the cell, including signal transduction pathways, cytokine production and cell proliferation and differentiation. The p38 proteins are subject to phosphoryation on Thr and Tyr residues, an event which is thought to activate the phosphorylated protein.
