This gene encodes coagulation factor VIII, which participates in the intrinsic pathway of blood coagulation; factor VIII is a cofactor for factor IXa which, in the presence of Ca+2 and phospholipids, converts factor X to the activated form Xa. This gene produces two alternatively spliced transcripts. Transcript variant 1 encodes a large glycoprotein, isoform a, which circulates in plasma and associates with von Willebrand factor in a noncovalent complex. This protein undergoes multiple cleavage events. Transcript variant 2 encodes a putative small protein, isoform b, which consists primarily of the phospholipid binding domain of factor VIIIc. GCF (GC-rich sequence DNA-binding factor), also known as C2orf3 (chromosome 2 open reading frame 3), transcription factor 9 (TCF-9) or DNABF, is a 781 amino acid nuclear protein that belongs to the GCF family. Widely expressed, GCF binds the GC-rich sequences of _-Actin, EGFR and calcium-dependent protease (CANP) promoters. GCF contains multiple phosphoserine and phosphothreonine residues, and two GCF isoforms are produced due to alternative splicing events. GCF is considered a candidate for susceptibility to dyslexia (DYX3) as both genes reside in close proximity on human chromosome 2. Chromosome 2 is the second largest human chromosome and consists of 237 million bases, encodes over 1,400 genes and makes up approximately 8% of the human genome.

The endophilins comprise a family of three SH3 domain-containing proteins designated Endophilin I, II and III, or alternatively known as SH3P4, SH3P8 and SH3P13, respectively. These proteins associate with Amphiphysin, Synaptojanin and Dynamin and are implicated in presynaptic vesicle trafficking at nerve terminals. The expression patterns of the endophilins are consistent with their cellular functions at the neuronal synapse, as Endophilin I is ex- pressed only in the brain. Both Endophilin II and Endophilin III are detected in a variety of tissues. Endophilin I is also implicated in modulating G protein-coupled receptor signaling by functioning as an adapter protein and directing 1 adrenergic receptors to the endocytic machinery.

Auxiliary protein of DNA polymerase delta and is involved in the control of eukaryotic DNA replication by increasing the polymerase's processibility during elongation of the leading strand.
